    Development of GABAergic and glycinergic transmission in the neonatal rat dorsal horn

    Cutaneous spinal sensory transmission appears to lack inhibitory control in the newborn spinal cord, but the properties of GABAergic and glycinergic synapses in the neonatal dorsal horn have not been characterized. Whole-cell patch-clamp recordings from rat superficial dorsal horn neurons in spinal cord slices at postnatal day 0 (P0) to P2, P6 - P7, and P13 - P14 revealed an age-dependent increase in the frequency of spontaneous IPSCs, which were abolished by the GABA(A) receptor (GABA(A)R) antagonist bicuculline between P0 and P7 but not at P14. GABA(A)R-mediated miniature IPSCs (mIPSCs), but not glycinergic mIPSCs, were present at birth, and GABA mIPSCs remained more frequent than glycine mIPSCs at all ages. Sciatic nerve stimulation resulted in IPSCs with both GABAergic and glycinergic components, although a larger contribution arose from GABAA receptors at all ages. In gramicidin perforated patch-clamp recordings, exogenous GABA applications produced depolarization in 40% of neurons at P0 - P2, but the reversal potential of GABA-evoked currents (E-GABA) was consistently more negative than action potential threshold at this age. By P6 - P7, GABA evoked only membrane hyperpolarization. The GABA(B)R agonist baclofen elicited an outward current in all neurons with peak amplitudes observed by P6 - P7 and abolished sciatic nerve-evoked monosynaptic glutamatergic EPSCs in all groups. The results show considerable postnatal development of inhibitory processing in the dorsal horn with GABAergic mechanisms initially dominant over glycinergic events. GABA(A)R-mediated depolarizations during the first postnatal week are likely to be important for the maturation of spinal networks but do not provide a major excitatory drive to the newborn dorsal horn

    Magnetohydrodynamic normal mode analysis of plasma with equilibrium pressure anisotropy

    In this work, we generalise linear magnetohydrodynamic (MHD) stability theory to include equilibrium pressure anisotropy in the fluid part of the analysis. A novel 'single-adiabatic' (SA) fluid closure is presented which is complementary to the usual 'double-adiabatic' (CGL) model and has the advantage of naturally reproducing exactly the MHD spectrum in the isotropic limit. As with MHD and CGL, the SA model neglects the anisotropic perturbed pressure and thus loses non-local fast-particle stabilisation present in the kinetic approach. Another interesting aspect of this new approach is that the stabilising terms appear naturally as separate viscous corrections leaving the isotropic SA closure unchanged. After verifying the self-consistency of the SA model, we re-derive the projected linear MHD set of equations required for stability analysis of tokamaks in the MISHKA code. The cylindrical wave equation is derived analytically as done previously in the spectral theory of MHD and clear predictions are made for the modification to fast-magnetosonic and slow ion sound speeds due to equilibrium anisotropy.Comment: 19 pages.     Pathways to interleukin-6 in healthy males and serious leisure male athletes: physical activity, body composition and age.

    Physical activity (PA) is beneficial to overall health, in part due to physiological changes that lower risk factors for cardiovascular disease, including reduced inflammation. However, the mechanism by which PA reduces inflammation is unclear. One possible pathway is that PA improves body composition which in turn reduces inflammation. To test this hypothesis, we used structural equation modeling (SEM) to assess PA-body composition -inflammation pathways, as well as influences of age. In a sample of 72 healthy males with a range of PA profiles (age 18-65, mean ± sd = ), we measured PA as metabolic equivalent tasks (as per the International PA Questionnaire), body composition as percent body fat, lean mass, and fat mass, and inflammation as plasma interleukin-6 (IL-6). We treated body composition in the SEM analysis as a latent variable indicated by the three measures. We performed statistical corrections for missing values and one outlier. The model demonstrated significant effects of PA on IL-6 both directly and through body composition. Percent body fat, fat mass, and lean mass were significant indicators of the body composition latent variable. Additionally, age showed an indirect effect on IL-6 through body composition, but no direct effect. The findings suggest that PA does improve inflammatory profile through improving body composition, but that other pathways also exist

    End of project reportThere are almost 150,000 farms in Ireland and these contribute substantial quantities of N and P to inland and coastal waters. Some of these nutrients are carried from wet soils by overland flow and by leaching from dry soils. Farm practice can reduce the loss from farms by judicious management of nutrients. Improvements are required to diminish export of nutrients without impairing operations on the farm. Literature regarding nutrient loss from agriculture was reviewed in this project and maps were prepared to predict best slurry spreading times around Ireland. Two further maps were prepared to show slurry storage requirement on farms
