    Two-Dimensional Molecular Patterning by Surface-Enhanced Zn-Porphyrin Coordination

    In this contribution, we show how zinc-5,10,15,20-meso-tetradodecylporphyrins (Zn-TDPs) self-assemble into stable organized arrays on the surface of graphite, thus positioning their metal center at regular distances from each other, creating a molecular pattern, while retaining the possibility to coordinate additional ligands. We also demonstrate that Zn-TDPs coordinated to 3-nitropyridine display a higher tendency to be adsorbed at the surface of highly oriented pyrolytic graphite (HOPG) than noncoordinated ones. In order to investigate the two-dimensional (2D) self-assembly of coordinated Zn-TDPs, solutions with different relative concentrations of 3-nitropyridine and Zn-TDP were prepared and deposited on the surface of HOPG. STM measurements at the liquid-solid interface reveal that the ratio of coordinated Zn-TDPs over noncoordinated Zn-TDPs is higher at the n-tetradecane/HOPG interface than in n-tetradecane solution. This enhanced binding of the axial ligand at the liquid/solid interface is likely related to the fact that physisorbed Zn-TDPs are better binding sites for nitropyridines.

    Combination Service for Time-variable Gravity Fields: operational GRACE-FO combination and validation of Chinese GRACE time-series

    The Combination Service for Time-variable Gravity Fields (COST-G) of the International Association of Geodesy (IAG) provides combined monthly gravity fields of its associated and partner Analysis Centers (ACs). In November 2020, the combination of monthly GRACE-FO gravity fields started its operational mode, providing consolidated L2 (spherical harmonics) and L3 (gridded and post-processed) products with a latency of currently 3 months. We present an overview and quality assessment of the available products. COST-G aims at the extension of its service to include further GRACE and GRACE-FO analysis centers. In January 2020 a collaboration with representatives of five Chinese ACs was initiated, who provided GRACE time-series according to the COST-G requirements. We present the results of a test combination with the Chinese AC models, including comparison and quality assessment of all contributing time-series and validation of the combined gravity fields

    Since its start of operations in July 2019, IAGâ?Ts Combination Service for Time-variable Gravity fields (COST-G) is providing a complete time-series of combined monthly GRACE gravity fields and a regularly updated time-series of monthly gravity fields derived from kinematic Swarm orbits. Starting from October 2020, the COST-G product line is complemented by a time-series of operationally combined and monthly updated GRACE-FO gravity fields. All these combinations are performed by variance component estimation on the solution level. We report on new developments, i.e., a planned extension of COST-G to include Chinese analysis centers of GRACE and GRACE-FO data, a re-consideration of the combination strategy to better focus on the range of spherical harmonic coefficients most relevant for the users, and the potential application of COST-G products for orbit determination of altimeter satellites

The phytochemistry of grapevine is rich in a wide range of compounds. Many of them are renowned for their numerous medicinal uses. The production of grapevine metabolites is highly conditioned by many factors like environment or pathogen attack. Some grapevine phytoalexins have gained a great deal of attention due to their antimicrobial activities, being also involved in the induction of resistance in grapevine against those pathogens. Meanwhile grapevine biotechnology is still evolving, thanks to the technological advance of modern science, and biotechnologists are making huge efforts to produce grapevine cultivars of desired characteristics. In this paper, important metabolites from grapevine and grape derived products like wine will be reviewed with their health promoting effects and their role against certain stress factors in grapevine physiology

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Residual ocean tide signals from satellite altimetry, GRACE gravity fields, and hydrodynamic modelling

    P>Improvements of a global state-of-the art ocean tide model are identified and quantified by applying three independent approaches, namely (i) empirical ocean tide analysis of multimission altimeter data, (ii) evaluation of GRACE data and gravity field models and (iii) high resolution hydrodynamic modelling. Although these approaches have different capabilities to sense ocean tides they obtain results which are basically consistent one with each other. The analysis of altimeter data clearly identifies significant residual amplitudes over shallow water for all major diurnal and semidiurnal constituents and the non-linear tide M-4. GRACE data and the time-series of monthly gravity field models exhibit-on a larger scale-residual ocean tide signals over much the same areas. The analysis of dynamic residuals of hydrodynamic modelling with data assimilation proves the validity of linear dynamics in the deep ocean and shows correlation of dynamic residuals with energy dissipation in these areas

    Overweight and Obesity Based on Four Reference Systems in 18,382 Paediatric Patients with Type 1 Diabetes from Germany and Austria

    Aim. To evaluate the prevalence of overweight and obesity in paediatric type 1 diabetes (T1D) subjects, based on four commonly used reference populations. Methods. Using WHO, IOTF, AGA (German pediatric obesity), and KiGGS (German Health Interview and Examination Survey for Children and Adolescents) reference populations, prevalence of overweight (≥90th percentile) and obesity (≥97th percentile) and time trend between 2000 (n = 9,461) and 2013 (n = 18,382) were determined in 2–18-year-old T1D patients documented in the German/Austrian DPV database. Results. In 2000, the overweight prevalence was the highest according to IOTF (22.3%), followed by WHO (20.8%), AGA (15.5%), and KiGGS (9.4%). The respective rates in 2013 were IOTF (24.8%), WHO (22.9%), AGA (18.2%), and KiGGS (11.7%). Obesity prevalence in 2000 was the highest according to WHO (7.9%), followed by AGA (4.5%), IOTF (3.1%), and KiGGS (1.8%). In 2013, the respective rates were WHO (9.6%), AGA (6.2%), IOTF (4.5%), and KiGGS (2.6%). Overall, the prevalence of overweight and obesity increased from 2000 to 2006 (p < 0.001) but showed stabilization thereafter in girls and overweight in boys. Conclusion. Overweight and obesity prevalence in T1D subjects differs significantly if it is assessed by four separate reference populations. More detailed assessment of each child is required to determine obesity-related risks
