Access to consciousness of briefly presented visual events is modulated by transcranial direct current stimulation of left dorsolateral prefrontal cortex
Adaptive behaviour requires the ability to process goal-relevant events at the expense of irrelevant ones. However, perception of a relevant visual event can transiently preclude access to consciousness of subsequent events — a phenomenon called attentional blink (AB). Here we investigated involvement of the left dorsolateral prefrontal cortex (DLPFC) in conscious access, by using transcranial direct current stimulation (tDCS) to potentiate or reduce neural excitability in the context of an AB task. In a sham-controlled experimental design, we applied between groups anodal or cathodal tDCS over the left DLPFC, and examined whether this stimulation modulated the proportion of stimuli that were consciously reported during the AB period. We found that tDCS over the left DLPFC affected the proportion of consciously perceived target stimuli. Moreover, anodal and cathodal tDCS had opposing effects, and exhibited different temporal patterns. Anodal stimulation attenuated the AB, enhancing conscious report earlier in the AB period. Cathodal stimulation accentuated the AB, reducing conscious report later in the AB period. These findings support the notion that the DLPFC plays a role in facilitating information transition from the unconscious to the conscious stage of processing
Cognitive and personality components underlying spoken idiom comprehension in context. An exploratory study.
In this exploratory study, we investigated whether and to what extent individual differences in cognitive and personality variables are associated with spoken idiom comprehension in context. Language unimpaired participants were enrolled in a cross-modal lexical decision study in which semantically ambiguous Italian idioms (i.e., strings with both a literal and an idiomatic interpretation as, for instance, break the ice), predictable or unpredictable before the string offset, were embedded in idiom-biasing contexts. To explore the contributions of different cognitive and personality components, participants also completed a series of tests respectively assessing general speed, inhibitory control, short-term and working memory, cognitive flexibility, crystallized and fluid intelligence, and personality. Stepwise regression analyses revealed that online idiom comprehension was associated with the participants\u2019 working memory, inhibitory control and crystallized verbal intelligence, an association modulated by idiom type. Also personality-related variables (State Anxiety and Openness to Experience) were associated with idiom comprehension, although in marginally significant ways. These results contribute to the renewed interest on how individual variability modulates language comprehension, and for the first time document contributions of individual variability on lexicalized, high frequency multi-word expressions as idioms adding new knowledge to the existing evidence on metaphor and sarcasm
Anodal tDCS over the right parietal but not frontal cortex enhances the ability to overcome task set inhibition during task switching
Switching between tasks requires individuals to inhibit mental representations of the previous task demands and to activate representations of the new task demands. The inhibition of the executed task remains active for a while so that when the inhibited task set must be re-activated shortly after, the need to overcome residual task set inhibition leads to behavioral costs. In a sham-controlled balanced-order within-subjects experimental design we investigated whether applying right anodal/left cathodal transcranial direct current stimulation (tDCS) over the dorsolateral prefrontal or parietal cortex modulated the ability to overcome persistent task inhibition during task switching. Results showed that right anodal/left cathodal tDCS over the parietal cortex improves performance selectively when switching back to a recently inhibited task that requires previous inhibition to be overcome. Right Anodal/left cathodal tDCS over the prefrontal cortex improves performance during task switching in general, either when re-engaging in a inhibited task or when engaging in a non-inhibited task. Results suggest a different contribution of prefrontal and parietal regions to task switching, with parietal cortex being selectively involved in overcoming persistent task inhibition and prefrontal cortex being more generally involved in the control of task set during task switching
Driving attitudes, behaviours, risk perception and risk concern among young student car-drivers, motorcyclists and pedestrians in various EU countries
The number of car crashes has gradually reduced in the last decade across all European Countries, but the number of motorcycle crashes has remained nearly the same. In our research we investigate whether there are differences in attitudes towards road safety issues, driving behaviours in specific imagined situations, risk perception and risk concern, among young student drivers and riders. The study involved a large sample taken from across six European countries. The results reveal that although there are no differences between motorcyclists and car drivers in their attitudes toward road safety rules, differences do appear when the road rules compliance is assessed in specific imagined situations, with motorcyclists reporting to be more prone than car-drivers to violations of traffic rules. Moreover, despite the perceived risk during driving is the same for motorcyclists and car-drivers, differences do appear on their concern about this risk, with motorcyclists reporting to be less concerned than car-drivers about the risk of a road crash. This could lead to a high probability of risky driving behaviour in motorcyclists than in car-drivers. Present findings have important practical implications for road safety training courses

