The UDP-glucosyltransferase multigene family in Bombyx mori
<p>Abstract</p> <p>Background</p> <p>Glucosidation plays a major role in the inactivation and excretion of a great variety of both endogenous and exogenous compounds. A class of UDP-glycosyltransferases (UGTs) is involved in this process. Insect UGTs play important roles in several processes, including detoxication of substrates such as plant allelochemicals, cuticle formation, pigmentation, and olfaction. Identification and characterization of <it>Bombyx mori </it>UGT genes could provide valuable basic information for this important family and explain the detoxication mechanism and other processes in insects.</p> <p>Results</p> <p>Taking advantage of the newly assembled genome sequence, we performed a genome-wide analysis of the candidate UGT family in the silkworm, <it>B. mori</it>. Based on UGT signature and their similarity to UGT homologs from other organisms, we identified 42 putative silkworm UGT genes. Most of them are clustered on the silkworm chromosomes, with two major clusters on chromosomes 7 and 28, respectively. The phylogenetic analysis of these identified 42 UGT protein sequences revealed five major groups. A comparison of the silkworm UGTs with homologs from other sequenced insect genomes indicated that some UGTs are silkworm-specific genes. The expression patterns of these candidate genes were investigated with known expressed sequence tags (ESTs), microarray data, and RT-PCR method. In total, 36 genes were expressed in tissues examined and showed different patterns of expression profile, indicating that these UGT genes might have different functions.</p> <p>Conclusion</p> <p><it>B. mori </it>possesses a largest insect UGT gene family characterized to date, including 42 genes. Phylogenetic analysis, genomic organization and expression profiles provide an overview for the silkworm UGTs and facilitate their functional studies in future.</p
Preparation and biological application of antibodies against leucoanthocyanidin reductase and anthocyanidin reductase from grape berry
Proanthocyanidins (PAs) endow wine with the flavor of bitterness and astringency. Both leucoanthocyanidin reductase (LAR) and anthocyanidin reductase (ANR) are two key enzymes of PA biosynthesis in grape berries, but the previous studies on these enzymes only focused on the transcriptional expression of these genes. Here, the full-length cDNAs of VvLAR1, VvLAR2 and VvANR, respectively, were cloned from wine grape berries and were then introduced into the pGEX-4T-1 expression vectors, which were highly expressed in Escherichia coli DH5α cells with the induction of the isopropyl-β-D-thiogalactoside (IPTG). The purified fusion proteins were used as the antigens to immunize rabbits, separately. The obtained antiserums were further purified to obtain the immunoglobulin G (Ig G) fractions, which were demonstrated to be capable of specifically immuno-recognizing the VvLAR1, VvLAR2 and VvANR from the crude protein extracts from grape berries with weight masses of approximately 43 kD. The analyses of translational expression of these enzyme genes during berry development and immunohistochemical localization of these proteins, by using the obtained antibodies, showed that a high amount of VvLAR1, VvLAR2 or VvANR was present at the pre-veraison stage and these enzyme proteins were all localized on the outer layer of the berry skin and the vascular bundle, as well as in the inner layer of the seed coat. This work provides an important basis for further studies on PA biosynthesis in grape berries.
