Hadron beam test of a scintillating fibre tracker system for elastic scattering and luminosity measurement in ATLAS
A scintillating fibre tracker is proposed to measure elastic proton
scattering at very small angles in the ATLAS experiment at CERN. The tracker
will be located in so-called Roman Pot units at a distance of 240 m on each
side of the ATLAS interaction point. An initial validation of the design
choices was achieved in a beam test at DESY in a relatively low energy electron
beam and using slow off-the-shelf electronics. Here we report on the results
from a second beam test experiment carried out at CERN, where new detector
prototypes were tested in a high energy hadron beam, using the first version of
the custom designed front-end electronics. The results show an adequate
tracking performance under conditions which are similar to the situation at the
LHC. In addition, the alignment method using so-called overlap detectors was
studied and shown to have the expected precision.Comment: 12 pages, 8 figures. Submitted to Journal of Instrumentation (JINST
Health-related quality of life associated with trifluridine/tipiracil in heavily pretreated metastatic gastric cancer: results from TAGS
Background
In TAGS, an international, double-blind, phase 3 trial, trifluridine/tipiracil significantly improved overall survival and progression-free survival compared with placebo in heavily pretreated metastatic gastric cancer patients. This paper reports pre-specified quality of life (QoL) outcomes for TAGS.
Methods
Patients were randomized 2:1 to trifluridine/tipiracil (35 mg/m2 twice daily on days 1â5 and 8â12 of each 28-day cycle) plus best supportive care (BSC) or placebo plus BSC. QoL was evaluated at baseline and at each treatment cycle, using the EORTC QLQ-C30 and EORTC QLQ-STO22 questionnaires; results were considered valid for analysis only ifââ„â10% of patients completed the questionnaires. Key QoL outcomes were mean changes from baseline and time to deterioration in QoL. A post hoc analysis assessed the association between QoL and time to deterioration of Eastern Cooperative Oncology Group performance score (ECOG PS) toââ„â2.
Results
Of 507 randomized patients, 496 had baseline QoL data available. The analysis cut-off was 6 cycles for trifluridine/tipiracil and 3 cycles for placebo. In both treatment groups, there were no clinically significant deteriorations in the mean QLQ-C30 Global Health Status (GHS) score, or in most subscale scores. In a sensitivity analysis including death and disease progression as events, there was a trend towards trifluridine/tipiracil reducing the risk of deterioration of QoL scores compared with placebo. Deterioration in the GHS score was associated with deterioration in ECOG PS.
Conclusion
QoL was maintained in TAGS, and there was a trend towards trifluridine/tipiracil reducing the risk of QoL deterioration compared with placebo
Permanent Neonatal Diabetes Caused by Creation of an Ectopic Splice Site within the INS Gene
PublishedCase ReportsJournal ArticleResearch Support, Non-U.S. Gov'tBACKGROUND: The aim of this study was to characterize the genetic etiology in a patient who presented with permanent neonatal diabetes at 2 months of age. METHODOLOGY/PRINCIPAL FINDINGS: Regulatory elements and coding exons 2 and 3 of the INS gene were amplified and sequenced from genomic and complementary DNA samples. A novel heterozygous INS mutation within the terminal intron of the gene was identified in the proband and her affected father. This mutation introduces an ectopic splice site leading to the insertion of 29 nucleotides from the intronic sequence into the mature mRNA, which results in a longer and abnormal transcript. CONCLUSIONS/SIGNIFICANCE: This study highlights the importance of routinely sequencing the exon-intron boundaries and the need to carry out additional studies to confirm the pathogenicity of any identified intronic genetic variants.Centro de Investigación Biomédica en Red de Diabetes y Enfermedades Metabólicas (CIBERDEM)Instituto de Salud Carlos III of the Spanish Ministry of HealthFIS-programsWellcome Trus
Functional Annotation and Identification of Candidate Disease Genes by Computational Analysis of Normal Tissue Gene Expression Data
Background: High-throughput gene expression data can predict gene function through the ââguilt by associationâ â principle: coexpressed genes are likely to be functionally associated. Methodology/Principal Findings: We analyzed publicly available expression data on normal human tissues. The analysis is based on the integration of data obtained with two experimental platforms (microarrays and SAGE) and of various measures of dissimilarity between expression profiles. The building blocks of the procedure are the Ranked Coexpression Groups (RCG), small sets of tightly coexpressed genes which are analyzed in terms of functional annotation. Functionally characterized RCGs are selected by means of the majority rule and used to predict new functional annotations. Functionally characterized RCGs are enriched in groups of genes associated to similar phenotypes. We exploit this fact to find new candidate disease genes for many OMIM phenotypes of unknown molecular origin. Conclusions/Significance: We predict new functional annotations for many human genes, showing that the integration of different data sets and coexpression measures significantly improves the scope of the results. Combining gene expression data, functional annotation and known phenotype-gene associations we provide candidate genes for several geneti