Seven horses were submitted to conventional drilling sessions, during two consecutive weeks. Blood samples were drawn before and after drilling sessions. Compared to basal values, horses submitted to drilling sessions during 2 weeks showed lower glucose levels post exercise at the 3 rd day, and higher lactate levels post exercise both after the 1st and 2nd week of drilling. Compared to basal values, Hgb
and Hct showed higher levels post exercise at the 1st day of 1st week; Plt and Pct showed higher levels post exercise at the 2nd and 3rd day of both 1st and 2nd week. HR, RR, RT showed a trend to increase in all postexercise sampling times. No significant differences were observed between the 2 drilling week sessions on hematological, hematochemical and functional values
Symptomatic carotid atherosclerotic plaques are associated with increased infiltration of natural killer (NK) cells and higher serum levels of NK activating receptor ligands
A wide array of immune cells, including lymphocytes, is known to be present and to play a pathogenetic role in atherosclerotic lesions. However, limited information is currently available regarding the presence of Natural Killer (NF cell subsets within vessel plaque, and more in general, regarding their role in human atherosclerosis. We evaluated the distribution of NK cells in human carotid atherosclerotic plaques, dissecting asymptomatic and symptomatic patients (identified as affected by stroke, transient ischemic attack, or amaurosis fugax within 6 months) with the aim of shedding light on the putative contribution of NK cells to the pathogenic process that leads to plaque instability and subsequent clinical complications. We observed that carotid plaques were consistently infiltrated by NK cells and, among them, CD56(bright)perforin(low) NK cells were abundantly present and displayed different markers of tissue residency (i.e., CD103 CD69 and CD49a). Interestingly, carotid atherosclerotic plaques of symptomatic patients showed a higher content of NK cells and an increased ratio between CD56(bright)perforin(low) NK cells and their CD56(dim)perforin(high)counterpart. NK cells isolated from plaques of symptomatic patients were also stronger producers of IFN-gamma. Analysis of the expression of NK activating receptor ligands (including MICA/B, ULBP-3, and B7-H6) in atherosclerotic carotid plaques revealed that they were abundantly expressed by a HLA-DR(+)CD11c(+) myeloid cell population resident in the plaques. Remarkably, sera of symptomatic patients contained significant higher levels of soluble ligands for NK activating receptors. Our observations indicate that CD56(bright)( )NK cells accumulate within human atherosclerotic lesions and suggest a possible contribution of NK cells to the process determining plaque instability
CB 1 Receptor Autoradiographic Characterization of the Individual Differences in Approach and Avoidance Motivation
Abstract Typically, approach behaviour is displayed in the context of moving towards a desired goal, while avoidance behaviour is displayed in the context of moving away from threatening or novel stimuli. In the current research, we detected three subpopulations of C57BL/6J mice that spontaneously responded with avoiding, balancing or approaching behaviours in the presence of the same conflicting stimuli. While the balancing animals reacted with balanced responses between approach and avoidance, the avoiding or approaching animals exhibited inhibitory or advance responses towards one of the conflicting inputs, respectively. Individual differences in approach and avoidance motivation might be modulated by the normal variance in the level of functioning of different systems, such as endocannabinoid system (ECS). The present research was aimed at analysing the ECS involvement on approach and avoidance behavioural processes. To this aim, in the three selected sub-populations of mice that exhibited avoiding or balancing or approaching responses in an approach/ avoidance Y-maze we analysed density and functionality of CB 1 receptors as well as enzyme fatty acid amide hydrolase activity in different brain regions, including the networks functionally responsible for emotional and motivational control. The main finding of the present study demonstrates that in both approaching and avoiding animals higher CB 1 receptor density in the amygdaloidal centro-medial nuclei and in the hypothalamic ventro-medial nucleus was found when compared with the CB 1 receptor density exhibited by the balancing animals. The characterization of the individual differences to respond in a motivationally based manner is relevant to clarify how the individual differences in ECS activity are associated with differences in motivational and affective functioning
Protein carbonyl group content in patients affected by familiar chronic nail candidiasis.