Shenfu injection: a review of pharmacological effects on cardiovascular diseases
Shenfu injection (SFI), composed of ginseng and aconite, is a Chinese patent developed from the classic traditional prescription Shenfu Decoction created more than 700 years ago. SFI has been widely used in China for over 30 years for treating cardiovascular diseases. The main components in it include ginsenosides and aconitum alkaloids. In recent years, the role of SFI in the treatment of cardiovascular diseases has attracted much attention. The pharmacological effects and therapeutic applications of SFI in cardiovascular diseases are summarized here, highlighting pharmacological features and potential mechanisms developments, confirming that SFI can play a role in multiple ways and is a promising drug for treating cardiovascular diseases
Superconductivity at 22.3 K in SrFe2-xIrxAs2
By substituting the Fe with the 5d-transition metal Ir in SrFe2As2, we have
successfully synthesized the superconductor SrFe2-xIrxAs2 with Tc = 22.3 K at x
= 0.5. X-ray diffraction indicates that the material has formed the
ThCr2Si2-type structure with a space group I4/mmm. The temperature dependence
of resistivity and dc magnetization both reveal sharp superconducting
transitions at around 22 K. An estimate on the diamagnetization signal reveals
a high Meissner shielding volume. Interestingly, the normal state resistivity
exhibits a roughly linear behavior up to 300 K. The superconducting transitions
at different magnetic fields were also measured yielding a slope of -dHc2/dT =
3.8 T/K near Tc. Using the Werthamer-Helfand-Hohenberg (WHH) formula, the upper
critical field at zero K is found to be about 58 T. Counting the possible
number of electrons doped into the system in SrFe2-xIrxAs2, we argue that the
superconductivity in the Ir-doped system is different from the Co-doped case,
which should add more ingredients to the underlying physics of the iron

Reproductive traits and mandibular gland pheromone of anarchistic honey bee workers Apis mellifera occurring in China
International audienceAbstractIn honey bee colonies, workers, in particular of “anarchistic” lineages, can activate their ovaries and lay eggs, even in the presence of the queen. We identified three queenright colonies showing typical signs of worker reproduction. To characterize this new lineage, we extracted the mandibular gland and analyzed it using gas chromatography. The total amounts of the five main components of the mandibular gland, namely methyl p-hydroxyben-zoate (HOB), 9-oxo-2(E)-decenoic acid (9-ODA), (S)-9-hydroxy-(E)-2-decenoic acid (9-HDA), 10-HDA, and 10-hydroxyde-canoic acid (10-HDAA) were significantly higher in the mandibular gland profiles of workers with activated ovaries (AWs, 8.88 ± 1.71 μg) compared to workers with inactivated ovaries (IAWs, 4.00 ± 2.09 μg). Furthermore, the chemical profiles of IAWs were dominated by the “worker substances” 10-HDA (34.64 ± 8.19 %) and its precursor 10-HDAA (22.88 ± 4.95 %), while the chemical profiles in AWs were dominated by the precursor of the queen substance 9-HDA (40.04 ± 7.55 %). The ratios of two precursor substances 10-HDAA/9-HDA of IAWs were more worker like (>1.0) whereas AWs were more queen like (≤1.0). These results suggest that the mandibular pheromones of anarchistic workers resemble a more queen-like reproductive active profile and that these workers may represent a reversion to a more basal reproductive phenotype
Guidezilla extension catheter for percutaneous interventional therapy of complex lesions via a transradial approach: Case series from a single-center experience
Background: Guide catheter extension systems have become one of the most powerful tools for addressing complex lesions during percutaneous coronary intervention (PCI), but data on a new-generation rapid exchange extension catheter — the Guidezilla catheter — are limited. Summarized herein reports on experience using the Guidezilla catheter for complex coronary lesions via a transradial approach at the documented institution an evaluation of its safety and efficacy.
Methods: A total of 25 patients (19 males and 6 females) who underwent PCI via the radial approach with the Guidezilla catheter for adequate back-up support and to facilitate equipment delivery were enrolled. The clinical, angiographic and procedural data of all 26 procedures in 25 patients (1 patient underwent two PCI procedures on different lesions) were collected to evaluate the safety and efficacy of this novel equipment.
Results: The mean age of the enrolled patients was 67.7 ± 8.41 years old. The mean depth of intubation was 27.90 ± 12.23 mm. Stent implantation was successful in 23 out of 26 procedures (88.5%) and failed in 3 cases: 1 case of tortuosity and severe angulation in a chronic total occlusion lesion; 1 case of an existing type B dissection (NHLBI classification system for coronary artery dissection types); and 1 case in which a stent was stripped off its balloon. None of the patients experienced coronary dissection, perforation, air embolism, pressure dampening or other major complications during the procedure.
