    Phylogeny and taxonomy of the Ophiostoma piceae complex and the Dutch elm disease fungi

    The Ophiostoma piceae complex forms a monophyletic group of insect-dispersed pyrenomycetes with synnemata (Pesotum) and micronematous (Sporothrix) synanamorphs. Other species of Ophios-toma outside of the O. piceae complex that form syn-nemata lack the Sporothrix state. The nine recognized species within the 0. piceae complex are delimited by synnema morphology, growth rate at 32 C, mating reactions and sequences of the internal transcribed spacer (ITS) region of the rDNA operon. Phyloge-netic analysis of the ITS region suggests two major clades in the complex, one that causes bluestain in primarily coniferous hosts and the other on primarily hardwood hosts. In the coniferous group are O. pi-ceae, O. canum, O. floccosum and the recently de-scribed O. setosum (anamorph Pesotum cupulatum sp. nov.). In the hardwood group are O. querci, O. caton-ianum, and the Dutch elm disease fungi: O. ulmi, O. novo-ulmi and O. himal-ulmi. Restriction fragment length polymorphisms of the ITS region are shown to be a convenient diagnostic tool for delimiting these species

    The Plume Impingement Contamination II Experiment: Motivation, Design, and Implementation Plan

    The International Space Station (ISS) will have a long service life during which it must be able to serve as a capable platform for a wide variety of scientific investigations. In order to provide this capability, the ISS has, at the system level, a design requirement of no more than 100 Angstroms of contaminant deposition per year from "non-quiescent" sources. Non-quiescent sources include the plumes resulting from the firing of reaction control system (ReS) engines on space vehicles visiting the ISS as well as the engines on the ISS itself. Unfortunately, good general plume contamination models do not yet exist. This is due both to the complexity of the problem, making the analytic approach difficult, and to the difficulty in obtaining empirical measurements of contaminant depositions. To address this lack of flight data, NASA Johnson Space Center is planning to fly an experiment, Plume Impingement Contamination-II, to measure the contamination deposition from the Shuttle Orbiter's primary RCS engines as a function angle from plume centerline. This represents the first direct on-orbit measurement of plume impingement contamination away from the nozzle centerline ever performed, and as such is extremely important in validating mathematical models which will be used to quantify the cumulative plume impingement contamination to the ISS over its lifetime. The paper will elaborate further upon the motivation behind making these measurements as well as present the design and implementation plan of this planned experiment

    Rapid development of spiral garnets during subduction zone metamorphism revealed from high-resolution Sm-Nd garnet geochronology

    Multiple studies have applied zoned garnet geochronology to place temporal constraints on the rates of metamorphism and deformation during orogenesis. We report new high-resolution isotope dilution–thermal ionization mass spectrometry Sm-Nd isochron ages on concentric growth zones from microstructurally and thermodynamically characterized garnets from the Betic Cordillera, southern Spain. Our ages for the garnet core (13.64 ± 0.31 Ma), mantle (13.41 ± 0.37 Ma), and rim (13.34 ± 0.45 Ma) indicate rapid garnet growth and are consistent with published garnet ages interpreted to reflect high-pressure metamorphism in the region. Thermodynamic analysis indicates garnets grew during subduction at ∼1.5–2.0 GPa and 570–600 °C. The core to rim duration of spiral garnet growth was just a few hundred thousand years. While other zoned garnet studies have shown similar rapid growth in subduction zone settings, this is the first documentation of such rapid growth of a spiral garnet. Combining this garnet growth duration with the magnitude of spiral inclusion trail curvature, we compute a strain rate of ∼10−13 s−1, an order of magnitude faster than all previous spiral garnet studies.     Phase I Study of Ipilimumab Combined with Whole Brain Radiation Therapy or Radiosurgery for Melanoma Patients with Brain Metastases

