Regional Assessment of Soybean Brown Stem Rot, Phytophthora sojae, and Heterodera glycines Using Area-Frame Sampling: Prevalence and Effects of Tillage
The prevalence of brown stem rot (caused by Phialophora gregata), Heterodera glycines, and Phytophthora sojae in the north central United States was investigated during the fall of 1995 and 1996. Soybean fields were randomly selected using an area-frame sampling design in collaboration with the National Agricultural Statistics Service. Soil and soybean stem samples, along with tillage information, were collected from 1,462 fields in Illinois, Iowa, Minnesota, Missouri, and Ohio. An additional 275 soil samples collected from Indiana were assessed for H. glycines. For each field, the incidence and prevalence of brown stem rot was assessed in 20 soybean stem pieces. The prevalence and recovery (expressed as the percentage of leaf disks colonized) of P. sojae and the prevalence and population densities of H. glycines were determined from the soil samples. The prevalence of brown stem rot ranged from 28% in Missouri to 73% in Illinois; 68 and 72% of the fields in Minnesota and Iowa, respectively, showed symptomatic samples. The incidence of brown stem rot was greater in conservation-till than in conventional-till fields in all states except Minnesota, which had few no-till fields. P. sojae was detected in two-thirds of the soybean fields in Ohio and Minnesota, whereas 63, 55, and 41% of the fields in Iowa, Missouri, and Illinois, respectively, were infested with the pathogen. The recovery rates of P. sojae were significantly greater (P ⤠0.05) in conservation-till than in conventional-till fields in all states except Iowa. H. glycines was detected in 83% of the soybean fields in Illinois, 74% in Iowa, 71% in Missouri, 60% in Ohio, 54% in Minnesota, and 47% in Indiana. Both the prevalence and population densities of H. glycines were consistently greater in tilled than in no-till fields in all states for which tillage information was available
Hypothenar hammer syndrome: Proposed etiology
AbstractPurpose: Finger ischemia caused by embolic occlusion of digital arteries originating from the palmar ulnar artery in a person repetitively striking objects with the heel of the hand has been termed hypothenar hammer syndrome (HHS). Previous reports have attributed the arterial pathology to traumatic injury to normal vessels. A large experience leads us to hypothesize that HHS results from trauma to intrinsically abnormal arteries. Methods: We reviewed the arteriography, histology, and clinical outcome of all patients treated for HHS in a university clinical research center study of hand ischemia, which prospectively enrolled more than 1300 subjects from 1971 to 1998. Results: Twenty-one men had HHS. All had occupational (mechanic, carpenter, etc) or avocational (woodworker) exposure to repetitive palmar trauma. All patients underwent upper-extremity and hand arteriography, unilateral in eight patients (38%) and bilateral in 13 patients (62%). By means of arteriogram, multiple digital artery occlusions were shown in the symptomatic hand, with either segmental ulnar artery occlusion in the palm or characteristic âcorkscrewâ elongation, with alternating stenoses and ectasia. Similar changes in the contralateral asymptomatic (and less traumatized) hand were shown by means of 12 of 13 bilateral arteriograms (92%). Twenty-one operations, consisting of segmental ulnar artery excision in the palm and vein grafting, were performed on 19 patients. Histology was compatible with fibromuscular dysplasia with superimposed trauma. Patency of arterial repairs at 2 years was 84%. One patient (5%) required amputative debridement of necrotic finger tips. No other tissue loss occurred. There have been no recurrences of ischemia in patients with patent bypass grafts. Conclusion: To our knowledge, this is the largest reported group of HHS patients. The characteristic angiographic appearance, histologic findings, and striking incidence of bilateral abnormalities in patients with unilateral symptoms lead us to conclude that HHS occurs when persons with preexisting palmar ulnar artery fibrodysplasia experience repetitive palmar trauma. This revised theory for the etiology of HHS explains why HHS does not develop in most patients with repetitive palmar trauma. (J Vasc Surg 2000;31:104-13.