Familiar chronic nail candidiasis (FCNC) is a rare disorder characterized by early-onset infections caused by different species of Candida, restricted to the nail of the hands and feet, and associated with a low serum concentration of intercellular adhesion molecule 1. Host defense mechanisms against candidiasis require the cooperation of many immune cells through several candidacidal mechanisms, including oxygen-dependent killing mechanisms, mediated by a superoxide anion radical myeloperoxidase--H2O2--halide system, and reactive nitrogen intermediates. We analyzed protein carbonyl groups (considered a useful marker of oxidative stress) in the serum of patients belonging to a five-generation Italian family with an isolated form of FCNC. Serum protein carbonyl groups in FCNC patients were significantly lower than those measured in healthy donors. Also, if this hypothesis is merely speculative, we could suggest that the decreased circulating level of protein carbonyl groups in these patients is not a marker of a lower oxidative stress condition, but might be linked to a lower protease activity
A human coronavirus responsible for the common cold massively kills dendritic cells but not monocytes

Although airway epithelial cells represent an important target for infection, the respiratory epithelium is also composed of an elaborate network of dendritic cells (DCs) that are essential sentinels of the immune system, sensing pathogens and presenting foreign antigens to T lymphocytes. In this report, we show that in vitro infection by human coronavirus 229E (HCoV-229E) induces massive cytopathic effects in DCs, including the formation of large syncytia and cell death within only few hours. In contrast, monocytes are much more resistant to infection and cytopathic effects despite similar expression levels of CD13, the membrane receptor for HCoV-229E. While the differentiation of monocytes into DCs in the presence of granulocyte-macrophage colony-stimulating factor and interleukin-4 requires 5 days, only 24 h are sufficient for these cytokines to sensitize monocytes to cell death and cytopathic effects when infected by HCoV-229E. Cell death induced by HCoV-229E is independent of TRAIL, FasL, tumor necrosis factor alpha, and caspase activity, indicating that viral replication is directly responsible for the observed cytopathic effects. The consequence of DC death at the early stage of HCoV-229E infection may have an impact on the early control of viral dissemination and on the establishment of long-lasting immune memory, since people can be reinfected multiple times by HCoV-229E
Adjunctive brivaracetam and sustained seizure frequency reduction in very active focal epilepsy
Objective: This study aimed to explore the effectiveness of brivaracetam (BRV) according to baseline seizure frequency and past treatment history in subjects with focal epilepsy who were included in the Brivaracetam Add-On First Italian Network Study (BRIVAFIRST). Methods: BRIVAFIRST was a 12-month retrospective, multicenter study including adults prescribed adjunctive BRV. Study outcomes included sustained seizure response (SSR), sustained seizure freedom (SSF), and the rates of treatment discontinuation and adverse events (AEs). Baseline seizure frequency was stratified as <5, 5–20, and >20 seizures per month, and the number of prior antiseizure medications (ASMs) as <5 and ≥6. Results: A total of 994 participants were included. During the 1-year study period, SSR was reached by 45.8%, 39.3%, and 22.6% of subjects with a baseline frequency of <5, 5–20, and >20 seizures per month (p <.001); the corresponding figures for the SSF were 23.4%, 9.8%, and 2.8% (p <.001). SSR was reached by 51.2% and 26.5% participants with a history of 1–5 and ≥6 ASMs (p <.001); the corresponding rates of SSF were 24.7% and 4.5% (p <.001). Treatment discontinuation due to lack of efficacy was more common in participants with >20 seizures compared to those with <5 seizures per month (25.8% vs. 9.3%, p <.001), and in participants with history of ≥6 prior ASMs compared to those with history of 1–5 ASMs (19.6% vs. 12.2%, p =.002). There were no differences in the rates of BRV withdrawal due to AEs and the rates of AEs across the groups of participants defined according to the number of seizures at baseline and the number of prior ASMs. Significance: The baseline seizure frequency and the number of previous ASMs were predictors of sustained seizure frequency reduction with adjunctive BRV in subjects with focal epilepsy
