Lactate and Pyruvate Activate Autophagy and Mitophagy that Protect Cells in Toxic Model of Parkinson's Disease
Intracellular quality control regulated by autophagy process is important for maintenance of cellular homeostasis. Deregulation of autophagy and more specifically mitophagy leads to accumulation of the misfolded proteins and damaged mitochondria that in turn leads to the cell loss. Alteration of autophagy and mitophagy has shown to be involved in the number of disorders including neurodegenerative diseases. Autophagy and mitophagy could be activated by short-time acidification of the cytosol; however, most of the compounds which can induce it are toxic. Here, we tested several organic compounds which are involved in cellular metabolism on their ability to change intracellular pH and induce mitophagy/autophagy. We have found that lactate and pyruvate are able to reduce intracellular pH in non-toxic concentrations. Short-term (2 h) and long-term (24 h) incubation of the cells with lactate and pyruvateinduced mitophagy and autophagy. Incubation of the SH-SY5Y cells or primary neurons and astrocytes with lactate or pyruvate also activated mitophagy and autophagy after MPP + treatment that led to recovery of mitochondrial function and protection of these cells against apoptotic and necrotic death. Thus, pyruvate- or lactate-induced acidification of cytosol activates cell protective mitophagy and autophagy
Semantic transformations of maritime historicisms in modern English
The article is dedicated to the study of English maritime historicisms in the modern English language. The author conducts research of English maritime historicisms aiming to prove that, firstly, they are actively used in modern English, secondly they changed their meanings and thirdly, their functions are not limited to the nominative one. Moreover, the author singles out numerous transformations that affect the meaning of maritime historicisms. The methods used by the author include: descriptive, comparative, systematizing methods and method of contextual analysis. The choice of maritime vocabulary as the material for analysis accounts for by two factors: 1) maritime affairs have always been closely associated with human activities of the British, hence, the English maritime vocabulary includes a large number of historicisms that were used to denote objects and events not existing now; 2) a considerable amount of written records where historicisms are used are still available, and on their basis maritime dictionaries which include historical terms have been compiled. The factual materials comprise numerous marine dictionaries, news columns, literary works, articles and encyclopedias. Conclusions reached by the author are as follows: English maritime historicisms form part of active vocabulary in the English language; four ways how they can change their meaning have been singled out and; apart from nominative function they perform expressive and evaluative functions as well. Theoretical and practical significance of the article consists not only in contributing to the knowledge about English maritime historicisms but also in tracing the association between Maritime History and its reflection in the English language. Moreover, the article can provide the basis for future research in discourse studies (e.g. studying the functional characteristics of historicisms in professional and non-professional communication) and terminological studies, translation of historicisms etc

Generation of recombinant adenoviral vectors encoding neural cell adhesion molecules ncam1, ncam2 and l1cam
To succed in gene therapy it is necessary to observe few basic conditions, such as targeted delivery of the therapeutic gene into the target organ and its effective expression. Nowadays targeted delivery of therapeutic genes is one of the critical problems of gene therapy. Delivery of recombinant genes using cell carriers (vectors), expressing tissue-specific cell adhesion molecules, allows us to move toward solving this problem. Using Gateway cloning technology (Invitrogen) we have created recombinant expression constructs pAd-NCAM1, pAd-NCAM2 and pAd-L1CAM, encoding neural cell adhesion molecules. Expression of recombinant proteins has been confirmed by immunofluorescent analysis. Based on these genetic constructs recombinant adenoviruses (serotype 5) were generated and titered. Obtained viral vectors encoding neural cell adhesion molecules may be subsequently used to modify cells carrying additional therapeutic genes to increase the efficiency of gene-cell therapeutics delivery to target organ. © Human stem cells institute, 2013
