    Professor Pimenta Claro (1957-2018): Pioneer in dynamics of mechanical systems at the University of Minho

    This work highlights the importance of Professor Pimenta Claro in the genesis and development of a new scientific area at the Department of Mechanical Engineering of the University of Minho, namely Dynamics of Mechanical Systems. Professor Pimenta Claro initiated his academic career in October 1980, coming from industry, where he was a well-recognized engineer in the field of mechanical design. Professor Pimenta Claro concluded his Pedagogical Aptitude and Scientific Capacity Tests (PAPCC) – MSc equivalent – in 1987, with dissertation title “Estudo Comparativo das Previsões Teóricas do Desempenho de Chumaceiras Radiais Hidrodinâmicas com Resultados Experimentais”. Professor Pimenta Claro received his doctorate degree in 1994 with thesis “Reformulação de Método de Cálculo de Chumaceiras Radiais Hidrodinâmicas – Análise do Desempenho Considerando Condições de Alimentação” under the mentorship of Professor Sousa Miranda, which was in fact the first PhD in Mechanical Engineering defended at the University of Minho. In 1997, Professor Pimenta Claro broken with his past background – classical tribology – to open a new research domain – Dynamics of Mechanical Systems. Since then, Professor Pimenta Claro has coordinated and participated in several scientific projects both with national and international partners, as well as projects with industrial partners. Professor Pimenta Claro was author of numerous publications, including scientific papers, books, conference papers, etc., and supervised PhD and MSc students. From 2007 to 2013 he coordinated the research group called Dynamics of Mechanical Systems. Professor Pimenta Claro was also pioneer and responsible for the creation of new courses on dynamics of mechanical systems offered in different degrees at the School of Engineering of the University of Minho. Thus, the main purpose of this work is to highlight Pimenta Claro’s contributions to the vast scientific area of Dynamics of Mechanical Systems at the Department of Mechanical Engineering of the University of Minho

    Characterization of Unplanned Metropolitan Wireless Networks

    Mobile Internet penetration has grown steadily over the last few years. Although most of today’s users have access through their 3G Mobile Operators, there are still regions that are under-covered for various reasons. Wireless Mesh Networks (WMN) can play an important role by providing the means to fully cover those underserved regions. Due to their intrinsic nature, WMN require a critical mass of nodes belonging to the mesh in order to be effective. In this paper we present a study conducted in Aveiro, Portugal which intends to draw some conclusions on the feasibility of deploying a WMN in small to medium cities based on the cooperation of its inhabitants and on off-the-shelf wireless equipment

    Análise da estabilidade de pilares de zircónia 3Y-TZP após carga cíclica e termociclagem

    Objectives: Although implant abutments made of yttria-stabilized tetragonal zirconia poly crystal (3Y-TZP) show great predictability, some issues related to its aging require further study. The objective of this in vitro study is to assess the stability of 3Y-TZP zirconia implant abutments with two different implant connections when subjected to aging simulation through thermocycling and mechanical loading (TCML).Methods: Ten 3Y-TZP zirconia abutments were selected and equally divided into two groups (n = 5): CEZr, abutments for externally hexed implants; and CIZr, abutments for implants with an internal conical connection. The samples were subjected to thermocycling (5000 cycles; 5-55 C) and mechanical loading (1.2 x 106 cycles; 88.8 N; 4 Hz). Before and after the aging procedures, X-ray diffraction (XRD) analysis was conducted to observe tetragonal monoclinic (t-m) phase transformation, and topographic surface analysis was performed by 3D profilometry, and data were analyzed using Mann-Witney test (p 0.05), before and after TCML, and between implants connections (test U= 57.0, p = 0.053 > 0.05) were not detected.Conclusions: After a 5-year simulation of its clinical use, the analyzed 3Y-TZP zirconia abutments did not show signs of aging. The connection's geometry does not interfere in aging.Objetivos Os pilares de zircónia tetragonal policristalina (Y-TZP) apresentam grande previsibilidade na sua utilização clínica. No entanto, existem ainda alguns aspetos relacionados com o envelhecimento que necessitam de ser estudados. O presente trabalho pretendeu avaliar a estabilidade de pilares de zircónia 3Y-TZP de 2 conexões implantares diferentes, quando submetidos a simulação de envelhecimento (TCML). Métodos Foram selecionados 10 pilares em zircónia 3Y-TZP, que foram divididos em 2 grupos (n = 5): CEZr, pilares para implantes de conexão de hexágono externo; e CIZr, pilares para implantes de conexão interna cónica-lobular. As amostras foram submetidas a termociclagem (5000 ciclos; 5-55 °C) e carga cíclica (1,2 × 106 ciclos; 88,8 N; 4 Hz). Antes e após TCML, os pilares foram sujeitos a análise por difração de raios X para determinação de alteração de fase cristalográfica de tetragonal para monoclínica (t-m) e a interferometria ótica sobre a superfície de assentamento das 2 conexões para a medição da sua topografia por perfilometria 3D. Análise estatística: Mann-Whitney test (p < 0,05). Resultados Após TCML não foi detetada fase monoclínica em nenhum dos pilares. A análise comparativa da rugosidade (com utilização do parâmetro Sa) sobre as plataformas protéticas dos pilares revelou ligeiro aumento da Sa em ambas as conexões após TCML. Não foram observadas diferenças estatisticamente significativas nos valores de Sa (test U = 57,0, p = 0,161 > 0,05), antes e após TCML, e entre conexões dos implantes (test U = 57,0, p = 0,053 > 0,05). Conclusões Após uma simulação de 5 anos de utilização clínica, os pilares de zircónia 3Y-TZP analisados não apresentaram sinais de envelhecimento.     Alternative models for the calculation of the RMR and Q indexes for granite rock masses

