    Dosage des polyphénols et activité anti-oxydante de feuilles et d’inflorescences mâles de Borassus aethiopum, Mart. (Arecaceae)

    Borassus aethiopum est une plante de la famille des Arécacées. C’est un géant palmier qui présente un tronc dur appelé stipe, des racines fasciculées et des feuilles longues en forme d’éventail. Elle est presque cosmopolite et ses différentes parties sont utilisées dans nombreux domaines : économiques, écologiques,  alimentaires et sanitaire. En effet, sur le plan sanitaire, la pharmacopée utilise l’alcinât des feuilles en boisson dans l’eau pour traiter la bilharziose viscérale  (Sakande, 2004). L’objet de notre étude est de déterminer la teneur en polyphénols et de tester l’activité antioxydante d’extraits de feuilles en comparaison avec ceux des inflorescences mâles. Ainsi, la méthode colorimétrique de Folin Denis, décrite par Bassène (2012) a été utilisée pour le dosage des polyphénols. L’activité antioxydante a été déterminée par les méthodes chromatographique sur couche mince selon Takao et al. (1994) et colorimétrique décrite par Sakande (2004). Les teneurs en polyphénols totaux obtenues sont 13,56% et 5,6% Eq d’acide tannique respectivement dans les feuilles et les inflorescences mâles. Les teneurs  respectives en tanins dans les feuilles et les inflorescences mâles ont été évaluées à 13,46% et 5,47% Eq d’acide tannique. La chromatographie sur couche mince a révélé une activité antioxydante dans les extraits de feuilles et d’inflorescences mâles. Par la méthode colorimétrique utilisant le DPPH, les  résultats obtenus en mg Eq d’acide ascorbique sont 4 et 5 mg Eq respectivement dans les extraits d’acétate éthyle et butanolique de feuilles ; les extraits d’acétate d’éthyle et  butanolique  d’inflorescences mâles respectifs ont une activité antioxydante évaluée à 10 et 20 mgEq d’acide ascorbique. Les résultats obtenus de cette étude  montre que les extraits de feuilles ont des teneurs en polyphénols et en tanins supérieures à celles des extraits d’inflorescences mâles. L’activité antioxydante a été deux fois plus importante dans les extraits de feuilles que dans les extraits d’inflorescences mâles. Cependant, dans les feuilles l’extrait d’acétate d’éthyle  renferme deux fois plus de polyphénols et de tanins que l’extrait butanolique ; l’activité antioxydante est aussi plus élevée dans ce premier extrait.Mots clés : Borassus aethiopum, feuilles, inflorescences mâles, polyphénols, tanins, activité antioxydante

    Le rônier est un palmier très utilisé dans la pharmacopée traditionnelle ; Le calcinât des feuilles est utilisé en boisson dans l’eau pour traiter la bilharziose viscérale. La tigelle de la noix germée en décoction lutte contre l’impuissance sexuelle, une activité anti-inflammatoire avec une inhibition de 70% de l’oedème induite par la carragénine a été révélée dans les extraits d’inflorescences mâles. La méthode de l’oedème induit par la carragénine de Winter a été utilisée sur des rats de souche Wistar pour mettre en évidence l’activité antiinflammatoire des feuilles de Borassus aethiopum. L’extrait d’acétate d’éthyle et sa fraction F1 à la dose 30 mg/kg ont une activité anti-inflammatoire voisine de celle de l’aspirine à 30 mg/kg; cette activité (exprimée en pourcentage d’inhibition) est respectivement de 79,21%, 73,16 et 92,53% à la première heure. Ainsi à la cinquième heure ces pourcentages sont 69,18%, 54,59% et 58,75% respectivement pour la référence, l’extrait d’acétate d’éthyle et la fraction F1.Mots clés: Borassus aethiopum Mart, feuilles et activité anti-inflammatoire

    Etude de l’activité antioxydante des extraits des feuilles de Vitex doniana (Verbenacea)

