    Osteochondrosis lesions of the lateral trochlear ridge of the distal femur in four ponies

    Lesions of the lateral trochlear ridge (LTR) of the distal femur were investigated in four pony or pony cross horses. The animals were all geldings and were six to 15 months of age. Lesions were bilateral in three ponies and unilateral in one. Femoropatellar joint effusion and lameness were present in two ponies; clinical signs were absent in the others. The proximal LTR was affected in all four animals. The radiographic appearance of the lesions was a subchondral defect containing mineralised bodies. Arthroscopic and postmortem examination findings included an osteochondral flap, a fissured or irregular articular surface and a smooth surface overlying focally thickened cartilage that extended into subchondral bone. Thickened articular cartilage was a histological feature of all the lesions. Among the other histological features, the most common were chondronecrosis, chondrocyte clusters, phenotypically abnormal chondrocytes, horizontal fissures at the osteochondral junction and retained blood vessels. The signalment of the four ponies, their clinical signs and the pathological features of their lesions were consistent with osteochondrosis of the LTR in horses. The use of multiple criteria was considered to be important in making a specific diagnosis

    Peripheral mechanisms of arthritic pain: a proposal to leverage large animals for in vitro studies

    Pain arising from musculoskeletal disorders such as arthritis is one of the leading causes of disability. Whereas the past 20-years has seen an increase in targeted therapies for rheumatoid arthritis (RA), other arthritis conditions, especially osteoarthritis, remain poorly treated. Although modulation of central pain pathways occurs in chronic arthritis, multiple lines of evidence indicate that peripherally driven pain is important in arthritic pain. To understand the peripheral mechanisms of arthritic pain, various in vitro and in vivo models have been developed, largely in rodents. Although rodent models provide numerous advantages for studying arthritis pathogenesis and treatment, the anatomy and biomechanics of rodent joints differ considerably to those of humans. By contrast, the anatomy and biomechanics of joints in larger animals, such as dogs, show greater similarity to human joints and thus studying them can provide novel insight for arthritis research. The purpose of this article is firstly to review models of arthritis and behavioral outcomes commonly used in large animals. Secondly, we review the existing in vitro models and assays used to study arthritic pain, primarily in rodents, and discuss the potential for adopting these strategies, as well as likely limitations, in large animals. We believe that exploring peripheral mechanisms of arthritic pain in vitro in large animals has the potential to reduce the veterinary burden of arthritis in commonly afflicted species like dogs, as well as to improve translatability of pain research into the clinic

    Identificação dos níveis séricos do fator de crescimento tipo insulina 1 em potros com osteocondrose

    A osteocondrose é uma das doenças ortopédicas do desenvolvimento mais comuns em cavalos, e deve ser encarada como uma desordem multifatorial. Caracteriza-se por ser uma doença dinâmica, onde o processo de reparo inicia-se quase que imediatamente após a formação da lesão osteocondral. A intensidade e eficácia deste reparo são fortemente relacionados à idade, e estão associados a reposição dos componentes da matriz cartilagínea. Uma vez que o fator de crescimento tipo insulina-1 (IGF-1) age regulando o crescimento da cartilagem articular, este estudo objetivou descrever os níveis séricos de IGF-I, insulina e glicose em potros Puro Sangue Lusitano hígidos ou com osteocondrose, desde o nascimento até 18 meses de idade. Dos potros acompanhados, 76,08% apresentaram sinais radiográficos de osteocondrose com um mês de idade, porém somente 16,2% mantiveram as lesões aos 18 meses. As concentrações de IGF-1 alcançaram picos entre 14 e 16 meses, equivalente ao início da puberdade. As concentrações de IGF-1 nos potros com osteocondrose foram menores que potros sadios, aos 2 e 5 meses (P<0,05), e os de insulina aos 2 meses (P<0,05). Em criatórios com alta ocorrência de osteocondrose recomenda-se que as mensurações dos níveis séricos de IGF-1 e insulina ocorram durante os primeiros 6 meses de idade, para que haja tempo hábil de se estabelecer medidas profiláticas e de tratamento
