Al reestudiar la corteza y madera de tallos subterráneos de esta planta fue posible aislar un alcaloide dimérico, la paquistanina, y el lignano siringarresinol. Este lignano parece ser un componente común a varias especies chilenas de Berberís; la paquistanina, por el contrario, es una base del tipo aporfinabencilisoquinolina que hasta ahora sólo habia sido encontrada en especies asiáticas del género

    Monitoring of evoked potentials during spinal cord ischaemia: Experimental evaluation in a rabbit model

    Objectives:somatosensory evoked potentials (SEPs), spinal evoked potentials (Spinal-EPs), and motor-evoked potentials (MEPs) were monitored in a rabbit model of spinal cord ischaemia to evaluate their accuracy and relationship to clinical status.Methods:a modified rabbit spinal cord ischaemia model of infrarenal aortic occlusion for 21 min was employed (30 rabbits). After baseline SEPs, Spinal-EPs, and MEPs were obtained, evoked potentials were recorded continuously during and after clamping of the aorta (30 min). Neurological outcome at 24 h was correlated with evoked potentials, and histopathological findings.Results:fifteen animals became paraplegic. MEPs were always abolished after clamping of the aorta while Spinal-EPs and SEPs remained. The sensory evoked potentials (SEPs and Spinal-EPs) were the least sensitive to spinal cord ischaemia, and their presence had no correlation with the final clinical status (50% of false negatives). This was consistent with histopathological examination that showed damage almost entirely confined to the anterior horn, while the dorsal columns were generally well preserved. High spine MEPs evoked by twitch stimulation was the best predictor of clinical outcome (0% of false negatives, 0% of false positives).Conclusions:SEPs and Spinal-EPs cannot be used as safe monitors of ischaemia of the spinal cord. High spine MEPs evoked by twitch stimulation was the most useful for real-time evaluation of spinal cord ischaemia, and the best predictor of neurologic outcome during reperfusion

    Molecular variants of Grapevine rupestris stem pitting-associated virus infecting grapevines (Vitis spp.) in Brazil.

    Grapevine rupestris stem pitting-associated virus (GRSPaV) is one of the most common viruses of grapevine. It is involved in the graft-transmissible disease rupestris stem pitting of the rugose wood complex. The objective of the research was to perform the molecular characterization of the coat protein (CP) gene of sixteen Brazilian GRSPaV isolates aiming to determine the occurrence of molecular variants (strains) of this virus. Nine grapevine samples were evaluated, from which dsRNA was extracted. Nucleotide sequences were generated by Next generation sequencing (NGS). Fifteen complete sequences of the GRSPaV CP gene were obtained and phylogenetically analyzed. Multiple alignments of the sequences showed identities of nucleotides ranging from 82% to 99%, suggesting high variability among the CPs of Brazilian isolates. The study revealed that genetic variability of GRSPaV comprising three molecular variants is also present in Brazilian grapevine genotypes.Key words: GRSPaV, Foveavirus, strains, lineages, genetic variability. O GRSPaV é um dos vírus mais comuns da videira. Está associado à doença transmissível por enxertia denominada caneluras de rupestris que compõe o complexo do lenho rugoso. O objetivo do trabalho foi realizar a caracterização molecular do gene da proteína capsidial (CP) de 16 isolados brasileiros de GRSPaV visando determinar a ocorrência de variantes moleculares desse vírus. Nove amostras de videira foram avaliadas das quais foi extraído dsRNA. As sequências de nucleotídeos foram geradas pelo sequenciamento de nova geração (NGS). Quinze sequências completas do gene CP de GRSPaV foram obtidas e filogeneticamente analisadas. Os alinhamentos múltiplos entre as sequências mostraram identidades de nucleotídeos variando de 82% a 99%, sugerindo alta variabilidade entre as CPs de isolados brasileiros. O estudo revelou que a variabilidade genética de GRSPaV compreendendo três variantes moleculares também está presente nos genótipos de videira no Brasil.     The Low Velocity Wind from the Circumstellar Matter Around the B9V Star sigma Herculis

    We have obtained FUSE spectra of sigma Her, a nearby binary system, with a main sequence primary, that has a Vega-like infrared excess. We observe absorption in the excited fine structure lines C II* at 1037 A, N II* at 1085 A, and N II** at 1086 A that are blueshifted by as much as ~30 km/sec with respect to the star. Since these features are considerably narrower than the stellar lines and broader than interstellar features, the C II and N II are circumstellar. We suggest that there is a radiatively driven wind, arising from the circumstellar matter, rather than accretion as occurs around beta Pic, because of sigma Her's high luminosity. Assuming that the gas is liberated by collisions between parent bodies at 20 AU, the approximate distance at which blackbody grains are in radiative equilibrium with the star and at which 3-body orbits become unstable, we infer dM/dt ~ 6 * 10^-12 M_{sun}/yr.     Neste estudo, plantas da cv. Royal Gala foram submetidas a PEV (procedimento de eliminação de vírus) por quimioterapia in vitro com ribavirina por 60 dias (tratamento 0 (controle negativo), sem ribavirina; 1, 1 pg/ml; 2, 5 pg/ml; 3, 10 pg/ml de ribavirina), com 4 repetições independentes, aclimatadas e avaliadas quanto à eficácia do PEV por RT-PCR convencional, imediatamente após o tratamento (maio-julho/2010) e após mais 1 ano (julho/2011).     New molecular tools: application of the μAQUA phylochip and concomitant FISH probes to study freshwater pathogens from samples taken along the Tiber River, Italy

    Current knowledge about aquatic pathogens are scarce because bacteria, protozoans, algae and their toxins occur at low concentrations, making them difficult to measure directly or to filter sufficient volumes to facilitate detection. We developed and validated tools to detect pathogens in freshwater systems. To evaluate impacts on water quality, a phylogenetic microarray was developed in the EU project μAQUA to detect simultaneously numerous pathogens and was applied in MicroCoKit, to samples taken from four locations from two seasons for two years along the length of the Tiber River, Italy. The sites ranged from a pristine site near its source to ones contaminated by agricultural, industrial and anthropogenic waste moving downstream to near its mouth. Fifty litres were collected and concentrated using a hollow-fibre ultrafiltration, a rapid method with minimal cell loss to provide a concentrate for downstream analysis. The 60 Da cut-off ensures many organics, such as toxins, will be concentrated for analysis. Aliquots from the concentrate were preserved in TRI-Reagent and total RNA extracted, labelled and hybridised to the phylochip to detect pathogenic bacteria, protozoa and toxic cyanobacteria. The microarray results gave positive signals for all pathogens. Calibration curves enabled us to infer cell concentrations. Cross validation was performed using FISH probes for selected toxic cyanobacteria and hybridised to aliquots taken from the raw water prior to concentration by the hollow fibre filters

    Plant height and hydraulic vulnerability to drought and cold

    Understanding how plants survive drought and cold is increasingly important as plants worldwide experience dieback with drought in moist places and grow taller with warming in cold ones. Crucial in plant climate adaptation are the diameters of water-transporting conduits. Sampling 537 species across climate zones dominated by angiosperms, we find that plant size is unambiguously the main driver of conduit diameter variation. And because taller plants have wider conduits, and wider conduits within species are more vulnerable to conduction-blocking embolisms, taller conspecifics should be more vulnerable than shorter ones, a prediction we confirm with a plantation experiment. As a result, maximum plant size should be short under drought and cold, which cause embolism, or increase if these pressures relax. That conduit diameter and embolism vulnerability are inseparably related to plant size helps explain why factors that interact with conduit diameter, such as drought or warming, are altering plant heights worldwide
