    Comment on ``Evidence Against Instanton Dominance of Topological Charge Fluctuations in QCD''

    We comment on the recent paper (hep-lat/0102003) by Horvath, Isgur, McCune, and Thacker, which concludes that the local chiral structure of fermionic eigenmodes is not consistent with instanton dominance.     Investigating and Optimizing the Chiral Properties of Lattice Fermion Actions

    We study exceptional modes of both the Wilson and the clover action in order to understand why quenched clover spectroscopy suffers so severely from exceptional configurations. We show that, in contrast to the case of the Wilson action, a large clover coefficient can make the exceptional modes extremely localized and thus very sensitive to short distance fluctuations. We describe a way to optimize the chiral behavior of Wilson-type lattice fermion actions by studying their low energy real eigenmodes. We find a candidate action, the clover action with fat links with a tuned clover term. We present a calculation of spectroscopy and matrix elements at Wilson gauge coupling beta=5.7. When compared to simulations with the standard (nonperturbatively improved) clover action at small lattice spacing, the action shows good scaling behavior, with an apparent great reduction in the number of exceptional configurations.Comment: 29 pages, LaTeX with 24 eps figures; Due to the suggestion of a referee, the previous version of this paper has been merged with hep-lat/9807002.     miR-221/222 control luminal breast cancer tumor progression by regulating different targets

    α6β4 integrin is an adhesion molecule for laminin receptors involved in tumor progression. We present a link between β4 integrin expression and miR-221/222 in the most prevalent human mammary tumor: luminal invasive carcinomas (Lum-ICs). Using human primary tumors that display different β4 integrin expression and grade, we show that miR-221/222 expression inversely correlates with tumor proliferating index, Ki67. Interestingly, most high-grade tumors express β4 integrin and low miR-221/222 levels. We ectopically transfected miR-221/222 into a human-derived mammary tumor cell line that recapitulates the luminal subtype to investigate whether miR-221/222 regulates β4 expression. We demonstrate that miR-221/222 overexpression results in β4 expression downregulation, breast cancer cell proliferation, and invasion inhibition. The role of miR-221/222 in driving β4 integrin expression is also confirmed via mutating the miR-221/222 seed sequence for β4 integrin 3′UTR. Furthermore, we show that these 2 miRNAs are also key breast cancer cell proliferation and invasion regulators, via the post-transcriptional regulation of signal transducer and activator of transcription 5A (STAT5A) and of a disintegrin and metalloprotease-17 (ADAM-17). We further confirm these data by silencing ADAM-17, using a dominant-negative or an activated STAT5A form. miR-221/222-driven β4 integrin, STAT5A, and ADAM-17 did not occur in MCF-10A cells, denoted “normal” breast epithelial cells, indicating that the mechanism is cancer cell-specific.   These results provide the first evidence of a post-transcriptional mechanism that regulates β4 integrin, STAT5A, and ADAM-17 expression, thus controlling breast cancer cell proliferation and invasion. Pre-miR-221/222 use in the aggressive luminal subtype may be a powerful therapeutic anti-cancer strategy

    We test a recently proposed improved lattice-fermion action, the fat link clover action, examining indicators of pathological small-quark-mass lattice artifacts ("exceptional configurations") on quenched lattices of spacing 0.12 fm and studying scaling properties of the light hadron spectrum for lattice spacing a=0.09 and 0.16 fm.     Low-lying fermion modes, topology and light hadrons in quenched QCD

    We explore the properties of low lying eigenmodes of fermions in the quenched approximation of lattice QCD. The fermion action is a recently proposed overlap action and has exact chiral symmetry. We find that chiral zero-eigenvalue modes are localized in space and their positions correlate strongly with the locations (as defined through the density of pure gauge observables) of instantons of the appropriate charge. Nonchiral modes are also localized with peaks which are strongly correlated with the positions of both charges of instantons. These correlations slowly die away as the fermion eigenvalue rises. Correlators made of quark propagators restricted to these modes closely reproduce ordinary hadron correlators at small quark mass in many channels. Our results are in qualitative agreement with the expectations of instanton liquid models.Comment: 21 pages, Revtex, 21 postscript figures. COLO-HEP-45

    By drawing on the most recent archival and historiographical investigations, this volume examines the development of Franciscanism in Pesaro and the life of its local saints during the period between the thirteenth and the fifteenth centuries. Investigating the religious context of Pesaro in the fourteenth and fifteenth centuries implies considering the relations between the mendicant religious orders and the seigniorial power. In this perspective, the Malatestis and the Sforzas strongly endorsed Franciscanism, supported and promoted its foundations, the administration of its convents and the cult of women and men saints within Pesaro’s spirituality
