    Partitioning optimization of proteins from Zea mays malt in ATPS PEG 6000/CaCl2

    This work aimed to establish the relationship between the compositions and pH of ATPS PEG 6000/CaCl2 and the proteins partition from maize malt and also to simplify the process optimization in ATPS for a statistical model, established by response surface methodology (RSM). Results showed that these were no influence of pH on the phase diagrams and on the composition of tie line length of PEG 6000/CaCl2 ATPS. SRM analyses showed that elevated pH and larger tie line length were the best conditions for recovering of maize malt proteins. The maximum partition coefficient by PEG 6000/CaCl2 ATPS was about 4.2 and was achieved in ATPS in a single purification step. The theoretical maximum partition coefficient was between 4.1-4.3. The process was very suitable for continuous aqueous two phase purification due to the stability of proteins (e.g. alpha and beta-amylases) and could increase their content into middle.50355756

    Coordination of microtubule and microfilament dynamics by Drosophila Rho1, Spire, and Cappuccino

    The actin nucleation factors Spire and Cappuccino regulate the onset of ooplasmic streaming in Drosophila1-5. Although this streaming event is microtubule-based, actin assembly is required for its timing. It is not understood how the interaction of microtubules and microfilaments is mediated in this context. Here we demonstrate that Cappuccino and Spire have microtubule and microfilament crosslinking activity. The spire locus encodes several distinct protein isoforms (SpireA, SpireC, and SpireD). SpireD was recently shown to nucleate actin, but the activity of the other isoforms has not been addressed. We find that SpireD does not have crosslinking activity, while SpireC is a potent crosslinker. We show that SpireD binds to Cappuccino and inhibits Factin/ microtubule crosslinking, and activated Rho1 abolishes this inhibition, establishing a mechanistic basis for the regulation of Capu and Spire activity. We propose that Rho1, cappuccino and spire are elements of a conserved developmental cassette that is capable of directly mediating crosstalk between microtubules and microfilaments

    Radionuclide Imaging of Viable Myocardium: Is it Underutilized?

    Coronary artery disease is the major cause of heart failure in North America. Viability assessment is important as it aims to identify patients who stand to benefit from coronary revascularization. Radionuclide modalities currently used in the assessment of viability include 201Tl SPECT, 99mTc-based SPECT imaging, and 18F-fluorodexoyglucose (18F-FDG)-PET imaging. Different advances have been made in the last year to improve the sensitivity and specificity of these modalities. In addition, the optimum amount of viable (yet dysfunctional) myocardium is important to identify in patients, as a risk–benefit ratio must be considered. Patients with predominantly viable/hibernating myocardium can benefit from revascularization from a mortality and morbidity standpoint. However, in patients with minimal viability (predominantly scarred myocardium), revascularization risk may certainly be too high to justify revascularization without expected benefit. Understanding different radionuclide modalities and new developments in the assessment of viability in ischemic heart failure patients is the focus of this discussion

    Formin homology 2 domains occur in multiple contexts in angiosperms

    BACKGROUND: Involvement of conservative molecular modules and cellular mechanisms in the widely diversified processes of eukaryotic cell morphogenesis leads to the intriguing question: how do similar proteins contribute to dissimilar morphogenetic outputs. Formins (FH2 proteins) play a central part in the control of actin organization and dynamics, providing a good example of evolutionarily versatile use of a conserved protein domain in the context of a variety of lineage-specific structural and signalling interactions. RESULTS: In order to identify possible plant-specific sequence features within the FH2 protein family, we performed a detailed analysis of angiosperm formin-related sequences available in public databases, with particular focus on the complete Arabidopsis genome and the nearly finished rice genome sequence. This has led to revision of the current annotation of half of the 22 Arabidopsis formin-related genes. Comparative analysis of the two plant genomes revealed a good conservation of the previously described two subfamilies of plant formins (Class I and Class II), as well as several subfamilies within them that appear to predate the separation of monocot and dicot plants. Moreover, a number of plant Class II formins share an additional conserved domain, related to the protein phosphatase/tensin/auxilin fold. However, considerable inter-species variability sets limits to generalization of any functional conclusions reached on a single species such as Arabidopsis. CONCLUSIONS: The plant-specific domain context of the conserved FH2 domain, as well as plant-specific features of the domain itself, may reflect distinct functional requirements in plant cells. The variability of formin structures found in plants far exceeds that known from both fungi and metazoans, suggesting a possible contribution of FH2 proteins in the evolution of the plant type of multicellularity

