    Avaliação toxicológica pré-clinica do fitoterápico contendo Gossypium herbaceum (Tintura de Algodoeiro Cangeri) em ratos wistar

    O presente estudo avaliou a segurança de um fitoterápico, constituído de extrato fluído de Gossypium herbaceum (algodoeiro) (Tintura de Algodoeiro Cangeri®), através de estudos de toxicidade subcrônica, tendo como base a resolução Nº 90, de 16 de março de 2004 da Agência Nacional de Vigilância Sanitária (ANVISA). O objetivo era verificar se o presente fitoterápico causava toxicidade sistêmica de doses repetidas em ratos Wistar. Para o teste de toxicidades repetidas constituiu-se quatro grupos de ratos Wistar, cada um composto por 10 machos e dez fêmeas, tratados por via oral (gavagem) com 0,4ml.kg-1, 2ml.kg-1e 4ml.kg-1, respectivamente a dose terapêutica indicada pelo fabricante para seres humanos adultos, 5 vezes e 10 vezes a dose terapêutica, além de um grupo controle, que recebeu o veículo do fitoterápico. Os resultados revelaram que os animais tratados com a dose terapêutica não mostraram alterações sistêmicas que indique toxicidade. As alterações observadas nas variáveis de ganho de massa corporal, dados hematológicos e de bioquímica sangüínea nos grupos tratados com as doses 5 e 10 vezes a dose terapêutica revelaram a ocorrência de toxicidade sistêmica, agravada com a elevação da mesma.The phytoterapic constituted by Gossypium herbaceaum (cotton plant) (Tintura de Algodoeiro Cangeri®) was investigated from the potential of toxicological effects when orally administered to male and female Wistar rats for 30 days. The studies were based in the resolution n. 90, March 16th, 2004 from National Agency of Sanitary Vigilance (ANVISA). The aim of the study was watch if daily doses of the fitoterapic would became in to systemic toxicity. Four experimental groups were formed (10 animals/sex/dose) receiving daily doses of 0.4ml, 2ml and 4ml.kg-1 respectively the therapeutic dose recommended by the producer for adult humans, 5 times and 10 times the therapeutic dose, besides a control group treated with the phytotherapic vehicle. The results revealed the absence of systemic toxicity with the therapeutic dose. The alterations observed in the weight gain, hematological values and biochemical blood analyses in the doses equivalent to 5 and 10 times the therapeutic doses revealed systemic toxicity, in a dose related manner

    Genomic and temporal analyses of Mycobacterium bovis in southern Brazil.

    Mycobacterium bovis is a causal agent of bovine tuberculosis (bTB), one of the most important diseases currently facing the cattle industry worldwide. Tracing the source of M. bovis infections of livestock is an important tool for understanding the epidemiology of bTB and defining control/eradication strategies. In this study, whole genome sequencing (WGS) of 74 M. bovis isolates sourced from naturally infected cattle in the State of Rio Grande do Sul (RS), southern Brazil, was used to evaluate the population structure of M. bovis in the region, identify potential transmission events and date the introduction of clonal complex (CC) European 2 (Eu2). In silico spoligotyping identified 11 distinct patterns including four new profiles and two CCs, European 1 (Eu1) and Eu2. The analyses revealed a high level of genetic diversity in the majority of herds and identified putative transmission clusters that suggested that within- and between-herd transmission is occurring in RS. In addition, a comparison with other published M. bovis isolates from Argentina, Brazil, Paraguay and Uruguay demonstrated some evidence for a possible cross-border transmission of CC Eu1 into RS from Uruguay or Argentina. An estimated date for the introduction of CC Eu2 into RS in the middle of the 19th century correlated with the historical introduction of cattle into RS to improve existing local breeds. These findings contribute to the understanding of the population structure of M. bovis in southern Brazil and highlight the potential of WGS in surveillance and helping to identify bTB transmission
