    Assessing infection patterns in Chinese mystery snails from Wisconsin, USA using field and laboratory approaches

    The success of invasive species establishment in new habitats depends, in part, on interactions with other members of the invaded community. Cipangopaludina chinensis, the Chinese mystery snail (CMS), is an exotic, invasive snail in North America. Since its introduction in the 1890s, CMS have spread to a number of watersheds across the United States and parts of Canada. This invasion has generated questions about the snail’s capacity to serve as a host for parasites within local habitats, including parasites with conservation implications. To begin addressing these questions, we necropsied 147 wild-caught snails from 22 lakes across Wisconsin, finding only two CMS individuals harboring trematode (flatworm) parasites. We also conducted experimental exposures using a trematode (Sphaeridiotrema pseudoglobulus) implicated in waterfowl die-offs and found that CMS infection levels were significantly lower than those in co-occurring snail species. Furthermore, the parasites that did successfully infect CMS were often found encased in the shells of the snails in a non-viable state. Together these results 1) provide insight into the importance of CMS as a host for parasites in the region, and 2) may help to explain the wide distribution of CMS across WI and the Midwest

This repository contains the data file associated with all surveys utilized in the analyses presented in this research article.Format of Data Files: Data files are in .csv format. The codebook contains the variable names, variable labels, and value labels for all variables contained within the "Dataset.csv" file.An abundance of literature has examined barriers to women's equal representation in science, technology, engineering, and math (STEM) fields, with many studies showing that STEM fields are not perceived to afford communal goals, a key component of women's interest in future careers. Using Goal Congruity Theory as a framework, we tested the longitudinal impact of perceptions of STEM career goal affordances, communal and agentic goals, and their congruity on persistence in science from the second through fourth years of college among women in STEM majors. We found that women's intent to persist in science were highest in fall of their second year, that persistence intentions exhibited a sharp decline, and that eventually leveled off by their fourth year of college. This pattern was moderated by perceptions of agentic affordances in STEM, such that women with higher perceptions of agentic affordances experienced smaller declines. Similar to prior research, we found that higher perceptions of communal goal affordances in STEM consistently predicted higher persistence intentions. Finally, we found an agentic goal-affordance congruity interaction, such that higher perceptions of agentic affordances in STEM predicted higher persistence intentions; however, the positive relationship was stronger for women with higher agentic goals. We conclude that because STEM fields are stereotyped as affording agentic goals, women who identify interest in a STEM major during their first year of college may be drawn to these fields for this reason, and may benefit from perceptions that STEM affords both communal and agentic goals.Funding for this work was provided by the National Science Foundation through grant number DUE- 1431795, DUE-1431823, and DUE-1460229

    Role modeling is a viable retention strategy for undergraduate women in the geosciences

    Gender diversity leads to better science; however, a number of science, technology, engineering, and mathematics (STEM) disciplines, including many geoscience subdisciplines, show a persistent gender gap. PROmoting Geo- science Research, Education, and SuccesS (PROGRESS) is a theory-driven role modeling and mentoring program aimed at supporting undergraduate women interested in geoscience-related degree and career pathways. This study is unique because it is being conducted in a long-term applied setting, rather than as a laboratory exercise. We compare female STEM majors in PROGRESS to a matched control group (N = 380) using a longitudinal prospec- tive multisite quasi-experimental design. College women in PROGRESS par- ticipated in a mentoring and role-modeling weekend workshop with follow- up support, while women in the control group participated in neither the workshop nor the follow-up support. PROGRESS members identified more female STEM career role models than controls (60% versus 42%, respectively), suggesting that deliberate interventions can develop the networks of under- graduate women. Undergraduate women that participate in PROGRESS have higher rates of persistence in geoscience-related majors (95% versus 73%), although the rates of switching into a geoscience-related major did not differ across groups. More strikingly, we also find that the persistence of undergrad- uate women in geoscience-related majors is related to the number of female STEM career role models they identify, as their odds of persisting approxi- mately doubles for each role model they identify. We conclude that our ability to retain undergraduate women in the geosciences will depend, in part, on helping them to identify same-gender career role models. Further, the suc- cess of PROGRESS points to steps universities and departments can take to sustain their students’ interest and persistence, such as hosting interactive panels with diverse female scientists to promote the attainability and social relevance of geoscience careers

    3D-cultured blastoids model human embryogenesis from pre-implantation to early gastrulation stages

