    3D mapping of the SPRY2 domain of ryanodine receptor 1 by single-particle Cryo-EM

    The type 1 skeletal muscle ryanodine receptor (RyR1) is principally responsible for Ca(2+) release from the sarcoplasmic reticulum and for the subsequent muscle contraction. The RyR1 contains three SPRY domains. SPRY domains are generally known to mediate protein-protein interactions, however the location of the three SPRY domains in the 3D structure of the RyR1 is not known. Combining immunolabeling and single-particle cryo-electron microscopy we have mapped the SPRY2 domain (S1085-V1208) in the 3D structure of RyR1 using three different antibodies against the SPRY2 domain. Two obstacles for the image processing procedure; limited amount of data and signal dilution introduced by the multiple orientations of the antibody bound in the tetrameric RyR1, were overcome by modifying the 3D reconstruction scheme. This approach enabled us to ascertain that the three antibodies bind to the same region, to obtain a 3D reconstruction of RyR1 with the antibody bound, and to map SPRY2 to the periphery of the cytoplasmic domain of RyR1.     Uniform accounting system for school cities and towns: a handbook of instructions

    The State Board of Accounts in collaboration with the State Department of Public Instruction, co-operating with an advisory committee of the Indiana City and Town Superintendents\u27 Association, have worked out in detail an accouting system designed to provide an adequate plan for Fund and Functional Accounting. Four ideas were kept constantly in mind while the forms constituting the system and the directions covering them were being drafted. (1) The system should be sound from an accounting point of view. (2) The system should be simple enough to enable clerks in the office of the small town superintendent\u27s or secretary\u27s office to keep it. (3) The system should be complete enough to yield desirable results. (4) The system should be flexible enough to meet adequately the needs of the largest school corporation as well as the smallest.     Two center multipole expansion method: application to macromolecular systems

    We propose a new theoretical method for the calculation of the interaction energy between macromolecular systems at large distances. The method provides a linear scaling of the computing time with the system size and is considered as an alternative to the well known fast multipole method.     Biophysical Measurements of Cells, Microtubules, and DNA with an Atomic Force Microscope

    Atomic force microscopes (AFMs) are ubiquitous in research laboratories and have recently been priced for use in teaching laboratories. Here we review several AFM platforms (Dimension 3000 by Digital Instruments, EasyScan2 by Nanosurf, ezAFM by Nanomagnetics, and TKAFM by Thorlabs) and describe various biophysical experiments that could be done in the teaching laboratory using these instruments. In particular, we focus on experiments that image biological materials and quantify biophysical parameters: 1) imaging cells to determine membrane tension, 2) imaging microtubules to determine their persistence length, 3) imaging the random walk of DNA molecules to determine their contour length, and 4) imaging stretched DNA molecules to measure the tensional force.Comment: 29 page preprint, 7 figures, 1 tabl

    Cardiac rehabilitation and psychological well-being

    The aim of this chapter is to address psychological issues associated with effective multidimensional cardiac rehabilitation programmes. Cardiac rehabilitation is defined as: “the sum of activities required to influence favourably the underlying cause of the disease, as well as the best possible, physical, mental and social conditions, so that they (people) may, by their own efforts preserve or resume when lost, as normal a place as possible in the community. Rehabilitation cannot be regarded as an isolated form or stage of therapy but must be integrated within secondary prevention services of which it forms only one facet” (World Health Organisation, 1993). The chapter will: discuss the impact of CHD in the UK, provide an overview of the Government strategy for reducing the burden of cardiac disease and disability, and review quantitative evidence discussing the effectiveness of cardiac rehabilitation on the mental health and well-being of cardiac patients, with particular reference to anxiety and depression states. The final part of this chapter presents results of a qualitative study, previously reported in Hudson, Board, and, Lavallee (2001) that examined the psychosocial impact of cardiac disease and rehabilitation for patients attending one cardiac rehabilitation scheme in England

    Tourist Shoppers’ Evaluation of Retail Service: A Study of Cross-Border versus International Outshoppers

    This article extends the concept of customer perceived value (CPV) to the tourist outshopping context and explores the differences in antecedents and outcomes of CPV between cross-border and international outshoppers. A large-scale field survey in Hong Kong with cross-border outshoppers from mainland China and international shoppers from four Western countries (Australia, Canada, the United Kingdom, and the United States) shows that perceived product quality, risk, and value for money have a stronger effect on CPV for cross-border outshoppers, and employee service quality and lifestyle congruence for international outshoppers. CPV also has a stronger positive effect on satisfaction, word of mouth, and repeat purchase intentions for cross-border outshoppers, whereas satisfaction has a stronger positive impact on word of mouth and repeat purchase intentions for international outshoppers. We discuss the conceptual contribution and managerial implications of our findings for international retailers, researchers, and tourism organizations