    Empirical classification systems like the RMR and Q are often used in current practice of geotechnical structures design built in rock masses. They allow obtaining an overall description of the rock mass and the calculation, through analytical solutions, of strength and deformability parameters which are determinant in design. To be applied these systems need a set of geomechanical information that may not be available or can be difficult to obtain. In this work it is intended to develop new alternative regression models for the calculation of the RMR and Q indexes using less data than the original formulations and keeping a high accuracy level. It is also intended to have an insight of which parameters are the most important for the prediction of the indexes and in the rock masses behaviour. This study was carried out applying Data Mining techniques to a database of the empirical classification systems applications in a granite rock mass. Data Mining is a relatively new area of computer science which concerns with automatically find, simplify and summarize patterns and relationships within large databases. The used Data Mining techniques were the multiple regression and artificial neural networks.     A synthetic biology approach to engineer "therapeutic" bacteria

    The high incidence and mortality of solid tumors like breast cancer makes the development of novel therapeutic agents a high priority. Curcumin, a natural substance from the rhizome of Curcuma longa, has captured the attention of the scientific community. Pre-clinical trials and extensive research has demonstrated its ability to prevent cancer. Indeed, curcumin has been shown to target critical genes involved in angiogenesis, apoptosis, cell cycle and metastasis, and consequently to inhibit cell growth. Currently, the clinical use of curcumin is mainly limited by its poor bioavailability which implies repetitive oral doses in order to achieve the therapeutic concentrations inside the cell. The idea of the present work is to design a strategy that could link the common technique used to treat solid tumors (ultrasound) with the therapeutic effects of curcumin. The plan is to use the temperature increase (consequence of ultrasound treatment) to trigger the in situ expression of curcumin by engineered bacteria. Escherichia coli was chosen as the model organism in which the genes involved in the curcumin pathway will be cloned. Those genes (4-coumarate: CoA ligase, diketide-CoA synthase and curcumin synthase) were successfully cloned under the control of a temperature sensitive promoter (dnaK). The proof-of-concept that the dnaK promoter can be induced by a temperature increase, leading to the expression of the 3 necessary genes, is currently being tested, using several biochemical assays. Moreover, several knockouts (KO) of specific genes from the E. coli K-12 MG1655 genome were performed in order to maximize the production of curcumin. The deletion strategy, as well as the definition of the non-essential genes to be KO, was determined in silico. This strategy included one single KO (gnd gene) and the multiple KO of five non-essential genes for aerobic growth (fumA, fumB, fumC, ccmA and argO) and serA gene for anaerobic growth. After optimizing the genes expression under the control of the temperature inducible promoter, the several KO will be transformed with this construction to confirm the improvement of curcumin production

    Gonorrhea in a tertiary care Portuguese hospital : a 10-year retrospective study of the evolution of cases and drug resistance of the isolates

