    Electrospinning Auricular Shaped Scaffolds for Tissue Engineering

    Poly(É›)caprolactone scaffolds have been electrospun directly into an auricular shaped conductive mould. Bovine chondrocytes were harvested from articular cartilage and seeded onto 16 of the produced scaffolds, which received either an ethanol (group A) or a plasma treatment (group B) for sterilisation before seeding. The seeded scaffolds were cultured for 3 weeks in vitro and analysed with regard to total DNA and GAG content as well as the expression of AGG, COL1, COL2, MMP3 and MMP13. Rapid cell proliferation and GAG accumulation was observed until week 2. However, total DNA and GAG content decreased again in week 3. qPCR data shows a slight increase in the expression of anabolic genes and a slight decrease for the catabolic genes, with a significant difference between the groups A and B only for COL2 and MMP1

    Ellbogengelenkserkrankungen gewinnen bei jungen Hunden großer, schnellwüchsiger und bewegungsfreudiger Rassen seit Jahren zunehmend an Bedeutung. Als mögliche Ursachen der Ellbogendysplasie wurden - neben genetischer Veranlagung - Übergewicht und Überlastung der gelenkbildenden Knochen benannt. In der vorliegenden Untersuchung wurde der Einfluss verschiedener biomechanischer Parameter auf die Lastübertragung in gesunden und pathologischen Hundeellbogen mit einem zweidimensionalen Finite-Elemente-Modell analysiert. Pathologische Veränderungen der Ellbogenstruktur, wie veränderte Materialeigenschaften oder asynchrones Knochenwachstum, veränderten deutlich die Kontaktdrücke in den Artikulationen, die Knochendeformation und die Spannungen in den Knochen. Die gewonnenen Erkenntnisse unterstützen die langjährigen empirischen Beobachtungen und bieten eine Erklärung für die bis anhin kaum verstandenen klinischen Erscheinungsbilde

    Ischemic preconditioning enhances critical power during a 3 minute all-out cycling test

    This study tested the hypothesis that ischemic preconditioning (IPC) would increase critical power (CP) during a 3 minute all-out cycling test. Twelve males completed two 3 minute all-out cycling tests, in a crossover design, separated by 7 days. These tests were preceded by IPC (4 x 5 minute intervals at 220 mmHg bilateral leg occlusion) or SHAM treatment (4 x 5 minute intervals at 20 mmHg bilateral leg occlusion). CP was calculated as the mean power output during the final 30 s of the 3 minute test with W′ taken as the total work done above CP. Muscle oxygenation was measured throughout the exercise period. There was a 15.3 ± 0.3% decrease in muscle oxygenation (TSI; [Tissue saturation index]) during the IPC stimulus, relative to SHAM. CP was significantly increased (241 ± 65 W vs. 234 ± 67 W), whereas W′ (18.4 ± 3.8 vs 17.9 ± 3.7 kJ) and total work done (TWD) were not different (61.1 ± 12.7 vs 60.8 ± 12.7 kJ), between the IPC and SHAM trials. IPC enhanced CP during a 3 minute all-out cycling test without impacting W′ or TWD. The improved CP after IPC might contribute towards the effect of IPC on endurance performance

    Eigenvalue sensitivity minimisation for robust pole placement by the receptance method

    The problem of robust pole placement in active structural vibration control by the method of receptance is considered in this paper. Expressions are derived for the eigenvalue sensitivities to parametric perturbations, which are subsequently minimised to improve performance robustness of the control of a dynamical system. The described approach has application to a vibrating system where variations are present due to manufacturing and material tolerances, damages and environment variabilities. The closed-loop eigenvalue sensitivities are expressed as a linear function of the velocity and displacement feedback gains, allowing their minimisation with carefully calculated feedback gains. The proposed algorithm involves curve fitting perturbed frequency response functions, FRFs, using the rational fraction polynomial method and implementation of a polynomial fit to the individual estimated rational fraction coefficients. This allows the eigenvalue sensitivity to be obtained entirely from structural FRFs, which is consistent with the receptance method. This avoids the need to evaluate the M,C,K matrices which are typically obtained through finite element modelling, that produces modelling uncertainty. It is also demonstrated that the sensitivity minimisation technique can work in conjunction with the pole placement and partial pole placement technique using the receptance method.     Absence of microemboli on transcranial Doppler identifies low-risk patients with asymptomatic carotid stenosis who do not warrant endarterectomy or stenting

