    SAG2 locus genotyping of Toxoplasma gondii in meat products of East Azerbaijan Province, North West of Iran During 2010-2011

    Toxoplasmosis is an infection caused by Toxoplasma gondii, an intracellular obligate parasite. Its transmission is usually attributed to ingestion of undercooked or raw meat. The aim of this study was the detection and genotyping of T. gondii in meat products using the molecular method in East Azerbaijan. DNA was extracted from 164 meat products sample obtained from 15 commercial establishments. Nested polymerase chain reaction with specific primers for Toxoplasma gondii SAG2 locus was used for detection of the parasite in samples. Genotyping was carried out by digestion of PCR products with restriction enzymes Cfo1 and Sau3AI. T. gondii DNA was detected in 16.6, 19.1, 15 and 56.6%, in salami, sausages, and hamburger and kebab samples, respectively. Genotyping by restriction fragment length polymorphism (RFLP) analysis of SAG2 locus revealed that all of the samples belonged to genotype I. The detection of the parasite in uncooked meat and commercial meat products, and the high ratio of seropositive slaughtered animals, emphasis that the risk still exists for food -born toxoplasmosis.Key words: Toxoplasma gondii, SAG2, genotyping, meat products

    Serological survey and comparison of two polymerase chain reaction (PCR) assays with enzyme-linked immunosorbent assay (ELISA) for the diagnosis of canine visceral leishmaniasis in dogs

    Visceral leishmaniasis (VL) is systemic zoonotic parasitic infection that is a health problem in some tropical and subtropical countries. The purpose of our study is to determine the seroprevalence of canine visceral leishmaniasis (CVL) in owned dogs of the Sarab area and to identify the species of Leishmania isolated from these dogs. We also compared the sensitivities and specificities of two polymerase chain reaction (PCR) assays (kDNA and ITS1) used for Leishmania infantum identification with culture, microscopic detection and enzyme-linked immunosorbent assay (ELISA) methods as well as validate the PCR techniques for the molecular diagnosis of CVL. Sera from 384 dogs of 30 villages around Sarab, were tested by ELISA and buffy coat blood fractions after sampling tested with PCR by specific primers (kDNA, ITS-18sRNA). Thirty-five dogs were seropositive by ELISA. The seroprevalence rate (SPR) of CVL was 9.1% (CI, 95% 6.6 -12.4). The most important serological result was a high proportion of seropositivity for leishmaniasis. Out of 361 (94%) asymptomatic dogs, 31 (8.6%) were seropositive, and out of 23 (6%) symptomatic dogs, 4 (17.4%) were seropositive. Agreement betweenthe ELISA test and clinical signs was 86.7%. Each assay was performed on 60 blood samples. PCR of kDNA (7/60 positives, 11.8%) was the most sensitive of the assays examined, followed by ELISA (3/60, 5%) and ITS1-PCR (2/60, 3.4 %). All diagnostic assays were highly specific (100 %) and had positive predictive values (PPV) >90% and negative predictive values (NPV) >88% for CVL. As expected, kDNAPCR proved to be the most sensitive (87.5 %) assay for leishmanial DNA in peripheral blood. This study shows that kDNA-PCR is significantly more sensitive than the other parasitological and serological methods, allowing the identification of infected dogs even before the appearance of serum L. infantum antibodies. Because kDNA-PCR is the most reliable, sensitive, and also a rapid diagnostic assay for CVL, it should be employed as the new standard for routine diagnosis.Key words: Leishmania infantum, polymerase chain reaction, kinetoplast DNA, enzyme-linked immunosorbent assay, Visceral leishmaniasis, dogs, prevalence

    An empirical study on measuring the relative efficiency using DEA method: A case study of bank industry

    Data Envelopment Analysis (DEA) has been widely used as an effective tool for measuring the relative efficiency of similar units by considering various input/output parameters. This paper examines DEA models for the estimation and improvement of organizational inputs and outputs in order to enhance management and decision making processes. We propose an empirical DEA analysis on banking sector by considering several financial and non-financial inputs and outputs. The relative efficiencies of various branches of banks are analyzed in different scenarios. The preliminary results indicate that there are some non-financial items that could significantly change the overall performance of a unit along with other financial items

