    The Spatial and Governance Dilemma of Small and Medium-Sized Italian Ports (SMPs): Maritime Spatial Planning (MSP) as a Potential Response

    The Italian coast has about 700 ports, which are different in typology, dimension, role, and ownership. Historically, this has led to the significant fragmentation of governance and space and a lack of cooperation that ports and cities still experience today. Among all ports, small and medium-sized ports (SMPs), such as marinas, small touristic harbors, and moorings, are the most affected. Unlike the main ports, where spatial and strategic regulation planning fall under the port authority’s responsibilities, SMPs are a combination of public and private management and are, therefore, excluded from national and regional planning and larger strategies. Improving SMPs’ cooperation at the regional level can drive more effective sustainable management among related activities (tourism and the fishing sector) and reduce pressures on the land–sea interaction (LSI). In filling the gaps, this article challenges the existing legal framework, planning tools, approaches, and initiatives and may pave the way to establishing a better-integrated national governance for SMPs. In conclusion, this paper identifies two main opportunities that can support the steady establishment of governance and the systematic harmonized development of these SMPs. The first one is offered by maritime spatial planning (MSP) as a strategic and legal tool whereby SMPs are recognized and, if financially supported, could find incentives and measures for their development. The second one is through European projects, programs, and initiatives such as Framesport as drivers in establishing a common ground among public and private interests and as a cooperation engine at a local scale

    The effects of chronic lifelong activation of the AHR pathway by industrial chemical pollutants on female human reproduction

    Environmental chemicals, such as heavy metals, affect female reproductive function. A biological sensor of the signals of many toxic chemical compounds seems to be the aryl hydrocarbon receptor (AHR). Previous studies demonstrated the environmental of heavy metals in Taranto city (Italy), an area that has been influenced by anthropogenic factors such as industrial activities and waste treatments since 1986. However, the impact of these elements on female fertility in this geographic area has never been analyzed. Thus, in the present study, we evaluated the AHR pathway, sex steroid receptor pattern and apoptotic process in granulosa cells (GCs) retrieved from 30 women, born and living in Taranto, and 30 women who are living in non-contaminated areas (control group), who were undergoing in vitro fertilization (IVF) protocol. In follicular fluids (FFs) of both groups the toxic and essential heavy metals, such as chromiun (Cr), Manganese (Mn), iron (Fe), cobalt (Co), nickel (Ni), copper (Cu), zinc (Zn), cadmium (Cd) and lead (Pb), were also analyzed. Higher levels of Cr, Fe, Zn and Pb were found in the FFs of the women from Taranto as compared to the control group, as were the levels of AHR and AHR-dependent cytochrome P450 1A1 and 1B1; while CYP19A1 expression was decreased. The anti-apoptotic process found in the GCs of women fromTaranto was associated with the highest levels of progesterone receptor membrane component 1(PGRMC1), a novel progesterone receptor, the expression of which is subjected to AHR activated by its highest affinity ligands (e.g., dioxins) or indirectly by other environmental pollutants, such as heavy metals. In conclusion, decreased production of estradiol and decreased number of retrieved mature oocytes found in women from Taranto could be due to chronic exposure to heavy metals, in particular to Cr and Pb

    MicroRNA co-expression networks exhibit increased complexity in pancreatic ductal compared to Vater’s papilla adenocarcinoma

    iRNA expression abnormalities in adenocarcinoma arising from pancreatic ductal system (PDAC) and Vater’s papilla (PVAC) could be associated with distinctive pathologic features and clinical cancer behaviours. Our previous miRNA expression profiling data on PDAC (n=9) and PVAC (n=4) were revaluated to define differences/ similarities in miRNA expression patterns. Afterwards, in order to uncover target genes and core signalling pathways regulated by specific miRNAs in these two tumour entities, miRNA interaction networks were wired for each tumour entity, and experimentally validated target genes underwent pathways enrichment analysis. One hundred and one miRNAs were altered, mainly over-expressed, in PDAC samples. Twenty-six miRNAs were deregulated in PVAC samples, where more miRNAs were down-expressed in tumours compared to normal tissues. Four miRNAs were significantly altered in both subgroups of patients, while 27 miRNAs were differentially expressed between PDAC and PVAC. Although miRNA interaction networks were more complex and dense in PDAC than in PVAC, pathways enrichment analysis uncovered a functional overlapping between PDAC and PVAC. However, shared signalling events were influenced by different miRNA and/or genes in the two tumour entities. Overall, specific miRNA expression patterns were involved in the regulation of a limited core signalling pathways in the biology landscape of PDAC and PVAC

    Cytotoxicity of an innovative pressurised cyclic solid–liquid (Pcsl) extract from artemisia annua