    Naive human pluripotent stem cells have the remarkable ability to self-organize into blastocyst-like structures ( blastoids ) that model lineage segregation in the pre-implantation embryo. However, the extent to which blastoids can recapitulate the defining features of human post-implantation development remains unexplored. Here, we report that blastoids cultured on thick three-dimensional (3D) extracellular matrices capture hallmarks of early post-implantation development, including epiblast lumenogenesis, rapid expansion and diversification of trophoblast lineages, and robust invasion of extravillous trophoblast cells by day 14. Extended blastoid culture results in the localized activation of primitive streak marker TBXT and the emergence of embryonic germ layers by day 21. We also show that the modulation of WNT signaling alters the balance between epiblast and trophoblast fates in post-implantation blastoids. This work demonstrates that 3D-cultured blastoids offer a continuous and integrated in vitro model system of human embryonic and extraembryonic development from pre-implantation to early gastrulation stages

    RAHI-SATHI Indo-U.S. Collaboration: The Evolution of a Trainee-Led Twinning Model in Global Health Into a Multidisciplinary Collaborative Program

    BACKGROUND: In recent years there has been a surge in the number of global health programs operated by academic institutions. However, most of the existing programs describe partnerships that are primarily faculty-driven and supported by extramural funding. PROGRAM DESCRIPTION: Research and Advocacy for Health in India (RAHI, or pathfinder in Hindi) and Support and Action Towards Health-Equity in India (SATHI, or partnership in Hindi) are 2 interconnected, collaborative efforts between the University of Massachusetts Medical School (UMMS) and Charutar Arogya Mandal (CAM), a medical college and a tertiary care center in rural western India. The RAHI-SATHI program is the culmination of a series of student/trainee-led research and capacity strengthening initiatives that received institutional support in the form of faculty mentorship and seed funding. RAHI-SATHI\u27s trainee-led twinning approach overcomes traditional barriers faced by global health programs. Trainees help mitigate geographical barriers by acting as a bridge between members from different institutions, garner cultural insight through their ability to immerse themselves in a community, and overcome expertise limitations through pre-planned structured mentorship from faculty of both institutions. Trainees play a central role in cultivating trust among the team members and, in the process, they acquire personal leadership skills that may benefit them in their future careers. CONCLUSION: This paradigm of trainee-led twinning partnership promotes sustainability in an uncertain funding climate and provides a roadmap for conducting foundational work that is essential for the development of a broad, university-wide global health program

    Degradation of GSPT1 causes TP53-independent cell death in leukemia whilst sparing normal hematopoietic stem cells

    Targeted protein degradation is a rapidly advancing and expanding therapeutic approach. Drugs that degrade GSPT1 via the CRL4CRBN ubiquitin ligase are a new class of cancer therapy in active clinical development with evidence of activity against acute myeloid leukemia in early phase trials. However, other than activation of the integrated stress response, the downstream effects of GSPT1 degradation leading to cell death are largely undefined, and no murine models are available to study these agents. We identified the domains of GSPT1 essential for cell survival and show that GSPT1 degradation leads to impaired translation termination, activation of the integrated stress response pathway, and TP53-independent cell death. CRISPR-Cas9 screens implicated decreased translation initiation as protective to GSPT1 degradation, suggesting that cells with higher levels of translation are more susceptible to GSPT1 degradation. We defined two Crbn amino acids that prevent Gspt1 degradation in mice, generated a knock-in mouse with alteration of these residues, and demonstrated the efficacy of GSPT1-degrading drugs in vivo with relative sparing of numbers and function of long-term hematopoietic stem cells. Our results provide a mechanistic basis for the use of GSPT1 degraders for the treatment of cancer, including TP53-mutant AML

    Disturbance and Recovery of Salt Marsh Arthropod Communities following BP Deepwater Horizon Oil Spill

    Oil spills represent a major environmental threat to coastal wetlands, which provide a variety of critical ecosystem services to humanity. The U.S. Gulf of Mexico is a hub of oil and gas exploration activities that historically have impacted intertidal habitats such as salt marsh. Following the BP Deepwater Horizon oil spill, we sampled the terrestrial arthropod community and marine invertebrates found in stands of Spartina alterniflora, the most abundant plant in coastal salt marshes. Sampling occurred in 2010 as oil was washing ashore and a year later in 2011. In 2010, intertidal crabs and terrestrial arthropods (insects and spiders) were suppressed by oil exposure even in seemingly unaffected stands of plants; however, Littoraria snails were unaffected. One year later, crab and arthropods had largely recovered. Our work is the first attempt that we know of assessing vulnerability of the salt marsh arthropod community to oil exposure, and it suggests that arthropods are both quite vulnerable to oil exposure and quite resilient, able to recover from exposure within a year if host plants remain healthy
