    Primerjalna študija genetske raznovrstnosti talnih bakterij in gliv v različnih sukcesijah vegetacije na krasu v provinci Guizhou, Kitajska

    To study the soil genetic diversity of bacteria and fungi in different vegetation successions (grassland, shrubbery, primary forest and secondary forest) from the karst area, the Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) technology was applied. The results showed that: (1) the diversity of bacterial communities and the fungal communities in karst area were higher than non karst area in each vegetation succession. Compared with the survey from bacterial (the Shannon index was 2.97 in primary forest, 2.91 in secondary forest, 3.18 in shrubbery, 3.14 in grassland and 2.68 in non karst), fungal diversity between karst areas (the Shannon index was 3.56 in primary forest, 3.78 in secondary forest, 3.73 in shrubbery and 3.70 in grassland) and non karst areas (the Shannon index was 3.08) was more evident, which may be related to the alterations of the composition of plant community and the source of carbon in soil with the vegetation succession of karst ecosystem; (2) The comparation of bacterial diversity index and the richness comprehensively evaluated as follows: shrubbery > grassland > primary forest > nsecondary forest. The diversity index and the richness of fungal communities was as follows: secondary forest > shrubbery > grassland > primary forest. The results suggest that the fungal communities have been greatly changed via vegetation successions, but the diversity index and the richness of the bacterial communities have not been seriously affected. The results provide scientific basis for understanding karst surface ecosystem, which contributes to the future aim of protecting the karst from desertification.Za proučevanje genetske pestrosti talnih bakterij in gliv v različnih sukcesijah vegetacije (travišče, grmičevje, primarni gozd in sekundarni gozd) na krasu je bila uporabljena tehnologija verižne reakcije s polimerazo-denaturirajoča gradientna gelska elektroforeza (PCR-DGGE). Rezultati raziskave so pokazali, da: (1) je bila v vsaki sukcesiji vegetacije pestrost bakterijskih in glivnih združb na kraškem območju višja kot na nekraškem. V primerjavi z bakterijsko raznovrstnostjo (Shannonov indeks je bil 2,97 v primarnem gozdu, 2,91 v sekundarnem gozdu, 3,18 v tleh grmičevja, 3,14 v tleh travišč in 2,68 v nekraškem območju) je bila raznovrstnost gliv med kraškimi območji (Shannonov indeks je bil v primarnem gozdu 3,56, 3,78 v sekundarnem gozdu, 3,73 v tleh grmičevja in 3,70 v tleh travišč) in nekraškimi (Shannonov indeks je bil 3,08) jasneje izražena. To je lahko povezano s spremembami v sestavi rastlinske združbe in vira ogljika v tleh glede na stanje sukcesije v vegetaciji kraškega ekosistema. (2) Primerjava kazalnikov bakterijske raznovrstnosti in abundance je bila celostno ovrednotena in sledi takole: grmičevje > travišče > primarni gozd > sekundarni gozd. Kazalnika raznovrstnosti in abundance glivnih združb kažeta sledeči trend: sekundarni gozd > grmičevje > travišče > primarni gozd. Rezultati izkazujejo, da so se glivne združbe precej spremenile zaradi sukcesije v vegetaciji, vendar pa na drugi strani ni bilo bistvenega vpliva na kazalnika bakterijske raznovrstnosti in abundance. Rezultati med drugim dajejo tudi znanstveno podlago za razumevanje delovanja kraškega površinskega ekosistema, kar ključno prispeva k cilju zaščite krasa pred dezertifikacijo (širjenjem puščav)

    Laparoscopic distal gastrectomy demonstrates acceptable outcomes regarding complications compared to open surgery for gastric cancer patients with pylorus outlet obstruction

    BackgroundFor gastric cancer (GC) patients with pylorus outlet obstruction (POO), whether laparoscopic surgery has advantages over open surgery remains unclear. This study aims to investigate the differences between patients with and without POO in open and laparoscopic groups and to determine the differences between laparoscopic distal gastrectomy (LDG) and open distal gastrectomy (ODG) in GC patients with POO.MethodsA total of 241 GC patients with POO who underwent distal gastrectomy at the Department of Gastric Surgery of the First Affiliated Hospital of Nanjing Medical University between 2016 and 2021 were included in this study. A total of 1,121 non-POO patients who underwent laparoscopic surgery and 948 non-POO patients who underwent open surgery from 2016 to 2021 were also enrolled in the study. We compared complication rates and hospital stays between open and laparoscopic groups.ResultsThere was no significant difference for LDG between GC patients with and without POO regarding the overall complication rates (P = 0.063), the Grade III–V complication rate (P = 0.673), and the anastomotic complication rate (P = 0.497) from 2016 to 2021. The patients with POO had longer preoperative hospital stay (P = 0.001) and postoperative hospital stay (P=0.007) compared to patients without POO. No significant difference was observed for open patients between POO and non-POO patients regarding the overall complication rate (P = 0.357), grade III–V complication rate (P = 1.000), and anastomosis-related complication rate (P = 0.766). Compared with open surgery in GC patients with POO (n = 111), the total complication rate of the LDG group was 16.2%, which was significantly lower than that of the open group (26.1%, P = 0.041). No significant differences in the Grade III–V complication rate (P = 0.574) and anastomotic complication rate (P = 0.587) were observed between laparoscopic and open groups. Patients receiving laparoscopic surgery had shorter postoperative hospital stay than open surgery (P = 0.001). More resected lymph nodes (LNs) were also observed in the laparoscopic group (P = 0.0145).ConclusionThe comorbidity of GC with POO does not increase the complication rate after laparoscopic or open distal gastrectomy. In GC patients with POO, laparoscopic surgery shows advantages over open surgery with a lower overall complication rate, shorter postoperative hospital stay, and more harvested lymph nodes. Laparoscopic surgery is a safe, feasible, and effective treatment for GC with POO