    L’objectif de la présente étude était d’évaluer l’activité antioxydante des extraits des feuilles de Vitex doniana par spectrophotométrie en utilisant les méthodes de piégeage des radicaux libres 2,2-diphényl-1- picryl-hydrazyle (DPPH•) et acide 2,2’-azino-bis-(3-éthylbenzothiazoline-6-sulfonique) (ABTS+•). Une extraction éthanolique des feuilles de Vitex doniana a été effectuée au Soxhlet. L’extrait sec recueilli a été redissout dans de l’eau puis fractionné en utilisant successivement de l’hexane, du dichlorométhane et de l’acétate d’éthyle. Les propriétés antioxydantes de l’extrait éthanolique et celles de ses différentes fractions ont été évaluées aux concentrations de 2,5 ; 10 ; 100 et 200 μg/mL. Les pourcentages d’inhibition (PI) expriment l’effet antioxydant mesuré. L’extrait éthanolique et ses différentes fractions ont présenté une activité de piégeage des deux radicaux libres. Lors des tests d’inhibition de l’absorbance du radical DPPH•, les PI ont varié de (18,15±0,01)% pour la fraction hexanique (2,5 μg/mL) à (92,45±0,01)% pour la fraction d’acétate d’éthyle (100 μg/mL). Avec le radical ABTS+•, les PI ont varié de (52,76±0,05)% pour la fraction hexanique (2,5 μg/mL) à (98,27±0,12)% pour la fraction d’acétate d’éthyle (100 μg/mL). Ainsi, l’extrait éthanolique des feuilles de Vitex doniana et ses différentes fractions possèdent un pouvoir antioxydant significatif.Mots clés : Activité antioxydante, DPPH, ABTS, Vitex doniana

    Hybridized zoonotic schistosoma infections result in hybridized morbidity profiles: a clinical morbidity study amongst co-infected human populations of Senegal

    Hybridization of infectious agents is a major emerging public and veterinary health concern at the interface of evolution, epidemiology, and control. Whilst evidence of the extent of hybridization amongst parasites is increasing, their impact on morbidity remains largely unknown. This may be predicted to be particularly pertinent where parasites of animals with contrasting pathogenicity viably hybridize with human parasites. Recent research has revealed that viable zoonotic hybrids between human urogenital Schistosoma haematobium with intestinal Schistosoma species of livestock, notably Schistosoma bovis, can be highly prevalent across Africa and beyond. Examining human populations in Senegal, we found increased hepatic but decreased urogenital morbidity, and reduced improvement following treatment with praziquantel, in those infected with zoonotic hybrids compared to non-hybrids. Our results have implications for effective monitoring and evaluation of control programmes, and demonstrate for the first time the potential impact of parasite hybridizations on host morbidity

    Attributes and predictors of long COVID

    Reports of long-lasting coronavirus disease 2019 (COVID-19) symptoms, the so-called ‘long COVID’, are rising but little is known about prevalence, risk factors or whether it is possible to predict a protracted course early in the disease. We analyzed data from 4,182 incident cases of COVID-19 in which individuals self-reported their symptoms prospectively in the COVID Symptom Study app. A total of 558 (13.3%) participants reported symptoms lasting ≥28 days, 189 (4.5%) for ≥8 weeks and 95 (2.3%) for ≥12 weeks. Long COVID was characterized by symptoms of fatigue, headache, dyspnea and anosmia and was more likely with increasing age and body mass index and female sex. Experiencing more than five symptoms during the first week of illness was associated with long COVID (odds ratio = 3.53 (2.76–4.50)). A simple model to distinguish between short COVID and long COVID at 7 days (total sample size, n = 2,149) showed an area under the curve of the receiver operating characteristic curve of 76%, with replication in an independent sample of 2,472 individuals who were positive for severe acute respiratory syndrome coronavirus 2. This model could be used to identify individuals at risk of long COVID for trials of prevention or treatment and to plan education and rehabilitation services

    Symptom clusters in COVID-19: A potential clinical prediction tool from the COVID Symptom Study app

    As no one symptom can predict disease severity or the need for dedicated medical support in coronavirus disease 2019 (COVID-19), we asked whether documenting symptom time series over the first few days informs outcome. Unsupervised time series clustering over symptom presentation was performed on data collected from a training dataset of completed cases enlisted early from the COVID Symptom Study Smartphone application, yielding six distinct symptom presentations. Clustering was validated on an independent replication dataset between 1 and 28 May 2020. Using the first 5 days of symptom logging, the ROC-AUC (receiver operating characteristic - area under the curve) of need for respiratory support was 78.8%, substantially outperforming personal characteristics alone (ROC-AUC 69.5%). Such an approach could be used to monitor at-risk patients and predict medical resource requirements days before they are required