    Background and Purpose - Carotid endarterectomy clearly benefits patients with symptomatic severe stenosis (SCS), but the risk of stroke is so low for asymptomatic patients (ACS) that the number needed to treat is very high. We studied transcranial Doppler (TCD) embolus detection as a method for identifying patients at higher risk who would have a lower number needed to treat. Methods - Patients with carotid stenosis of ≥60% by Doppler ultrasound who had never been symptomatic (81%) or had been asymptomatic for at least 18 months (19%) were studied with TCD embolus detection for up to 1 hour on 2 occasions a week apart; patients were followed for 2 years. Results - 319 patients were studied, age (standard deviation) 69.68 (9.12) years; 32 (10%) had microemboli at baseline (TCD+). Events were more likely to occur in the first year. Patients with microemboli were much more likely to have microemboli 1 year later (34.4 versus 1.4%; P\u3c0.0001) and were more likely to have a stroke during the first year of follow-up (15.6%, 95% CI, 4.1 to 79; versus 1%, 95% CI, 1.01 to 1.36; P\u3c0.0001). Conclusions - Our findings indicate that TCD- ACS will not benefit from endarterectomy or stenting unless it can be done with a risk \u3c1%; TCD+ may benefit as much as SCS if their surgical risk is not higher. These findings suggest that ACS should be managed medically with delay of surgery or stenting until the occurrence of symptoms or emboli. © 2005 American Heart Association, Inc

However, at high concentrations it is toxic to the parasite. A heat-labile component is apparently responsible for the stage-specific toxicity to parasites within infected erythrocytes 12–42 h after invasion, i.e. during trophozoite maturation. The effects of HDL on parasite metabolism (as determined by nucleic acid synthesis) are evident at about 30 h after invasion. Parasites treated with HDL show gross abnormalities by light and electron microscopy.Professor Hajduk was supported by NIH.     The effect of pathological shoulder rhythm on muscle and joint forces after reverse shoulder arthroplasty, a numerical analysis.

    BACKGROUND Compromised abduction ability after reverse shoulder arthroplasty is primarily linked to limited glenohumeral range of motion while scapulothoracic mobility can typically be maintained. Glenohumeral joint forces strongly depend on the resulting scapulohumeral rhythm, however, an association between the acting muscle and joint forces and the subject-specific scapulohumeral rhythm after reverse shoulder arthroplasty has not been established. METHODS Eleven reverse shoulder arthroplasty patients were divided into groups of poor and excellent abduction ability. Subject-specific models were developed and scaled for each patient using existing motion capture data in AnyBody™. Shoulder muscle and joint forces were obtained using inverse dynamics calculations during shoulder abduction to 100° in the scapula plane. The scapulohumeral rhythm, the resting abduction angle and internal body forces between the outcome groups were compared using a Mann Whitney U test. FINDINGS The mean glenohumeral and scapulothoracic contribution to overall shoulder abduction for the excellent group was on average 9.7% higher and 21.4% lower, respectively, compared to the mean of the poor group. For shoulder abduction angles between 30° and 60°, the excellent group demonstrated on average 25% higher muscle forces in the anterior deltoid which was significantly higher compared to the poor outcome patients. Scapulothoracic muscle activity did not differ significantly between the two functional groups. INTERPRETATION Accordingly, rehabilitation strategies focusing on strengthening the anterior part of the deltoid in particular may improve clinical outcomes

    The development and validation of a human screening model of tobacco abstinence

    Introduction Given the low efficacy of smoking cessation methods, an experimental medicine model indicating smoking abstinence would be of great benefit to the development of new treatments. Hence the sensitivity of cognitive tasks and ambulatory craving measures to smoking abstinence were investigated. Methods Cognitive tasks and ambulatory ratings of craving were assessed for sensitivity to acute abstinence (experiment 1), and nicotine replacement therapy administration (NRT) (experiment 2). Results In experiment 1 go/no-go performance was improved (Mean Difference [MD] -0.99, 95% CI: −1.90 to −0.08) and craving was lower (Regression Coefficient [RC] −33.39, 95% CI: -39.96 to -26.82) in satiated compared with abstinent smokers. There was no clear evidence that N-back (MD 0.64, 95% CI: −0.42 to 2.51), delay discounting (MD 0.01, 95% CI: 0.001 to 0.005) or dot probe performance (MD 0.61, 95% CI: −0.87 to 1.54) were sensitive to acute abstinence. In experiment 2 go/no-go performance was improved (MD 1.12, 95% CI: 0.16–2.08) and craving was lower (RC −18.59, 95% CI: −24.63 to −12.55) smokers abstinent overnight receiving NRT compared with placebo. There was no clear evidence that N-back (MD −0.25, 95% CI: −1.45 to 0.94), delay discounting (MD 0.01, 95% CI: -0.002 to 0.004) or dot probe performance (MD −0.49, 95% CI: −1.61 to −0.64) were sensitive to NRT. Conclusions Findings from two experiments converge to suggest that abstinence in smokers reliably increases ambulatory craving assessments and, to a lesser extent, decreases go/no-go task performance. These findings can be utilized in the development of an experimental medicine model to test novel treatments for smoking cessation
