    Cytochrome oxidase subunit VI of Trypanosoma brucei is imported without a cleaved presequence and is developmentally regulated at both RNA and protein levels

    Mitochondrial respiration in the African trypanosome undergoes dramatic developmental stage regulation. This requires co-ordinated control of components encoded by both the nuclear genome and the kinetoplast, the unusual mitochondrial genome of these parasites. As a model for understanding the co-ordination of these genomes, we have examined the regulation and mitochondrial import of a nuclear-encoded component of the cytochrome oxidase complex, cytochrome oxidase subunit VI (COXVI). By generating transgenic trypanosomes expressing intact or mutant forms of this protein, we demonstrate that COXVI is not imported using a conventional cleaved presequence and show that sequences at the N-terminus of the protein are necessary for correct mitochondrial sorting. Analyses of endogenous and transgenic COXVI mRNA and protein expression in parasites undergoing developmental stage differentiation demonstrates a temporal order of control involving regulation in the abundance of, first, mRNA and then protein. This represents the first dissection of the regulation and import of a nuclear-encoded protein into the cytochrome oxidase complex in these organisms, which were among the earliest eukaryotes to possess a mitochondrion

    'An outsider in our midst': narratives of Neil Lennon, soccer and ethno-religious igotry in the Scottish press

    This essay offers a critique of media narratives concerning soccer and those of 'difference' in contemporary Scotland, in particular those who have Irishness as their different identity. It examines certain newspaper narratives concerning Neil Lennon of Celtic FC during autumn 2005. During this period Lennon was characterized as a soccer villain. The commentaries drew on existing perceptions concerning his personality and style of play. More importantly Lennon's national identity (Irish) and his religious background (Catholic) were integrated into the narratives, marking him as an outsider in Scotland. These narratives resonate with public and private discourses of 'otherness' concerning the Irish Catholic diaspora community in Scotland. These broader discourses are manifest as ethno-religious prejudices directed against this community. The discourses of 'outsider' and 'otherness' that surround Lennon, Celtic FC and the Irish Catholic community expose the myth of Scotland's collective self-image as an egalitarian and inclusive society

    Mitochondrial Development during Life Cycle Differentiation of African Trypanosomes: Evidence for a Kinetoplast-dependent Differentiation Control Point

    Life cycle differentiation of African trypanosomes entails developmental regulation of mitochondrial activity. This requires regulation of the nuclear genome and the kinetoplast, the trypanosome's unusual mitochondrial genome. To investigate the potential cross talk between the nuclear and mitochondrial genome during the events of differentiation, we have 1) disrupted expression of a nuclear-encoded component of the cytochrome oxidase (COX) complex; and 2) generated dyskinetoplastid cells, which lack a mitochondrial genome. Using RNA interference (RNAi) and by disrupting the nuclear COX VI gene, we demonstrate independent regulation of COX component mRNAs encoded in the nucleus and kinetoplast. However, two independent approaches (acriflavine treatment and RNA interference ablation of mitochondrial topoisomerase II) failed to establish clonal lines of dyskinetoplastid bloodstream forms. Nevertheless, dyskinetoplastid forms generated in vivo could undergo two life cycle differentiation events: transition from bloodstream slender to stumpy forms and the initiation of transformation to procyclic forms. However, they subsequently arrested at a specific point in this developmental program before cell cycle reentry. These results provide strong evidence for a requirement for kinetoplast DNA in the bloodstream and for a kinetoplast-dependent control point during differentiation to procyclic forms

    RURAL-URBAN DIFFERENCES IN THE UTILIZATION AND AMELIORATIVE EFFECTS OF WELFARE PROGRAMS

    At a time when welfare policy analysts are preoccupied with the problem of welfare dependency, this paper refocuses attention on the ability of welfare (AFDC and general assistance) to reduce poverty among families, by highlighting and explaining rural-urban differences in this ameliorative effect. Descriptive and multivariate methods are used to analyze data from the March 1987 Current Population Survey. Results show that despite comparatively high poverty rates in nonmetro areas, the nonmetro poor were much less likely than their urban counterparts t o receive welfare. Moreover, nonmetro welfare recipients received considerably less welfare income, on average, than metro recipients. Accordingly, the ameliorative effect of welfare was lowest in nonmetro areas and highest in central cities. Logistic regression analysis revealed that nonmetro poor families were less likely to receive welfare than those in metro areas because they were more likely to be working, older, childless, and headed by a married couple.