    Purpose: We performed a phase I study to determine the maximum tolerable dose (MTD) and safety of ipilimumab with stereotactic radiosurgery (SRS) or whole brain radiotherapy (WBRT) in patients with brain metastases (BM) from melanoma. Methods: Based on intracranial (IC) disease burden, patients were treated with WBRT (Arm A) or SRS (Arm B). Ipilimumab starting dose was 3 mg/kg (every 3 weeks, starting on day 3 of WBRT or 2 days after SRS). Ipilimumab was escalated to 10 mg/kg using a two-stage, 3+3 design. The primary endpoint was to determine the MTD of ipilimumab combined with radiotherapy. Secondary endpoints were overall survival (OS), IC and extracranial (EC) control, progression free survival (PFS), and toxicity. This trial is regis- tered with ClinicalTrials.gov, number NCT01703507. Results: Characteristics of the 16 patients enrolled between 2011 and 2014 were: mean age, 60; median BM, 2 (1 to \u3e10); number with EC disease, 13 (81%). Treatment included WBRT (n=5), SRS (n=11), ipilimumab 3mg/kg (n=7), 10 mg/kg (n=9). Median follow-up was 8 months (Arm A) and 10.5 months (Arm B). There were 21 grade 1-2 neuro- toxic effects with no dose-limiting toxicities (DLTs). One patient experienced grade 3 neurotoxicity prior to ipilimumab administration. Ten additional grade 3 toxicities were reported with gastrointestinal (n=5, 31%) as the most common. There were no grade 4/5 toxicities. Median PFS and OS, respectively, in Arm A were 2.5 months and 8 months, and in Arm B were 2.1 months and not reached. Conclusion: Concurrent ipilimumab 10 mg/kg with SRS is safe. The WBRT arm was closed early due to slow accrual, but demonstrated safety with ipilimumab 3 mg/kg. No patient experienced DLT. Larger studies with ipilimumab 10 mg/kg and SRS are warranted

    A new globular cluster black hole in NGC 4472

    We discuss CXOU~1229410+075744, a new black hole candidate in a globular cluster in the elliptical galaxy NGC~4472. By comparing two Chandra observations of the galaxy, we find a source that varies by at least a factor of 4, and has a peak luminosity of at least 2×10392\times10^{39} ergs/sec. As such, the source varies by significantly more than the Eddington luminosity for a single neutron star, and is a strong candidate for being a globular cluster black hole. The source's X-ray spectrum also evolves in a manner consistent with what would be expected from a single accreting stellar mass black hole. We consider the properties of the host cluster of this source and the six other strong black hole X-ray binary candidates, and find that there is suggestive evidence that black hole X-ray binary formation is favored in bright and metal rich clusters, just as is the case for bright X-ray sources in general.Comment: 6 pages, one 2-panel figure, 2 tables; accepted to MNRA

    Jekyll and Hyde: men's constructions of feminism and feminists

    Research and commentary on men's responses to feminism has demonstrated the range of ways in which men have mobilised both against and for feminist principles. This paper argues that further analyses of men's responses require a sophisticated theory of discourse acknowledging the fragmented and contradictory nature of representation. A corpus of men's talk on feminism and feminists was studied to identify the pervasive patterns in men's accounting and regularities in rhetorical organisation. Material from two samples of men was included: a sample of white middle-class 17-18 year old school students and a sample of 60 interviews with a more diverse sample of older men aged 20 to 64. Two interpretative repertoires of feminism and feminists were identified. These set up a 'Jekyll and Hyde' binary and positioned feminism along with feminists very differently as reasonable versus extreme and monstrous. Both repertoires tended to be deployed together and the paper explores the ideological and interactional consequences of typical deployments along with the identity work accomplished by the men as they positioned themselves in relation to these

    Elucidation of The Behavioral Program and Neuronal Network Encoded by Dorsal Raphe Serotonergic Neurons

    Elucidating how the brain's serotonergic network mediates diverse behavioral actions over both relatively short (minutes–hours) and long period of time (days–weeks) remains a major challenge for neuroscience. Our relative ignorance is largely due to the lack of technologies with robustness, reversibility, and spatio-temporal control. Recently, we have demonstrated that our chemogenetic approach (eg, Designer Receptors Exclusively Activated by Designer Drugs (DREADDs)) provides a reliable and robust tool for controlling genetically defined neural populations. Here we show how short- and long-term activation of dorsal raphe nucleus (DRN) serotonergic neurons induces robust behavioral responses. We found that both short- and long-term activation of DRN serotonergic neurons induce antidepressant-like behavioral responses. However, only short-term activation induces anxiogenic-like behaviors. In parallel, these behavioral phenotypes were associated with a metabolic map of whole brain network activity via a recently developed non-invasive imaging technology DREAMM (DREADD Associated Metabolic Mapping). Our findings reveal a previously unappreciated brain network elicited by selective activation of DRN serotonin neurons and illuminate potential therapeutic and adverse effects of drugs targeting DRN neurons
