Architecture of the Hin Synaptic Complex during Recombination The Recombinase Subunits Translocate with the DNA Strands
AbstractMost site-specific recombinases can be grouped into two mechanistically distinct families. Whereas tyrosine recombinases exchange DNA strands through a Holliday intermediate, serine recombinases such as Hin generate double-strand breaks in each recombining partner. Here, site-directed protein crosslinking is used to elucidate the configuration of protein subunits and DNA within the Hin synaptic complex and to follow the movement of protein subunits during DNA strand exchange. Our results show that the protein interface mediating synapsis is localized to a region within the catalytic domains, thereby positioning the DNA strands on the outside of the Hin tetrameric complex. Unexpected crosslinks between residues within the dimerization helices provide evidence for a conformational change that accompanies DNA cleavage. We demonstrate that the Hin subunits, which are linked to the cleaved DNA ends by serine-phosphodiester bonds, translocate between synapsed dimers to exchange the DNA strands
Oscillations in meta-generalized-gradient approximation potential energy surfaces for dispersion-bound complexes

It has been pointed out by Johnson et al. [Chem. Phys. Lett. 394, 334 (2004) ] that meta-GGA potential energy curves in dispersion-bound complexes are susceptible to spurious oscillations unless very large integration grids are used. This grid sensitivity originates from the saddle-point region of the density near the intermonomer midpoint. Various dimensionless ratios involving the kinetic-energy density, found in typical meta-GGAs, may be ill-behaved in this region. Grid sensitivity thus arises if the midpoint region is sampled by too sparse a grid. For most meta-GGAs, standard grids do not suffice. Care must be taken to avoid this problem when using, or constructing, meta-GGAs
Avian Foraging Patterns in Crop Field Edges Adjacent to Woody Habitat
As natural predators of pest insects, woodland birds provide biological pest suppression in crop fields adjacent to woody edges. Although many birds using these habitats forage widely, earlier studies have found that most foraging activity occurs within 50 m of the woody edge. The goals of this study were to determine the primary area of use, or functional edge, for birds foraging in crop fields adjacent to woody edges, and to evaluate their foraging distance patterns. During the summers of 2005 and 2006, avian foraging behavior was observed at 12 research sites in east central Nebraska that contained either a shelterbelt or woody riparian edge. At each site, perches were provided at 10 m intervals out from the edge and insect larvae were placed in feeders at random locations to simulate a pest insect food resource. Birds were recorded foraging in five distance categories out from the edge (0–10, 10–20, 20–30, 30–40, and 40–50 m). Seven species foraged primarily within 20 m of the edge (72% all observations; 79% without perch or feeder observations). Ten species foraged throughout the plots but six of these generally foraged more often (45% and 49%) and four less often (30% and 30%) within 20 m of the edge. The 13 species that tended to forage more often within 20 m of the edge, with 56% of their foraging overall in this area, also tended to forage farther when perch and feeder observations were included, indicating willingness to forage farther when food resources were available. Based on a repeated measures analysis of variance, foraging distances appeared to be greater at sites with soybean as the planted crop, although this apparent trend was significant for only some species. There was no clear difference in foraging distances outward from shelterbelt versus riparian sites. These results indicate that conservation efforts within the 20 m functional edge offer potential to enhance the sustainability of both birds and crops in agricultural

Creation of a Southwestern Authors Archive: Revisiting the LaVerne Harrell Clark Collection Ten Years Later
This article details the processing of the LaVerne Harrell Clark Photographic Collection by University of Arizona Poetry Center librarians, archivists, and interns, and the current status and next steps of the collection. The LaVerne Harrell Clark Photographic Collection, which contains thousands of portraits and candids of poets taken circa 1960-2000 by LaVerne Harrell Clark, the Poetry Center’s first director, is deeply entwined with the Poetry Center’s institutional identity. In addition, the authors explored ways in which the collection and the institution as a whole are positioned as uniquely Western and also of national significance. Those processing the collection became interested in the ways that this identity might affect other collections within the institution, such as the book collection and other archival collections. At the core of this exploration were questions about how the process of documentation and identity making occurs
Yttrium-90 Selective Internal Radiotherapy As Bridge to Curative Hepatectomy for Recurrent Malignant Solid Pseudopapillary Neoplasm of Pancreas: Case Report and Review of Literature
Recurrent malignant solid pseudopapillary neoplasms of the pancreas (SPNP) are rare tumors with unpredictable clinical and histopathological features. There is a lack of consensus regarding utilization of adjuvant modalities in conjunction with or in lieu of curative metastatectomy. We present a remarkable case where Yttrium-90 selective internal radiation therapy (Y-90 SIRT) was successfully utilized to elucidate underlying tumor biology and aid resection of a large multifocal recurrent metastatic SPNP in the right hemi-liver of a 59-year-old female. Thus, in cases where curative metastatectomy remains the treatment goal in management of recurrent and/or metastatic SPNPs, Y-90 SIRT is a safe and effective adjunct treatment to facilitate curative resection
Understanding uncertainty in temperature effects on vector-borne disease: A Bayesian approach
Extrinsic environmental factors influence the distribution and population
dynamics of many organisms, including insects that are of concern for human
health and agriculture. This is particularly true for vector-borne infectious
diseases, like malaria, which is a major source of morbidity and mortality in
humans. Understanding the mechanistic links between environment and population
processes for these diseases is key to predicting the consequences of climate
change on transmission and for developing effective interventions. An important
measure of the intensity of disease transmission is the reproductive number
. However, understanding the mechanisms linking and temperature, an
environmental factor driving disease risk, can be challenging because the data
available for parameterization are often poor. To address this we show how a
Bayesian approach can help identify critical uncertainties in components of
and how this uncertainty is propagated into the estimate of . Most
notably, we find that different parameters dominate the uncertainty at
different temperature regimes: bite rate from 15-25 C; fecundity across
all temperatures, but especially 25-32 C; mortality from
20-30 C; parasite development rate at 15-16C and again at
33-35C. Focusing empirical studies on these parameters and
corresponding temperature ranges would be the most efficient way to improve
estimates of . While we focus on malaria, our methods apply to improving
process-based models more generally, including epidemiological, physiological