    A Roadmap for the Production of a GMP-Compatible Cell Bank of Allogeneic Bone Marrow-Derived Clonal Mesenchymal Stromal Cells for Cell Therapy Applications

    Background: Allogeneic mesenchymal stromal cells (MSCs) have been used extensively in various clinical trials. Nevertheless, there are concerns about their efficacy, attributed mainly to the heterogeneity of the applied populations. Therefore, producing a consistent population of MSCs is crucial to improve their therapeutic efficacy. This study presents a good manufacturing practice (GMP)-compatible and cost-effective protocol for manufacturing, banking, and lot-release of a homogeneous population of human bone marrow-derived clonal MSCs (cMSCs). Methods: Here, cMSCs were isolated based on the subfractionation culturing method. Afterward, isolated clones that could reproduce up to passage three were stored as the seed stock. To select proliferative clones, we used an innovative, cost-effective screening strategy based on lengthy serial passaging. Finally, the selected clones re-cultured from the seed stock to establish the following four-tired cell banking system: initial, master, working, and end of product cell banks (ICB, MCB, WCB, and EoPCB). Results: Through a rigorous screening strategy, three clones were selected from a total of 21 clones that were stored during the clonal isolation process. The selected clones met the identity, quality, and safety assessments criteria. The validated clones were stored in the four-tiered cell bank system under GMP conditions, and certificates of analysis were provided for the three-individual ready-to-release batches. Finally, a stability study validated the EoPCB, release, and transport process of the frozen final products. Conclusion: Collectively, this study presents a technical and translational overview of a GMP-compatible cMSCs manufacturing technology that could lead to the development of similar products for potential therapeutic applications.     Flat Spectrum Radio Quasars through the MAGIC glasses

    The detection of Flat Spectrum Radio Quasars (FSRQs) in the Very High Energy (VHE, E > 100 GeV) range is challenging, mainly because of their steep spectra in this energy band. Up to now, only five FSRQs are known to be VHE γ-ray emitters, all of them have been detected by the MAGIC telescopes, that discovered four of them in the VHE band. The observations in the VHE band are crucial to understand their emission, specially to constrain the location of the emitting region within the jet due to the absorption from their broad line region (BLR). Typically, FSRQs are detected during high flux states, enhancing the probability of detection with the current instruments sensitivities. However, the last observation campaigns performed with the MAGIC telescopes show emission during moderate states, thus challenging our understanding of the emission mechanisms in FSRQs. In this contribution, we give an overview and present the most recent results of the three FSRQs 3C279, PKS1222+21 and PKS1510-089 in a multi-wavelength context with special focus on MAGIC and Fermi-LAT simultaneous observations

    Role of the employment status and education of mothers in the prevalence of intestinal parasitic infections in Mexican rural schoolchildren

    <p><b>Background:</b> Intestinal parasitic infections are a public health problem in developing countries such as Mexico. As a result, two governmental programmes have been implemented: a) "National Deworming Campaign" and b) "Opportunities" aimed at maternal care. However, both programmes are developed separately and their impact is still unknown. We independently investigated whether a variety of socio-economic factors, including maternal education and employment levels, were associated with intestinal parasite infection in rural school children.</p> <p><b>Methods:</b> This cross-sectional study was conducted in 12 rural communities in two Mexican states. The study sites and populations were selected on the basis of the following traits: a) presence of activities by the national administration of albendazole, b) high rates of intestinal parasitism, c) little access to medical examination, and d) a population having less than 2,500 inhabitants. A total of 507 schoolchildren (mean age 8.2 years) were recruited and 1,521 stool samples collected (3 per child). Socio-economic information was obtained by an oral questionnaire. Regression modelling was used to determine the association of socio-economic indicators and intestinal parasitism.</p> <p><b>Results:</b> More than half of the schoolchildren showed poliparasitism (52%) and protozoan infections (65%). The prevalence of helminth infections was higher in children from Oaxaca (53%) than in those from Sinaloa (33%) (p < 0.0001). Giardia duodenalis and Hymenolepis nana showed a high prevalence in both states. Ascaris lumbricoides, Trichuris trichiura and Entamoeba hystolitica/dispar showed low prevalence. Children from lower-income families and with unemployed and less educated mothers showed higher risk of intestinal parasitism (odds ratio (OR) 6.0, 95% confidence interval (CI) 1.6–22.6; OR 4.5, 95% CI 2.5–8.2; OR 3.3, 95% CI 1.5–7.4 respectively). Defecation in open areas was also a high risk factor for infection (OR 2.4, 95% CI 2.0–3.0).</p> <p><b>Conclusion:</b> Intestinal parasitism remains an important public health problem in Sinaloa (north-western Mexico) and Oaxaca (south-eastern Mexico). Lower income, defecation in open areas, employment status and a lower education level of mothers were the significant factors related to these infections. We conclude that mothers should be involved in health initiatives to control intestinal parasitism in Mexico.</p&gt

