    Molecular characterization of paecilomyces fumosoroseus (deuteromycotina: hyphomycetes) isolates.

    ABSTRACT ITS and RAPD analyses were used to investigate molecular variations within samples of Paecilomyces isolates and to resolve five morphologically atypical isolates resembling P. fumosorosus, obtained from whitefly in Northern Paraná State. The ITS4-ITS5 amplicon was 700 base pairs (bp) long in all isolates. The five isolates of Paecilomyces not assigned to species produced restriction profiles identical to all the reference strains of P. fumosoroseus. The extent of fingerprint variability observed by RAPD was sufficient to discriminate all the isolates. The genetic similarity among unidentified isolates and strains of P. fumosoroseus was even higher than that observed among reference strains of this species, allowing us to conclude that isolates CNPso-P77, CNPso-P78, CNPso-P80, CNPso-P85 and CNPso-P91 are P. fumosoroseus.     Análise proteômica do estresse hídrico em cafeeiro.

    Déficit hídrico é um dos fatores ambientais mais importantes para a diminuição da produtividade do cafeeiro, tanto no Brasil quanto em outros países produtores. O estabelecimento de estratégias para obtenção de cultivares tolerantes ao estresse hídrico depende da compreensão das respostas biológicas ao nível genético, molecular e bioquímico. Para estudar a resposta ao estresse hídrico no gênero Coffea, foi estabelecida uma rede de pesquisa em proteômica (PROTEOPAR ? Programa Proteoma do Paraná) constituída de oito laboratórios. Quatro genótipos de Coffea, com diferentes respostas fisiológicas à seca, foram analisados: C. canephora (Clone 14, tolerante e Clone 109A, sensível) e C. arabica (BA10, tolerante e Geisha, sensível). Proteínas foram extraídas de folhas e raízes de plantas (18 meses de idade) mantidas em casa-de-vegetação, utilizando-se o método de SDS-Fenol modificado. Os regimes hídricos constituíram-se dos seguintes tratamentos: controle irrigado, estresse hídrico severo (-4,0 MPa de potencial de água) e recuperação pós-estresse (36 horas após irrigação). Os extratos protéicos foram distribuídos aos laboratórios do PROTEOPAR para obtenção e análise do padrão de expressão protéica diferencial via eletroforese bidimensional. O método de identificação de proteínas PMF (?Peptide mass fingerprinting?) foi aplicado utilizando-se um espectrômetro de massa (MS) do tipo MALDI-TOF e comparando os espectros de digestão tríptica contra bancos de dados baseados em ESTs de Coffea

    Extension of the core map of common bean with EST-SSR, RGA, AFLP, and putative functional markers

    Microsatellites and gene-derived markers are still underrepresented in the core molecular linkage map of common bean compared to other types of markers. In order to increase the density of the core map, a set of new markers were developed and mapped onto the RIL population derived from the ‘BAT93’ × ‘Jalo EEP558’ cross. The EST-SSR markers were first characterized using a set of 24 bean inbred lines. On average, the polymorphism information content was 0.40 and the mean number of alleles per locus was 2.7. In addition, AFLP and RGA markers based on the NBS-profiling method were developed and a subset of the mapped RGA was sequenced. With the integration of 282 new markers into the common bean core map, we were able to place markers with putative known function in some existing gaps including regions with QTL for resistance to anthracnose and rust. The distribution of the markers over 11 linkage groups is discussed and a newer version of the common bean core linkage map is proposed

    Genome of Herbaspirillum seropedicae Strain SmR1, a Specialized Diazotrophic Endophyte of Tropical Grasses

    The molecular mechanisms of plant recognition, colonization, and nutrient exchange between diazotrophic endophytes and plants are scarcely known. Herbaspirillum seropedicae is an endophytic bacterium capable of colonizing intercellular spaces of grasses such as rice and sugar cane. The genome of H. seropedicae strain SmR1 was sequenced and annotated by The Paraná State Genome Programme—GENOPAR. The genome is composed of a circular chromosome of 5,513,887 bp and contains a total of 4,804 genes. The genome sequence revealed that H. seropedicae is a highly versatile microorganism with capacity to metabolize a wide range of carbon and nitrogen sources and with possession of four distinct terminal oxidases. The genome contains a multitude of protein secretion systems, including type I, type II, type III, type V, and type VI secretion systems, and type IV pili, suggesting a high potential to interact with host plants. H. seropedicae is able to synthesize indole acetic acid as reflected by the four IAA biosynthetic pathways present. A gene coding for ACC deaminase, which may be involved in modulating the associated plant ethylene-signaling pathway, is also present. Genes for hemagglutinins/hemolysins/adhesins were found and may play a role in plant cell surface adhesion. These features may endow H. seropedicae with the ability to establish an endophytic life-style in a large number of plant species