    Associations of dietary factors with gastric cancer risk: insights from NHANES 2003–2016 and mendelian randomization analyses

    Background: Gastric cancer (GC) continues to be one of the leading causes of cancer-related deaths globally. Diet significantly influences the incidence and progression of GC. However, the relationship between dietary intake and GC is inconsistent.Methods: A study was conducted with adults who participated in the National Health and Nutrition Examination Survey (NHANES) from 2003 to 2016 to investigate possible associations between 32 dietary factors and GC. To further detect potential causal relationships between these dietary factors and the risk of GC, a two-sample Mendelian randomization (MR) analysis was conducted. The primary method employed was the inverse variance weighted (IVW) analysis, and its results were further validated by four other methods.Results: Of the 35,098 participants surveyed, 20 had a history of GC. Based on the results of weighted logistic multivariate analysis, it was observed that there was a positive correlation between total fat intake [odds ratio (OR) = 1.09, 95% confidence interval (CI): (1.01–1.17), p = 0.03] and GC as well as negative association of dietary monounsaturated fatty acids (MUFAs) intake [OR = 0.83, 95% CI: (0.76–0.92), p < 0.001]. Further evaluations of the odds of GC across the quartiles of dietary MUFAs showed that the top quartile of total MUFA intake was associated with a lower likelihood of GC in three different models [model1: OR = 0.03, 95% CI: (0.00–0.25), p < 0.01; model2: OR = 0.04, 95% CI: (0.00–0.38), p = 0.01; model3: OR = 0.04, 95% CI: (0.00–0.40), p = 0.01]. For the MR analyses, genetic instruments were selected from the IEU Open GWAS project; IVW analysis showed that GC risk was not associated with MUFAs [OR = 0.82, 95% CI: (0.59–1.14), p = 0.23] or the ratio of MUFAs to total fatty acids [OR = 1.00, 95% CI: (0.75–1.35), p = 0.98]. Similar results were observed when using the other MR methods.Conclusion: The NHANES study revealed that consuming MUFAs was linked to a lower risk of GC, although the results of MR analyses do not provide evidence of a causal relationship. Additional research is therefore necessary to clarify these findings

    Baichuan 2: Open Large-scale Language Models

    Large language models (LLMs) have demonstrated remarkable performance on a variety of natural language tasks based on just a few examples of natural language instructions, reducing the need for extensive feature engineering. However, most powerful LLMs are closed-source or limited in their capability for languages other than English. In this technical report, we present Baichuan 2, a series of large-scale multilingual language models containing 7 billion and 13 billion parameters, trained from scratch, on 2.6 trillion tokens. Baichuan 2 matches or outperforms other open-source models of similar size on public benchmarks like MMLU, CMMLU, GSM8K, and HumanEval. Furthermore, Baichuan 2 excels in vertical domains such as medicine and law. We will release all pre-training model checkpoints to benefit the research community in better understanding the training dynamics of Baichuan 2.Comment: Baichuan 2 technical report.     An Accurate Power Flow Method for Microgrids with Conventional Droop Control

    With conventional droop control, the droop relationship between the voltage and reactive power is not purely linear because of the filter reactance. This paper focuses on the theoretical analysis to account for this characteristic and presents a precise power flow method for conventional droop control. The proposed method is universal, which can handle not only conventional droop control, but also other control strategies, such as robust droop control, constant power control, and constant voltage–frequency control. It can also handle frequency-dependent active and reactive power loads and is adapted for islanded and grid-connected systems. The proposed method extends the applicability of conventional power flow methods to microgrids so that the framework of the method is generic; any conventional power flow algorithm can be adapted to this framework. Compared with the time-domain simulation method, the proposed method is accurate, simple, and easy to implement for industrial applications