    Therapeutic treatments with Artemisia annua have a long-established tradition in various diseases due to its antibacterial, antioxidant, antiviral, anti-malaria and anti-cancer effects. However, in relation to the latter, virtually all reports focused on toxic effects of A. annua extracts were obtained mostly through conventional maceration methods. In the present study, an innovative extraction procedure from A. annua, based on pressurised cyclic solid–liquid (PCSL) extraction, resulted in the production of a new phytocomplex with enhanced anti-cancer properties. This extraction procedure generated a pressure gradient due to compressions and following decompressions, allowing to directly perform the extraction without any maceration. The toxic effects of A. annua PCSL extract were tested on different cells, including three cancer cell lines. The results of this study clearly indicate that the exposure of human, murine and canine cancer cells to serial dilutions of PCSL extract resulted in higher toxicity and stronger propensity to induce apoptosis than that detected by subjecting the same cells to Artemisia extracts obtained through canonical extraction by maceration. Collected data suggest that PCSL extract of A. annua could be a promising and economic new therapeutic tool to treat human and animal tumours

    In the wake of the ongoing mass mortality events: Co-occurrence of Mycobacterium, Haplosporidium and other pathogens in Pinna nobilis collected in Italy and Spain (Mediterranean Sea)

    Following the Mass Mortality Events (MMEs) of the pen shell P. nobilis in Campania region and Sicily, a survey of moribund P. nobilis specimens was also conducted in other Italian regions (Campania, Tuscany, Sardinia, and Apulia) and Spain (Catalunya). Histopathological and molecular examination of 27 specimens of P. nobils revealed different types of pathogens associated with tissue lesions, morbidity and mortality. Presence of Mycobacterium, Vibrio species, Haplosporidium pinnae and Perkinsus sp. were detected, differently distributed into the areas. The Mycobacterium sp., previously reported in Campania and Sicily samples, was observed in all the analyzed areas and individuals, associated to systemic inflammatory lesions. In Spain, H. pinnae was observed in 36% of cases, always associated to the Mycobacterium sp. Molecular study using hsp65 genes and Internal Transcriber Spacer ITS support that a new species of Mycobacteria is infecting P. nobilis, close to M. triplex and belonging to the group of M. simiae complex with M. sherrisi. Presence of Perkinsus spp. resembling P. mediterraneus was observed in 2 out of 13 Italian individuals whose presence should be addressed as potential risk for shellfish aquaculture of the area. Vibrio spp. were also detected in some case. The preliminary results of this study suggest that Mycobacterium sp., Vibrio spp., H. pinnae and Perkinsus sp. cooperate to disease pathogenesis, being Mycobacterium and Haplosporidium most of the time involved. Vigilant inspection of those areas where MME is now starting, along with continuous systematic surveys, are crucial to define the spatiotemporal progress of mortality and the role of every single pathogen in the disease outcome.info:eu-repo/semantics/publishedVersio

    Mirna Expression Profiles Identify Drivers in Colorectal and Pancreatic Cancers

    Altered expression of microRNAs (miRNAs) hallmarks many cancer types. The study of the associations of miRNA expression profile and cancer phenotype could help identify the links between deregulation of miRNA expression and oncogenic pathways.Expression profiling of 866 human miRNAs in 19 colorectal and 17 pancreatic cancers and in matched adjacent normal tissues was investigated. Classical paired t-test and random forest analyses were applied to identify miRNAs associated with tissue-specific tumors.     Designing and implementing a multi-scalar approach to Maritime Spatial Planning: The case study of Italy

    The Italian coastal and marine space includes areas with remarkable differences in terms of oceanographic characteristics, maritime uses, natural habitats, species distribution, landscape and cultural heritage. In Italy, coastal and marine management competencies are shared among national, regional, and for some aspects even local authorities. This geographic heterogeneity and governance complexity required the adoption of a multiscalar approach to Maritime Spatial Planning (MSP). Such an approach aims at implementing decision-making and spatial planning at multiple and nested scales. In the case of Italy, the multi-scalar approach included the definition of national guidelines and the development of three maritime spatial (MS) plans, one for each maritime area (Adriatic, Ionian and Central Mediterranean, and Tyrrhenian and Western Mediterranean), including subareas and nested planning units. The development of the MS plans involved competent Ministries, the coastal Regions and several researchers. Based on the description of the adopted six-phase methodology and the exemplification of results of the Italian MSP process, this paper discusses the most relevant features and common challenges of multi-scalar MSP (i.e. co-planning, vertical and horizontal integration, multi-level governance, scalability, flexibility, integration of data and knowledge with different resolution, multi-scalar stakeholder engagement). Finally, the paper reflects on some novel aspects of the adopted multi-scalar approach and identifies actions to grant efficacy to this approach during the next phases of the Italian MSP proces