    Sotaque e telejornalismo: representações de comunicadores de mídia nordestinos

    RESUMO: Objetivo: descrever o conteúdo das representações sociais do comunicador de mídia nordestino acerca do seu sotaque. Métodos: a coleta de dados foi realizada em duas etapas. Na primeira, foi empregada a técnica de associação livre, na qual 50 universitários apresentaram o que pensavam diante da palavra "sotaque". Na segunda, por meio do Procedimento de Classificações Múltiplas (PCM), 25 comunicadores atuantes na Região Metropolitana do Recife classificaram as 15 palavras mais associadas pelos estudantes de duas maneiras, classificação livre e dirigida. Na classificação livre, os comunicadores agruparam as palavras segundo critérios por eles estabelecidos, enquanto que na classificação dirigida, as palavras foram classificadas de acordo com o grau de associação com o termo MEU SOTAQUE. A análise foi realizada utilizando-se métodos estatísticos multidimensionais, que permitiram construir a estruturação do campo das Representações Sociais. Resultados: três regiões delimitadas expressam a representação dos sujeitos sobre sotaque: Conceito, Identidade e Espaço. Na primeira, há seis categorias (engraçado, diferente, arrastado, matuto, preconceito, oxente), na segunda, as palavras são identidade, característica, língua, fala e cultura, próximas ao termo meu sotaque e, na terceira, região, regionalismo, nordeste e localidade. Identidade foi o termo mais relacionado ao termo MEU SOTAQUE, enquanto que os itens menos relacionados são os presentes na região conceito, exceto pelo termo oxente. Conclusão: o sotaque nordestino é tido como uma marca da identidade e cultura da região Nordeste. Os comunicadores suavizam características do falar nordestino durante o exercício profissional para se adequar ao padrão preconizado pelo mercado de trabalho sem perder a sua identidade

    A polarimetrically oriented X-ray stare at the accreting pulsar EXO 2030+375

    Accreting X-ray pulsars (XRPs) are presumed to be ideal targets for polarization measurements, as their high magnetic field strength is expected to polarize the emission up to a polarization degree of 80%. However, such expectations are being challenged by recent observations of XRPs with the Imaging X-ray Polarimeter Explorer (IXPE). Here, we report on the results of yet another XRP, namely, EXO 2030+375, observed with IXPE and contemporarily monitored with Insight-HXMT and SRG/ART-XC. In line with recent results obtained with IXPE for similar sources, an analysis of the EXO 2030+375 data returns a low polarization degree of 0%- 3% in the phase-averaged study and a variation in the range of 2%- 7% in the phase-resolved study. Using the rotating vector model, we constrained the geometry of the system and obtained a value of 60 for the magnetic obliquity. When considering the estimated pulsar inclination of 130, this also indicates that the magnetic axis swings close to the observera's line of sight. Our joint polarimetric, spectral, and timing analyses hint toward a complex accreting geometry, whereby magnetic multipoles with an asymmetric topology and gravitational light bending significantly affect the behavior of the observed source

    Polarization Properties of the Weakly Magnetized Neutron Star X-Ray Binary GS 1826-238 in the High Soft State

    The launch of the Imaging X-ray Polarimetry Explorer (IXPE) on 2021 December 9 has opened a new window in X-ray astronomy. We report here the results of the first IXPE observation of a weakly magnetized neutron star, GS 1826−238, performed on 2022 March 29-31 when the source was in a high soft state. An upper limit (99.73% confidence level) of 1.3% for the linear polarization degree is obtained over the IXPE 2-8 keV energy range. Coordinated INTEGRAL and NICER observations were carried out simultaneously with IXPE. The spectral parameters obtained from the fits to the broadband spectrum were used as inputs for Monte Carlo simulations considering different possible geometries of the X-ray emitting region. Comparing the IXPE upper limit with these simulations, we can put constraints on the geometry and inclination angle of GS 1826-238