    Detection of very high energy gamma-ray emission from the gravitationally-lensed blazar QSO B0218+357 with the MAGIC telescopes

    Context. QSO B0218+357 is a gravitationally lensed blazar located at a redshift of 0.944. The gravitational lensing splits the emitted radiation into two components, spatially indistinguishable by gamma-ray instruments, but separated by a 10-12 day delay. In July 2014, QSO B0218+357 experienced a violent flare observed by the Fermi-LAT and followed by the MAGIC telescopes. Aims. The spectral energy distribution of QSO B0218+357 can give information on the energetics of z ~ 1 very high energy gamma- ray sources. Moreover the gamma-ray emission can also be used as a probe of the extragalactic background light at z ~ 1. Methods. MAGIC performed observations of QSO B0218+357 during the expected arrival time of the delayed component of the emission. The MAGIC and Fermi-LAT observations were accompanied by quasi-simultaneous optical data from the KVA telescope and X-ray observations by Swift-XRT. We construct a multiwavelength spectral energy distribution of QSO B0218+357 and use it to model the source. The GeV and sub-TeV data, obtained by Fermi-LAT and MAGIC, are used to set constraints on the extragalactic background light. Results. Very high energy gamma-ray emission was detected from the direction of QSO B0218+357 by the MAGIC telescopes during the expected time of arrival of the trailing component of the flare, making it the farthest very high energy gamma-ray sources detected to date. The observed emission spans the energy range from 65 to 175 GeV. The combined MAGIC and Fermi-LAT spectral energy distribution of QSO B0218+357 is consistent with current extragalactic background light models. The broad band emission can be modeled in the framework of a two zone external Compton scenario, where the GeV emission comes from an emission region in the jet, located outside the broad line region.Comment: 11 pages, 6 figures, accepted for publication in A&

    A search for spectral hysteresis and energy-dependent time lags from X-ray and TeV gamma-ray observations of Mrk 421

    Blazars are variable emitters across all wavelengths over a wide range of timescales, from months down to minutes. It is therefore essential to observe blazars simultaneously at different wavelengths, especially in the X-ray and gamma-ray bands, where the broadband spectral energy distributions usually peak. In this work, we report on three "target-of-opportunity" (ToO) observations of Mrk 421, one of the brightest TeV blazars, triggered by a strong flaring event at TeV energies in 2014. These observations feature long, continuous, and simultaneous exposures with XMM-Newton (covering X-ray and optical/ultraviolet bands) and VERITAS (covering TeV gamma-ray band), along with contemporaneous observations from other gamma-ray facilities (MAGIC and Fermi-LAT) and a number of radio and optical facilities. Although neither rapid flares nor significant X-ray/TeV correlation are detected, these observations reveal subtle changes in the X-ray spectrum of the source over the course of a few days. We search the simultaneous X-ray and TeV data for spectral hysteresis patterns and time delays, which could provide insight into the emission mechanisms and the source properties (e.g. the radius of the emitting region, the strength of the magnetic field, and related timescales). The observed broadband spectra are consistent with a one-zone synchrotron self-Compton model. We find that the power spectral density distribution at 4×104\gtrsim 4\times 10^{-4} Hz from the X-ray data can be described by a power-law model with an index value between 1.2 and 1.8, and do not find evidence for a steepening of the power spectral index (often associated with a characteristic length scale) compared to the previously reported values at lower frequencies.Comment: 45 pages, 15 figure