Splenic differentiation and emergence of CCR5+CXCL9+CXCL10+ monocyte-derived dendritic cells in the brain during cerebral malaria
Dendritic cells have an important role in immune surveillance. After being exposed to microbial components, they migrate to secondary lymphoid organs and activate T lymphocytes. Here we show that during mouse malaria, splenic inflammatory monocytes differentiate into monocyte-derived dendritic cells (MO-DCs), which are CD11b+F4/80+CD11c+MHCIIhighDC-SIGNhighLy6c+ and express high levels of CCR5, CXCL9 and CXCL10 (CCR5+CXCL9/10+ MO-DCs). We propose that malaria-induced splenic MO-DCs take a reverse migratory route. After differentiation in the spleen, CCR5+CXCL9/10+ MO-DCs traffic to the brain in a CCR2-independent, CCR5-dependent manner, where they amplify the influx of CD8+ T lymphocytes, leading to a lethal neuropathological syndrome
Trees improve soil carbon stocks in integrated crop-livestock-forestry (ICLF) systems.
Soil organic carbon (SOC, determined with dichromate oxidation) accumulation was evaluated at three ICLF sites of different ages (2, 4 and 11 years) and soil textures (469, 198, 632 g kg-1 clay) in southern GoiĂĄs state of Brazil. SOC stocks were accessed at 0-30, 30-100 and 0-100 cm soil layers under the tree-rows and between tree-rows. SOC stocks were calculated based on equivalent soil mass, using pasture, in which the ICLFs were implemented, as reference. Our objective was to investigate the potential of trees in accumulating SOC in ICLFs. Dunnett?s test was applied to check the effect of trees on soil C stocks within each location using the linear mixed model. More SOC was measured under tree-rows at all sites and all soil layers, compared to the alley between the tree-rows, except for the youngest site (Morrinhos) at 0-30 cm. Trees, therefore, are important to maximize SOC accumulation in ICLFs. Further studies are, however, necessary to expand the database to support more robust analysis and to conclude on SOC sequestration (immobilization)
Development of a detector (ALFA) to measure the absolute LHC luminosity at ATLAS
The ATLAS collaboration plans to determine the absolute luminosity of the CERN LHC at Interaction Point 1 by measuring the trajectory of protons elastically scattered at very small angles (). A scintillating fibre tracker system called ALFA (Absolute Luminosity For ATLAS) is proposed for this measurement. Detector modules will be placed above and below the LHC beam axis in roman pot units at a distance of 240 m on each side of the ATLAS interaction point. They allow the detectors to approach the beam axis to millimeter distance. Overlap detectors also based on the scintillating fibre technology, will measure the precise relative position of the two detector modules. Results obtained during beam tests at DESY and at CERN validate the detectors design and demonstrate the achievable resolution. We also report about radiation hardness studies of the scintillating fibres to estimate the lifetime of the ALFA system at different operating conditions of the LHC
Driven Assembly of Lignin into Microcapsules for Storage and Delivery of Hydrophobic Molecules
Oil-filled microcapsules of kraft lignin were synthe-
sized by first creating an oil in water emulsion followed by a high-
intensity, ultrasound-assisted cross-linking of lignin at the water/oil
interface. The rationale behind our approach is based on promoting
documented lignin hydrophobic interactions within the oil phase,
followed by locking the resulting spherical microsystems by covalent
cross-linking using a high intensity ultrasound treatment. As further
evidence in support of our rationale, confocal and optical
microscopies demonstrated the uniformly spherical morphology of
the created lignin microparticles. The detailed elucidation of the
cross-linking processes was carried out using gel permeation
chromatography (GPC) and quantitative 31P NMR analyses. The
ability of lignin microcapsules to incorporate and release Coumarin-6
was evaluated in detail. In vitro studies and confocal laser scanning microscopy analysis were carried out to assess the internalization of capsules into Chinese hamster ovary (CHO) cells. This part of our work demonstrated that the lignin microcapsules are not cytotoxic and readily incorporated in the CHO cells