    The detection of polarized X-ray emission from the magnetar 1E 2259+586

    We report on IXPE, NICER, and XMM–Newton observations of the magnetar 1E 2259+586. We find that the source is significantly polarized at about or above 20 per cent for all phases except for the secondary peak where it is more weakly polarized. The polarization degree is strongest during the primary minimum which is also the phase where an absorption feature has been identified previously. The polarization angle of the photons are consistent with a rotating vector model with a mode switch between the primary minimum and the rest of the rotation of the neutron star. We propose a scenario in which the emission at the source is weakly polarized (as in a condensed surface) and, as the radiation passes through a plasma arch, resonant cyclotron scattering off of protons produces the observed polarized radiation. This confirms the magnetar nature of the source with a surface field greater than about 1015 G

    Discovery of strongly variable X-ray polarization in the neutron star low-mass X-ray binary transient XTE J1701-462

    CONTEXT: After about 16 years since its first outburst, the transient neutron star low-mass X-ray binary XTE J1701−462 turned on again in September 2022, allowing for the first study of its X-ray polarimetric characteristics by a dedicated observing program with the Imaging X-ray Polarimeter Explorer (IXPE). AIMS: Polarimetric studies of XTE J1701−462 have been expected to improve our understanding of accreting weakly magnetized neutron stars, in particular, the physics and the geometry of the hot inner regions close to the compact object. METHOD: The IXPE data of two triggered observations were analyzed using time-resolved spectroscopic and polarimetric techniques, following the source along its Z-track of the color–color diagram. RESULTS: During the first pointing on 2022 September 29, an average 2–8 keV polarization degree of (4.6 ± 0.4)% was measured, the highest value found up to now for this class of sources. Conversely, only a ∼0.6% average degree was obtained during the second pointing ten days later. CONCLUSIONS: The polarimetric signal appears to be strictly related to the higher energy blackbody component associated with the boundary layer (BL) emission and its reflection from the inner accretion disk, and it is as strong as 6.1% and 1.2% (> 95% significant) above 3–4 keV for the two measurements, respectively. The variable polarimetric signal is apparently related to the spectral characteristics of XTE J1701−462, which is the strongest when the source was in the horizontal branch of its Z-track and the weakest in the normal branch. These IXPE results provide new important observational constraints on the physical models and geometry of the Z-sources. Here, we discuss the possible reasons for the presence of strong and variable polarization among these sources

    The X-ray polarization of the Seyfert 1 galaxy IC 4329A

    We present an X-ray spectro-polarimetric analysis of the bright Seyfert galaxy IC 4329A. The Imaging X-ray Polarimetry Explorer (IXPE) observed the source for ∼500 ks, supported by XMM–Newton (∼60 ks) and NuSTAR (∼80 ks) exposures. We detect polarization in the 2–8 keV band with 2.97σ confidence. We report a polarization degree of 3.3 ± 1.1 per cent and a polarization angle of 78° ± 10° (errors are 1σ confidence). The X-ray polarization is consistent with being aligned with the radio jet, albeit partially due to large uncertainties on the radio position angle. We jointly fit the spectra from the three observatories to constrain the presence of a relativistic reflection component. From this, we obtain constraints on the inclination angle to the inner disc (<39° at 99 per cent confidence) and the disc inner radius (<11 gravitational radii at 99 per cent confidence), although we note that modelling systematics in practice add to the quoted statistical error. Our spectropolarimetric modelling indicates that the 2–8 keV polarization is consistent with being dominated by emission directly observed from the X-ray corona, but the polarization of the reflection component is completely unconstrained. Our constraints on viewer inclination and polarization degree tentatively favour more asymmetric, possibly out-flowing, coronal geometries that produce more highly polarized emission, but the coronal geometry is unconstrained at the 3σ level