Regulation of beta-amyloid production in neurons by astrocyte-derived cholesterol
Alzheimerâs disease (AD) is characterized by the presence of amyloid β (Aβ) plaques, tau tangles, inflammation, and loss of cognitive function. Genetic variation in a cholesterol transport protein, apolipoprotein E (apoE), is the most common genetic risk factor for sporadic AD. In vitro evidence suggests that apoE links to Aβ production through nanoscale lipid compartments (lipid clusters), but its regulation in vivo is unclear. Here, we use superresolution imaging in the mouse brain to show that apoE utilizes astrocyte-derived cholesterol to specifically traffic neuronal amyloid precursor protein (APP) in and out of lipid clusters, where it interacts with β- and Îł-secretases to generate Aβ-peptide. We find that the targeted deletion of astrocyte cholesterol synthesis robustly reduces amyloid and tau burden in a mouse model of AD. Treatment with cholesterol-free apoE or knockdown of cholesterol synthesis in astrocytes decreases cholesterol levels in cultured neurons and causes APP to traffic out of lipid clusters, where it interacts with Îą-secretase and gives rise to soluble APP-Îą (sAPP-Îą), a neuronal protective product of APP. Changes in cellular cholesterol have no effect on Îą-, β-, and Îł-secretase trafficking, suggesting that the ratio of Aβ to sAPP-Îą is regulated by the trafficking of the substrate, not the enzymes. We conclude that cholesterol is kept low in neurons, which inhibits Aβ accumulation and enables the astrocyte regulation of Aβ accumulation by cholesterol signaling
Small Group Tutorials in Radiology: a Pilot Study
Abstract Rationale: Radiology plays an integral role in the diagnosis and management of disease. As diagnostic images are frequently reviewed by junior doctors prior to the formal Radiology report being issued, it is essential that they are armed with the skills to correctly interpret pathological findings, especially out of hours. Effective radiology teaching is of considerable benefit to patients as critical findings are less likely to be missed or misinterpreted. Methodology: Quantitative methods were utilised for data collection. Sampling was purposive as questionnaires were disseminated at the end of the series of small group tutorials. The response rate was 81.4 % (228/280). Findings: The data extracted from this research demonstrated the positive impact the change initiative had on the 228 participants. 91% of postgraduates and 84% of undergraduates rated the small group tutorials as either essential or very helpful. 100% of participants would like more radiology teaching with 85% preferring the small group tutorial format. Worryingly, 85% of junior doctor's and 44 % of undergraduates felt that their radiology training had insufficiently prepared them for the clinical duties of a junior doctor. Conclusion: This research supports the implementation of small group tutorials in radiology into the medical undergraduate and postgraduate curriculum
Use of MMG signals for the control of powered orthotic devices: Development of a rectus femoris measurement protocol

The protocol is specified in terms of the following: measurement equipment, signal processing requirements, human postural requirements, test rig, sensor placement, sensor dermal fixation, and test procedure. Preliminary tests of the statistical nature of rectus femoris MMG signals were performed, and Gaussianity was evaluated by means of a two-sided Kolmogorov-Smirnov test. For all 100 MMG data sets obtained from the testing of two volunteers, the null hypothesis of Gaussianity was rejected at the 1%, 5%, and 10%âsignificance levels. Most skewness values were found to be greater than 0.0, while all kurtosis values were found to be greater than 3.0. A statistical convergence analysis also performed on the same 100 MMG data sets suggested that 25 MMG acquisitions should prove sufficient to statistically characterize rectus femoris MMG. This conclusion is supported by the qualitative characteristics of the mean rectus femoris MMG power spectral densities obtained using 25 averages
Phenotype of p53 wild-type epitope-specific T cells in the circulation of patients with head and neck cancer
CD8(+) cytotoxic T-cell (CTL) specific for non-mutated, wild type (wt) sequence p53 peptides derived from wt or mutant p53 molecules expressed in head and neck squamous cell carcinomas (HNSCC) have been detected in the circulation of patients with this disease. The frequency and differentiation/maturation phenotypes of these anti-tumor specific CTL can reflect the host's immunologic response. Therefore, we investigated the frequency and phenotypes of wt sequence p53 peptide-specific CTL in patients with HNSCC (n = 33) by flow cytometric analysis using HLA-A*0201 tetrameric peptides (tet) complexed with the wt sequence p53(264-272) or p53(149-157) peptide and co-staining with phenotypic markers. One main finding was that increasing frequencies of tet(+) CD8(+) T cells in patients' circulation correlated with increased frequencies of inactive naive tet(+) cells, while those with effector memory and terminally differentiated phenotypes, which are associated with positive anti-tumor immune responses, decreased. We also found that the frequency of circulating tet(+) CD8(+) T cells negatively correlated with p53 expression in tumor tissues and tumor stage. Our findings support further clinical-based investigations to define the frequencies and phenotypes of wt sequence p53 peptide-specific CD8(+) T cells to predict disease severity, enhance selection of patients for inclusion in vaccination trials and highlight prerequisites to enhance immune susceptibility by activation of inactive naive tet+ T cells and/or enhancing circulating effector T cell activity by checkpoint blockage

Drawing on Redhead's discussion of Baudrillard as a theorist of hyperreality, the paper considers the different ways in which the mediatized âliveâ football spectacle is often modelled on the âliveâ however eventually usurps the âliveâ forms position in the cultural economy, thus beginning to replicate the mediatized âliveâ. The blurring of the âliveâ and ârealâ through an accelerated mediatization of football allows the formation of an imagined community mobilized by the working class whilst mediated through the sanitization, selling of âeventsâ and the middle classing of football, through the re-encoding of sporting spaces and strategic decision-making about broadcasting. A culture of pub supporting then allows potential for working-class supporters to remove themselves from the panoptican gazing systems of late modern hyperreal football stadia and into carnivalesque performative spaces, which in many cases are hyperreal and simulated themselves
