    Screening hub genes in coronary artery disease based on integrated analysis

    Background: Coronary artery disease (CAD) is the leading cause of mortality worldwide. Identifying key pathogenic genes benefits the understanding molecular mechanism of CAD. Methods: In this study, 5 microarray data sets from the blood sample of 312 CADs and 277 healthy controls were downloaded. Limma and metaMA packages were used to identify differentially expressed genes. The functional enrichment analysis of differentially expressed genes was further performed by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes. Additionally, protein–protein interac­tion and transcript factors-target networks were performed based on top 10 up- and down-regulated differentially expressed genes to further study the biological function. Last, real-time quantitative poly­merase chain reaction (RT-qPCR) was used to validate the integrated analysis result. Results: A total of 528 differentially expressed genes were obtained. All differentially expressed genes were significantly involved in signal transduction and the MAPK signaling pathway. Among MAPK signaling pathway, IL1R2, ARRB2 and PRKX were associated with CAD. Furthermore, there were 4 common differentially expressed genes including PLAUR, HSPH1, ZMYND11 and S100A8 in the protein–protein interaction and transcript factors-target networks, which played crucial roles in the development of CAD. In quantitative RT-qPCR, the expression of PRKX, HSPH1 and ZMYND11 was down-regulated and consistent with the integrated analysis. Conclusions: Identified 7 differentially expressed genes (IL1R2, ARRB2, PRKX, PLAUR, HSPH1, ZMYND11 and S100A8) may play crucial roles in the development of CAD

    Multi-level Distillation of Semantic Knowledge for Pre-training Multilingual Language Model

    Pre-trained multilingual language models play an important role in cross-lingual natural language understanding tasks. However, existing methods did not focus on learning the semantic structure of representation, and thus could not optimize their performance. In this paper, we propose Multi-level Multilingual Knowledge Distillation (MMKD), a novel method for improving multilingual language models. Specifically, we employ a teacher-student framework to adopt rich semantic representation knowledge in English BERT. We propose token-, word-, sentence-, and structure-level alignment objectives to encourage multiple levels of consistency between source-target pairs and correlation similarity between teacher and student models. We conduct experiments on cross-lingual evaluation benchmarks including XNLI, PAWS-X, and XQuAD. Experimental results show that MMKD outperforms other baseline models of similar size on XNLI and XQuAD and obtains comparable performance on PAWS-X. Especially, MMKD obtains significant performance gains on low-resource languages.Comment: accepted at EMNLP 202

    抑郁发作患者抑郁严重程度与认知应对策略的关系

    Objective: To explore the relationship between the depressive mood and cognitive emotion regulation strategies.  Methods: A total of 143 major depression patients were assessed with a self-designed questionnaire, Beck Depression Inventory (BDI), the Chinese version of Cognitive Emotion Regulation Questionnaire (CERQ-C) and the twenty-item Toronto Scale (TAS-20), and 95 healthy people were involved as controls. Regression analysis was carried out for the influcing factors to depressive mood. Results: Compared with controls, Patients with Depressive Episode showed significantly higher total scores of inadaptability strategy,TAS and BDI(P<0.01). There were lots of factors, such as domestic stress and maladaptive strategy showed significant correlation with total score of BDI in major depression (P<0.05 or P<0.01). The factors of domestic stress and maladaptive strategy et al entered the regression equation for BDI by turns. Conclusions: Patients with depression suffered from maladaptive strategy. Cognitive coping strategies and stressl were the important influence factors of depression emotion in patients with depression.目的 探讨抑郁发作患者抑郁情绪与认知应对策略的关系。方法 采用自编一般情况问卷、贝克抑郁问卷(BDI)、认知情绪调节问卷中文版(CERQ-C)、多伦多述情量表(TAS-20) 中文版对143例抑郁发作患者及95例健康对照者进行评定,采用多元回归方法对抑郁的影响因素进行分析。结果 抑郁发作患者CERQ-C不适应性策略分、TAS总分及抑郁总分高于对照组(P<0.01)。抑郁发作患者抑郁总分与家庭应激、CERQ-C不适应性策略等因素密切相关(P<0.05或P<0.01)。家庭应激与CERQ-C不适应性策略等依次进入BDI总分的回归方程。结论 抑郁发作患者较多地使用了不适应性策略。认知应对策略、应激等为抑郁发作患者抑郁情绪的重要影响因素

    Fluorescent protein tagged hepatitis B virus capsid protein with long glycine-serine linker that supports nucleocapsid formation

    Fusion core proteins of Hepatitis B virus can be used to study core protein functions or capsid trafficking. A problem in constructing fusion core proteins is functional impairment of the individual domains in these fusion proteins, might due to structural interference. We reported a method to construct fusion proteins of Hepatitis B virus core protein (HBc) in which the functions of fused domains were partially kept. This method follows two principles: (1) fuse heterogeneous proteins at the N terminus of HBc; (2) use long Glycine-serine linkers between the two domains. Using EGFP and RFP as examples, we showed that long flexible G4S linkers can effectively separate the two domains in function. Among these fusion proteins constructed, GFP-G4S186-HBc and RFP-G4S47-HBc showed the best efficiency in rescuing the replication of an HBV replicon deficient in the core protein expression, though both of the two fusion proteins failed to support the formation of the relaxed circular DNA. These fluorescent protein-tagged HBcs might help study related to HBc or capsids tracking in cells
