    DUAL-USE RESEARCH AND SYNTHETIC BIOLOGY: AN INVITATION INTO PANDORA’S BOX

    The rapid advancement of synthetic biology along with the rapid emergence of technology has, and continues to provide, widespread benefit to humanity. While this advancement has been responsible for advanced medical treatments as well as environmental sustainability, there is inherent risk associated with some research attributed to synthetic biology. The benefit/risk trade-off, however, has raised uncertainty in both the scientific community and among world leaders and policy makers. The effects of the COVID-19 pandemic have increased concerns, globally, about dual-use and gain-of-function research. While benefits may ensue, unpredictable outcomes and unintended consequences could have a disastrous impact on the world. The potential harm an engineered biological organism could have on the global community presents a significant threat to the security of the United States. While some progress, in terms of processes and procedures, has been made to reduce potential harm, there is still further need for uniform regulation and oversight. An organism that has been engineered utilizing synthetic biology must not introduce a biological risk that produces a global threat.Civilian, Phoenix Fire DepartmentApproved for public release.     Sediment patterns near a model patch of reedy emergent vegetation

    This laboratory study describes the sediment patterns formed in a sand bed around circular patches of rigid vertical cylinders, representing a patch of reedy emergent vegetation. The patch diameter was much smaller than the channel width. Two patch densities (solid volume fraction 3% and 10%) and two patch diameters (22 and 10 cm) were considered. For flows above the threshold of sediment motion, patterns of sediment erosion and deposition were observed around and within the patch. Scouring within the patch was positively correlated with turbulent kinetic energy in the patch. For sparse patches, sediment scoured from within the patch was mostly deposited within one patch diameter downstream of the patch. For dense patches, which experience greater flow diversion, sediment scoured from the patch was carried farther downstream before deposition along the patch centerline. Differences between the sparse and dense patch patterns of deposition are explained in the context of flow diversion and wake structure, which are related to a nondimensional flow blockage parameter.     A Digital Neuromorphic Architecture Efficiently Facilitating Complex Synaptic Response Functions Applied to Liquid State Machines

    Information in neural networks is represented as weighted connections, or synapses, between neurons. This poses a problem as the primary computational bottleneck for neural networks is the vector-matrix multiply when inputs are multiplied by the neural network weights. Conventional processing architectures are not well suited for simulating neural networks, often requiring large amounts of energy and time. Additionally, synapses in biological neural networks are not binary connections, but exhibit a nonlinear response function as neurotransmitters are emitted and diffuse between neurons. Inspired by neuroscience principles, we present a digital neuromorphic architecture, the Spiking Temporal Processing Unit (STPU), capable of modeling arbitrary complex synaptic response functions without requiring additional hardware components. We consider the paradigm of spiking neurons with temporally coded information as opposed to non-spiking rate coded neurons used in most neural networks. In this paradigm we examine liquid state machines applied to speech recognition and show how a liquid state machine with temporal dynamics maps onto the STPU-demonstrating the flexibility and efficiency of the STPU for instantiating neural algorithms.Comment: 8 pages, 4 Figures, Preprint of 2017 IJCN

    Phytosanitary irradiation for export of fresh produce: commercial adoption in Hawaii and current issues

    Hawaii is a pioneer in the use of phytosanitary irradiation. Irradiation is an approved treatment to control quarantine insect pests in 17 fruits and 7 vegetables for export from Hawaii to the US mainland. The commercial X-ray irradiation facility, Hawaii Pride LLC, has been shipping tropical fruits and vegetables to the US mainland using irradiation since 2000. Hawaiian purple sweet potato is the highest volume product with annual exports of more than 12 million lbs (5,500 tonnes). The advent of generic radiation treatments for tephritid fruit flies (150 Gy) and other insects (400 Gy) has accelerated commodity export approvals and facilitated adoption by foreign trading partners. Current impediments to wider adoption include the labeling requirement, the 1 kGy limit for fresh horticultural products, and non-acceptance of phytosanitary irradiation in Japan and the European Union. USDA Animal and Plant Health Inspection Service (APHIS) has restricted the use of modified atmosphere packaging (MAP) for irradiated fresh fruits exported to the US due to possible increased radiotolerance in the target insects. Data are presented for response of melon fly in papaya to several types of MAP with radiation treatment at 45–65 Gy. Low oxygen atmospheres can increase insect radio-tolerance, but the effect is dependent on oxygen concentration and radiation dose

    The Development of Orphan Drugs; A Financial and Ethical Decision

    This paper explores the decision faced by a firm to invest in an orphan drug development project. Two primary areas of concern are considered: financial and ethical. In order to properly understand these two areas, the paper first summarizes the current development landscape for non-orphan and orphan drugs. Once the basic development structure is established, a discussion regarding the differences in the Net Present Value equation for a non-orphan and orphan product may occur. Once the differences in the financial decision are established, the paper will discuss the ethical considerations surrounding drug development and drug pricing. The combination of the financial model and the ethical guidelines for drug pricing form the argument for an increase in social corporate responsibility in the drug development industry to increase treatment accessibility for patients of rare diseases
