    Editorial: Towards an Archaeology of Television

    Over the last few years, ‘media archaeology’ has evolved from a marginal topic to an academic approach en vogue. Under its banner, conferences and publications bring together scholars from different disciplines who, revisiting the canon of media history and theory, emphasize the necessity for renewed historiographical narratives. Despite, or maybe because of profuse debates, media archaeology remains a loosely defined playground for researchers working at the intersection of history and theory. Far from offering uniform principles or constituting a homogeneous field, its prominent authors – Friedrich Kittler and Wolfgang Ernst, Siegfried Zielinksi, Jussi Parrika and Erkki Huhtamo, to name just a few – distinguish themselves by their heterogeneity regarding methodology and theoretical focus

    The popular music heritage of the Dutch pirates: illegal radio and cultural identity

    This article explores how cultural identities are negotiated in relation to the heritage of illegal radio in the Netherlands. The term ‘pirate radio’ commonly refers to the offshore radio stations that were broadcasting during the 1960s. These stations introduced commercial radio and popular music genres like beat music, which were not played by public broadcasters at the time. In their wake, land-based pirates began broadcasting for local audiences. This study examines the identities that are constituted by the narrative of pirate radio. Drawing on in-depth interviews with archivists, fans and broadcasters, this article explores the connection between pirate radio, popular music heritage and cultural identity. Moreover, it considers how new technologies such as internet radio provide platforms to engage with this heritage and thus to maintain these local identities. To examine how the memories of pirate radio live on in the present a narrative approach to identity will be used

    Nitrogen sources on TPOMW valorization through solid state fermentation performed by Yarrowia lipolytica

    This manuscript reports the valorization of two-phase olive mill waste (TPOMW) as raw material and carbon source for solid state fermentation using Yarrowia lipolytica as biocatalyst. Due to its chemical characteristics, a combination of different raw materials (TPOMW and wheat bran, WB) was evaluated and two distinct nitrogen sources were applied as supplementation for lipase production. A TPOMW/WB ratio of 1:1 and supplementation with ammonium sulfate was chosen as the best condition. The productivity in 24 h reached 7.8 U/gh and, after four days of process, only decreased about 35%. Process pH ranged from 5.5-5.9, remaining in an acid range. Thus, the successful use of TPOMW, a watery solid by-product with high content of lipids, as raw material for Yarrowia lipolytica growth and lipase production provided an environmental friendly alternative to valorize such waste.The authors kindly acknowledge the financial aid and research scholarships given by CAPES.     Identification and characterization of DGA2, an acyltransferase of the DGAT1 acyl-CoA:diacylglycerol acyltransferase family in the oleaginous yeast Yarrowia lipolytica. New insights into the storage lipid metabolism of oleaginous yeasts

    Triacylglycerols (TAG) and steryl esters (SE) are the principal storage lipids in all eukaryotic cells. In yeasts, these storage lipids accumulate within special organelles known as lipid bodies (LB). In the lipid accumulation-oriented metabolism of the oleaginous yeast Yarrowia lipolytica, storage lipids are mostly found in the form of TAG, and only small amounts of SE accumulate. We report here the identification of a new DAG acyltransferase gene, DGA2, homologous to the ARE genes of Saccharomyces cerevisiae. This gene encodes a member of the type 1 acyl-CoA:diacylglycerol acyltransferase family (DGAT1), which has not previously been identified in yeasts, but is commonly found in mammals and plants. Unlike the Are proteins in S. cerevisiae, Dga2p makes a major contribution to TAG synthesis via an acyl-CoA-dependent mechanism and is not involved in SE synthesis. This enzyme appears to affect the size and morphology of LB, suggesting a direct role of storage lipid proteins in LB formation. We report that the Are1p of Y. lipolytica was essential for sterol esterification, as deletion of the encoding gene (ARE1) completely abolished SE synthesis. Unlike its homologs in yeasts, YlARE1 has no DAG acyltransferase activity. We also reconsider the role and function of all four acyltransferase enzymes involved in the final step of neutral lipid synthesis in this oleaginous yeast

    Biotechnological production of Îł-decalactone, a peach like aroma, by Yarrowia lipolytica

    The request for new flavourings increases every year. Consumer perception that everything natural is better is causing an increase demand for natural aroma additives. Biotechnology has become a way to get natural products. Îł-Decalactone is a peach-like aroma widely used in dairy products, beverages and others food industries. In more recent years, more and more studies and industrial processes were endorsed to cost-effect this compound production. One of the best-known methods to produce -decalactone is from ricinoleic acid catalyzed by Yarrowia lipolytica, a generally regarded as safe status yeast. As yet, several factors affecting -decalactone production remain to be fully understood and optimized. In this review, we focus on the aromatic compound -decalactone and its production by Y. lipolytica. The metabolic pathway of lactone production and degradation are addressed.     The Kolumbo submarine volcano of Santorini island is a large pool of bacterial strains with antimicrobial activity

    Microbes in hydrothermal vents with their unique secondary metabolism may represent an untapped potential source of new natural products. In this study, samples were collected from the hydrothermal field of Kolumbo submarine volcano in the Aegean Sea, in order to isolate bacteria with antimicrobial activity. Eight hundred and thirty-two aerobic heterotrophic bacteria were isolated and then differentiated through BOX-PCR analysis at the strain level into 230 genomic fingerprints, which were screened against 13 different type strains (pathogenic and nonpathogenic) of Gram-positive, Gram-negative bacteria and fungi. Forty-two out of 176 bioactive-producing genotypes (76 %) exhibited antimicrobial activity against at least four different type strains and were selected for 16S rDNA sequencing and screening for nonribosomal peptide (NRPS) and polyketide (PKS) synthases genes. The isolates were assigned to genus Bacillus and Proteobacteria, and 20 strains harbored either NRPS, PKS type I or both genes. This is the first report on the diversity of culturable mesophilic bacteria associated with antimicrobial activity from Kolumbo area; the extremely high proportion of antimicrobial-producing strains suggested that this unique environment may represent a potential reservoir of novel bioactive compounds