Workplace Traumatic Stress and Mental Health Sequelae among Public Safety Telecommunications Officers in Florida
Background: Public safety telecommunications officers (PSTCOs), aka emergency “dispatchers,” are exposed to workplace traumatic stress and can experience situations characterized by uncertainty, communication difficulties, and a lack of resources. Traumatic stress experienced by emergency dispatchers has led to mental health symptoms. Purpose: This paper aims to describe the results of a study examining the patterns of workplace traumatic stress and the relationship between workplace traumatic stress and mental health concerns among a sample of PSTCOs. Methods: PSTCOs (n=54) participated in a cross-sectional, anonymous survey including screeners for depression, anxiety, post-traumatic stress disorder (PTSD), suicidal thoughts, and harmful alcohol use. Respondents also completed the Life Events Checklist (LEC) to screen for exposure to emergency calls identified as having the potential to induce traumatic stress. Results: 18.4% of respondents reported experiencing moderate to severe levels of depression, and 12% reported moderate to severe anxiety. Of the sample, 14% met the criteria for a provisional diagnosis of PTSD, 40% reported hazardous drinking levels, and 10% met the criteria of being at risk for suicide. Overall, 72.2% of PSTCOs experienced at least one form of job-related trauma and varied significantly by marital status. Poisson regression revealed statistically significant relationships between exposure to job-related traumatic events and screener scale scores. For example, those exposed to assault with a weapon had depression scores 2.29 points higher compared to those who were not exposed (p≤0.05). Discussion: Employing organizations of PSTCOs should strive to incorporate comprehensive wellness programs emphasizing education, prevention, early intervention, and recognition of traumatic stress among dispatchers. These programs should emphasize peer support and non-punitive policies to encourage help-seeking. In addition, given that findings in this study indicate exposure to traumatic emergency calls predicts PTSD symptomology, legislation could consider including PSTCOs in Florida Statute 118.1215
Associations between residual feed intake and apparent nutrient digestibility, in vitro methane-producing activity, and volatile fatty acid concentrations in growing beef cattle
The objectives of this study were to examine the relationship between residual feed intake (RFI) and DM and nutrient digestibility, in vitro methane production, and volatile fatty acid (VFA) concentrations in growing beef cattle. Residual feed intake was measured in growing Santa Gertrudis steers (Study 1; n = 57; initial BW = 291.1 ± 33.8 kg) and Brangus heifers (Study 2; n = 468; initial BW = 271.4 ± 26.1 kg) fed a high-roughage-based diet (ME = 2.1 Mcal/kg DM) for 70 d in a Calan-gate feeding barn. Animals were ranked by RFI based on performance and feed intake measured from day 0 to 70 (Study 1) or day 56 (Study 2) of the trial, and 20 animals with the lowest and highest RFI were identified for subsequent collections of fecal and feed refusal samples for DM and nutrient digestibility analysis. In Study 2, rumen fluid and feces were collected for in vitro methane-producing activity (MPA) and VFA analysis in trials 2, 3, and 4. Residual feed intake classification did not affect BW or BW gain (P \u3e 0.05), but low-RFI steers and heifers both consumed 19% less (P \u3c 0.01) DMI compared with high-RFI animals. Steers with low RFI tended (P \u3c 0.1) to have higher DM digestibility (DMD) compared with high-RFI steers (70.3 vs. 66.5 ± 1.6% DM). Heifers with low RFI had 4% higher DMD (76.3 vs. 73.3 ± 1.0% DM) and 4 to 5% higher (P \u3c 0.01) CP, NDF, and ADF digestibility compared with heifers with high RFI. Low-RFI heifers emitted 14% less (P \u3c 0.01) methane (% GE intake; GEI) calculated according to Blaxter and Clapperton (1965) as modified by Wilkerson et al. (1995), and tended (P = 0.09) to have a higher rumen acetate:propionate ratio than heifers with high RFI (GEI = 5.58 vs. 6.51 ± 0.08%; A:P ratio = 5.02 vs. 4.82 ± 0.14%). Stepwise regression analysis revealed that apparent nutrient digestibilities (DMD and NDF digestibility) for Study 1 and Study 2 accounted for an additional 8 and 6%, respectively, of the variation in intake unaccounted for by ADG and mid-test BW0.75. When DMD, NDF digestibility, and total ruminal VFA were added to the base model for Study 2, trials 2, 3, and 4, the R2 increased from 0.33 to 0.47, explaining an additional 15% of the variation in DMI unrelated to growth and body size. On the basis of the results of these studies, differences in observed phenotypic RFI in growing beef animals may be a result of inter-animal variation in apparent nutrient digestibility and ruminal VFA concentrations