    Complex Affine Arithmetic-Based Power Flow Analysis for Zonal Medium Voltage Direct Current Shipboard Power Systems in the Presence of Power Variation

    The medium-voltage direct-current (MVDC) ship power system has been extensively investigated and discussed in recent years. This paper presents an alternating current/direct-current (AC/DC) power flow algorithm based on complex affine arithmetic for the zonal MVDC shipboard power system in the presence of power variation. The power converter effect is considered, and an affine power converter model is proposed in the power flow model. An affine-analysis-based sequential method is adopted to solve AC/DC power flows. With the proposed algorithm, the bounds of bus voltages can be effectively obtained. A relative influence factor is defined to quantitatively assess the effect of power variation on voltage profile. Four cases are studied in detail to demonstrate the applicability of the proposed algorithm. The proposed algorithm is useful in network architecture design, planning, and online operations of MVDC power systems when such decisions are subject to power variation

    Complete chloroplast genome of a novel chlorophyll-deficient mutant (clm) in sweetpotato (Ipomoea batatas L.)

    The complete chloroplast genome of a novel chlorophyll-deficient mutant (clm) and its wild type (WT) in sweetpotato (Ipomoea batatas L.) was sequenced. The complete chloroplast genome of clm and WT was 161,393 bp and 161,429 bp in length, containing a large single copy (LSC) region of 87,561 bp and 87,597 bp, respectively, a small single copy (SSC) region with the same length of 30,890 bp and a pair of inverted repeat regions (IRs) with the same length of 12,052 bp. Both of them contained 132 genes including 87 protein-coding sequences, 37 tRNA, and eight rRNA. Comparing to the WT, four SNPs and three INDELs were detected and only one INDEL in the exon affecting the translation of rpoA gene. Phylogenetic analysis showed that clm and WT were closely related to Ipomoea tabascana. The complete chloroplast genome of clm and its WT will play a role in understanding the molecular mechanism of chlorophyll deficiency and developing molecular markers in sweetpotato

    Ecosystem health assessment in the pearl river estuary of China by considering ecosystem coordination.

    Marine ecosystem is a complex nonlinear system. However, ecosystem health assessment conventionally builds on a linear superposition of changes in ecosystem components and probably fails to evaluate nonlinear interactions among various components. To better reflect the intrinsic interactions and their impacts on ecosystem health, an ecosystem coordination index, defined as the matching level of ecosystem structure/services, is proposed and incorporated into the ecosystem health index for a systematic diagnosis in the Pearl River Estuary, China. The analysis results show that the ecosystem health index over the last three decades decreased from 0.91 to 0.50, indicating deteriorating from healthy to unhealthy status. The health index is 3-16% lower than that calculated using the common method without considering ecosystem coordination. Ecosystem health degradation in the Pearl River Estuary manifested as significant decreases in structure/services and somewhat mismatching among them. Overall, the introduction of coordination in ecosystem health assessment could improve the understanding of the mechanism of marine ecosystem change and facilitate effective restoration of ecosystem health

    An improved method for the molecular identification of single dinoflagellate cysts

    Background Dinoflagellate cysts (i.e., dinocysts) are biologically and ecologically important as they can help dinoflagellate species survive harsh environments, facilitate their dispersal and serve as seeds for harmful algal blooms. In addition, dinocysts derived from some species can produce more toxins than vegetative forms, largely affecting species through their food webs and even human health. Consequently, accurate identification of dinocysts represents the first crucial step in many ecological studies. As dinocysts have limited or even no available taxonomic keys, molecular methods have become the first priority for dinocyst identification. However, molecular identification of dinocysts, particularly when using single cells, poses technical challenges. The most serious is the low success rate of PCR, especially for heterotrophic species. Methods In this study, we aim to improve the success rate of single dinocyst identification for the chosen dinocyst species (Gonyaulax spinifera, Polykrikos kofoidii, Lingulodinium polyedrum, Pyrophacus steinii, Protoperidinium leonis and Protoperidinium oblongum) distributed in the South China Sea. We worked on two major technical issues: cleaning possible PCR inhibitors attached on the cyst surface and designing new dinoflagellate-specific PCR primers to improve the success of PCR amplification. Results For the cleaning of single dinocysts separated from marine sediments, we used ultrasonic wave-based cleaning and optimized cleaning parameters. Our results showed that the optimized ultrasonic wave-based cleaning method largely improved the identification success rate and accuracy of both molecular and morphological identifications. For the molecular identification with the newly designed dinoflagellate-specific primers (18S634F-18S634R), the success ratio was as high as 86.7% for single dinocysts across multiple taxa when using the optimized ultrasonic wave-based cleaning method, and much higher than that (16.7%) based on traditional micropipette-based cleaning. Discussion The technically simple but robust method improved on in this study is expected to serve as a powerful tool in deep understanding of population dynamics of dinocysts and the causes and consequences of potential negative effects caused by dinocysts