Conclusions: The Guidezilla extension catheter is an effective and safe tool that provides improved back-up support and increases the success rate of PCI for complex coronary lesion by radial access
A review of obstructive sleep apnea and lung cancer: epidemiology, pathogenesis, and therapeutic options
Despite undeniable advances in modern medicine, lung cancer still has high morbidity and mortality rates. Lung cancer is preventable and treatable, and it is important to identify new risk factors for lung cancer, especially those that can be treated or reversed. Obstructive sleep apnea (OSA) is a very common sleep-breathing disorder that is grossly underestimated in clinical practice. It can cause, exacerbate, and worsen adverse outcomes, including death and various diseases, but its relationship with lung cancer is unclear. A possible causal relationship between OSA and the onset and progression of lung cancer has been established biologically. The pathophysiological processes associated with OSA, such as sleep fragmentation, intermittent hypoxia, and increased sympathetic nervous excitation, may affect normal neuroendocrine regulation, impair immune function (especially innate and cellular immunity), and ultimately contribute to the occurrence of lung cancer, accelerate progression, and induce treatment resistance. OSA may be a contributor to but a preventable cause of the progression of lung cancer. However, whether this effect exists independently of other risk factors is unclear. Therefore, by reviewing the literature on the epidemiology, pathogenesis, and treatment of lung cancer and OSA, we hope to understand the relationships between the two and promote the interdisciplinary exchange of ideas between basic medicine, clinical medicine, respiratory medicine, sleep medicine, and oncology
Universal Information Extraction with Meta-Pretrained Self-Retrieval
Universal Information Extraction~(Universal IE) aims to solve different
extraction tasks in a uniform text-to-structure generation manner. Such a
generation procedure tends to struggle when there exist complex information
structures to be extracted. Retrieving knowledge from external knowledge bases
may help models to overcome this problem but it is impossible to construct a
knowledge base suitable for various IE tasks. Inspired by the fact that large
amount of knowledge are stored in the pretrained language models~(PLM) and can
be retrieved explicitly, in this paper, we propose MetaRetriever to retrieve
task-specific knowledge from PLMs to enhance universal IE. As different IE
tasks need different knowledge, we further propose a Meta-Pretraining Algorithm
which allows MetaRetriever to quicktly achieve maximum task-specific retrieval
performance when fine-tuning on downstream IE tasks. Experimental results show
that MetaRetriever achieves the new state-of-the-art on 4 IE tasks, 12 datasets
under fully-supervised, low-resource and few-shot scenarios.Comment: Accepted to ACL 202
Nitrogen Self-Doped Activated Carbons Derived from Bamboo Shoots as Adsorbent for Methylene Blue Adsorption
Bamboo shoots, a promising renewable biomass, mainly consist of carbohydrates and other nitrogen-related compounds, such as proteins, amino acids and nucleotides. In this work, nitrogen self-doped activated carbons derived from bamboo shoots were prepared via a simultaneous carbonization and activation process. The adsorption properties of the prepared samples were evaluated by removing methylene blue from waste water. The factors that affect the adsorption process were examined, including initial concentration, contact time and pH of methylene blue solution. The resulting that BSNC-800-4 performed better in methylene blue removal from waste water, due to its high specific surface area (2270.9 m2 g−1), proper pore size (2.19 nm) and relatively high nitrogen content (1.06%). Its equilibrium data were well fitted to Langmuir isotherm model with a maximum monolayer adsorption capacity of 458 mg g−1 and a removal efficiency of 91.7% at methylene blue concentration of 500 mg L−1. The pseudo-second-order kinetic model could be used to accurately estimate the carbon material’s (BSNC-800-4) adsorption process. The adsorption mechanism between methylene blue solution and BSNC-800-4 was controlled by film diffusion. This study provides an alternative way to develop nitrogen self-doped activated carbons to better meet the needs of the adsorption applications