Although traditionally considered treatable, the emergence of Neisseria gonorrhoeae resistance to antimicrobials is currently a serious problem. The goal of this study was to evaluate the incidence and trends of antimicrobial resistance over the last 10 years in N. gonorrhoeae isolates from a Portuguese Centre. Methods: Laboratorial confirmed N. gonorrhoeae infections diagnosed between 2009 and 2018 were evaluated. Susceptibilities to penicillin, tetracycline, ciprofloxacin, azithromycin and cefotaxime were studied, along with demographic and clinical characteristics. Results: From 2009 to 2018, 440 cases of N. gonorrhoeae infection were diagnosed in our center, with a significant yearly increase (p<0.05). Most cases occurred in males (97.9%), with a median age of 25 years. In 88.7% of the cases, treatment with ceftriaxone plus azithromycin was used. Resistances to penicillin, tetracycline and ciprofloxacin remained high throughout the study period. Conclusions: Antimicrobial resistance of N. gonorrhoeae appeared shortly after the introduction of antimicrobials. To combat this problem, improved surveillance and more studies combining susceptibility and epidemiological data are needed. In our population, N. gonorrhoeae remains highly susceptible to the antibiotics currently recommended for its treatment, whereas ciprofloxacin, azithromycin (in monotherapy) and penicillin should be avoided as empirical treatment.info:eu-repo/semantics/publishedVersio

    Molecular diversity within the genus Laeonereis (Annelida, Nereididae) along the west Atlantic coast: paving the way for integrative taxonomy

    The polychaete genus Laeonereis (Annelida, Nereididae) occurs over a broad geographic range and extends nearly across the entire Atlantic coast of America, from the USA to Uruguay. Despite the research efforts to clarify its diversity and systematics, mostly by morphological and ecological evidence, there is still uncertainty, mainly concerning the species Laeonereis culveri, which constitutes an old and notorious case of taxonomic ambiguity. Here, we revised the molecular diversity and distribution of Laeonereis species based on a multi-locus approach, including DNA sequence analyses of partial segments of the cytochrome c oxidase subunit I (COI), 16S rRNA, and 28S rRNA genes. We examined Laeonereis specimens collected from 26 sites along the American Atlantic coast from Massachusetts (USA) to Mar del Plata (Argentina). Although no comprehensive morphological examination was performed between different populations, the COI barcodes revealed seven highly divergent MOTUs, with a mean K2P genetic distance of 16.9% (from 6.8% to 21.9%), which was confirmed through four clustering algorithms. All MOTUs were geographically segregated, except for MOTUs 6 and 7 from southeastern Brazil, which presented partially overlapping ranges between Rio de Janeiro and Sao Paulo coast. Sequence data obtained from 16S rRNA and 28S rRNA markers supported the same MOTU delimitation and geographic segregation as those of COI, providing further evidence for the existence of seven deeply divergent lineages within the genus. The extent of genetic divergence between MOTUs observed in our study fits comfortably within the range reported for species of polychaetes, including Nereididae, thus providing a strong indication that they might constitute separate species.     Altered bone microarchitecture in a type 1 diabetes mouse model Ins2 Akita

    Type 1 diabetes mellitus (T1DM) has been associated to several cartilage and bone alterations including growth retardation, increased fracture risk, and bone loss. To determine the effect of long term diabetes on bone we used adult and aging Ins2(Akita) mice that developed T1DM around 3-4 weeks after birth. Both Ins2(Akita) and wild-type (WT) mice were analyzed at 4, 6, and 12 months to assess bone parameters such as femur length, growth plate thickness and number of mature and preapoptotic chondrocytes. In addition, bone microarchitecture of the cortical and trabecular regions was measured by microcomputed tomography and gene expression of Adamst-5, Col2, Igf1, Runx2, Acp5, and Oc was quantified by quantitative real-time polymerase chain reaction. Ins2(Akita) mice showed a decreased longitudinal growth of the femur that was related to decreased growth plate thickness, lower number of chondrocytes and to a higher number of preapoptotic cells. These changes were associated with higher expression of Adamst-5, suggesting higher cartilage degradation, and with low expression levels of Igf1 and Col2 that reflect the decreased growth ability of diabetic mice. Ins2(Akita) bone morphology was characterized by low cortical bone area (Ct.Ar) but higher trabecular bone volume (BV/TV) and expression analysis showed a downregulation of bone markers Acp5, Oc, and Runx2. Serum levels of insulin and leptin were found to be reduced at all-time points Ins2(Akita). We suggest that Ins2(Akita) mice bone phenotype is caused by lower bone formation and even lower bone resorption due to insulin deficiency and to a possible relation with low leptin signaling.F. R.