    Quantitative imaging of concentrated suspensions under flow

    We review recent advances in imaging the flow of concentrated suspensions, focussing on the use of confocal microscopy to obtain time-resolved information on the single-particle level in these systems. After motivating the need for quantitative (confocal) imaging in suspension rheology, we briefly describe the particles, sample environments, microscopy tools and analysis algorithms needed to perform this kind of experiments. The second part of the review focusses on microscopic aspects of the flow of concentrated model hard-sphere-like suspensions, and the relation to non-linear rheological phenomena such as yielding, shear localization, wall slip and shear-induced ordering. Both Brownian and non-Brownian systems will be described. We show how quantitative imaging can improve our understanding of the connection between microscopic dynamics and bulk flow.Comment: Review on imaging hard-sphere suspensions, incl summary of methodology.     Multiple populations in globular clusters. Lessons learned from the Milky Way globular clusters

    Recent progress in studies of globular clusters has shown that they are not simple stellar populations, being rather made of multiple generations. Evidence stems both from photometry and spectroscopy. A new paradigm is then arising for the formation of massive star clusters, which includes several episodes of star formation. While this provides an explanation for several features of globular clusters, including the second parameter problem, it also opens new perspectives about the relation between globular clusters and the halo of our Galaxy, and by extension of all populations with a high specific frequency of globular clusters, such as, e.g., giant elliptical galaxies. We review progress in this area, focusing on the most recent studies. Several points remain to be properly understood, in particular those concerning the nature of the polluters producing the abundance pattern in the clusters and the typical timescale, the range of cluster masses where this phenomenon is active, and the relation between globular clusters and other satellites of our Galaxy.Comment: In press (The Astronomy and Astrophysics Review

    Cytoplasmic CUG RNA Foci Are Insufficient to Elicit Key DM1 Features

    The genetic basis of myotonic dystrophy type I (DM1) is the expansion of a CTG tract located in the 3′ untranslated region of DMPK. Expression of mutant RNAs encoding expanded CUG repeats plays a central role in the development of cardiac disease in DM1. Expanded CUG tracts form both nuclear and cytoplasmic aggregates, yet the relative significance of such aggregates in eliciting DM1 pathology is unclear. To test the pathophysiology of CUG repeat encoding RNAs, we developed and analyzed mice with cardiac-specific expression of a beta-galactosidase cassette in which a (CTG)400 repeat tract was positioned 3′ of the termination codon and 5′ of the bovine growth hormone polyadenylation signal. In these animals CUG aggregates form exclusively in the cytoplasm of cardiac cells. A key pathological consequence of expanded CUG repeat RNA expression in DM1 is aberrant RNA splicing. Abnormal splicing results from the functional inactivation of MBNL1, which is hypothesized to occur due to MBNL1 sequestration in CUG foci or from elevated levels of CUG-BP1. We therefore tested the ability of cytoplasmic CUG foci to elicit these changes. Aggregation of CUG RNAs within the cytoplasm results both in Mbnl1 sequestration and in approximately a two fold increase in both nuclear and cytoplasmic Cug-bp1 levels. Significantly, despite these changes RNA splice defects were not observed and functional analysis revealed only subtle cardiac dysfunction, characterized by conduction defects that primarily manifest under anesthesia. Using a human myoblast culture system we show that this transgene, when expressed at similar levels to a second transgene, which encodes expanded CTG tracts and facilitates both nuclear focus formation and aberrant splicing, does not elicit aberrant splicing. Thus the lack of toxicity of cytoplasmic CUG foci does not appear to be a consequence of low expression levels. Our results therefore demonstrate that the cellular location of CUG RNA aggregates is an important variable that influences toxicity and support the hypothesis that small molecules that increase the rate of transport of the mutant DMPK RNA from the nucleus into the cytoplasm may significantly improve DM1 pathology

    Do you get what you pay for? Sales incentives and implications for motivation and changes in turnover intention and work effort

    This study investigated relations between pay-for-performance incentives designed to vary in instrumentality (annual pay-for-performance, quarterly pay-for-performance, and base pay level) and employee outcomes (self-reported work effort and turnover intention) in a longitudinal study spanning more than 2 years. After controlling for perceived instrumentality, merit pay increase, and the initial values of the dependent variables, the amount of base pay was positively related to work effort and negatively related to turnover intention, where both relationships were mediated by autonomous motivation. The amounts of quarterly and annual pay-for-performance were both positively related to controlled motivation, but were differently related to the dependent variables due to different relations with autonomous motivation
