    Comparative Recovery of Coliforms from Meat and Milk Using m-Coliblue24 and Standard Methods

    A new membrane filtration medium, m-ColiBlue24 (mCB), simultaneously detects Total Coliforms (TC) and Escherichia coli (EC) in a single 24-hour incubation. Samples of ground beef, ground pork, and milk were analyzed by mCB and compared to standard methods. When TC from beef and pork samples were compared, there were no significant difference between mCB and pour plating on Violet Red Bile (VRB) agar. In a most probable number (MPN) analysis, nine counts from mCB were within the 95% confidence interval (CI) and 13 were outside the CI. VRB yielded six counts within and 16 outside the CI. The raw milk samples contained confirmed TC counts as high as 9.00X10 2 on mCB and 2.20X10 3 on VRB. Furthermore, the raw milk had up to 1.73X10 3 confirmed EC population. The BBL Crystal system was used to identify representative isolates from mCB and VRB. Of 59 mCB isolates, 64.4% were classical TC. Of the 51 VRB isolates, 62.7% were identified as classical TC

    A New Technique for Escherichia coli Testing of Beef and Pork Carcasses

    A novel technique has been developed to monitor Escherichia coli contamination on carcasses using membrane filtration and m-ColiBlue24 (mCB). mCB is a membrane filtration medium that simultaneously detects total coliforms and E. coli (EC) in a period of 24 +/- 4 h. A study was conducted, using a sponge method to obtain samples from pork carcasses and the excision technique to remove samples from beef carcasses, that compared mCB to standard methods. On pork carcasses (n = 77), the mean values for mCB and violet red bile agar were 7.4 CFU/15 cm2 and 6.1 CFU/15 cm2, respectively. The paired t test (P \u3e 0.05) indicated no significant difference between the two methods (t = 0.5; P = 0.6). Samples from beef carcasses (n = 57) were used to compare mCB to both coliform count and EC Petrifilm. Of these samples, 27 were artificially inoculated with cattle manure. The mean total coliform count was 4.2 log CFU/cm2 and 4.0 log CFU/cm2 on mCB and coliform count Petrifilm, respectively. The mean EC count on mCB was 4.0 log CFU/cm2 and 3.5 log CFU/cm2 on EC Petrifilm. When comparing mCB to both coliform count (t = 2.4; P = 0.02) and EC (t = 3.5; P \u3c 0.01) Petrifilm, paired t tests (P \u3c or = 0.05) indicated significant differences

    A novel physiological role for ARF1 in the formation of bidirectional tubules from the Golgi.

    Capitalizing on CRISPR/Cas9 gene-editing techniques and super-resolution nanoscopy, we explore the role of the small GTPase ARF1 in mediating transport steps at the Golgi. Besides its well-established role in generating COPI vesicles, we find that ARF1 is also involved in the formation of long (∼3 µm), thin (∼110 nm diameter) tubular carriers. The anterograde and retrograde tubular carriers are both largely free of the classical Golgi coat proteins coatomer (COPI) and clathrin. Instead, they contain ARF1 along their entire length at a density estimated to be in the range of close packing. Experiments using a mutant form of ARF1 affecting GTP hydrolysis suggest that ARF1[GTP] is functionally required for the tubules to form. Dynamic confocal and stimulated emission depletion imaging shows that ARF1-rich tubular compartments fall into two distinct classes containing 1) anterograde cargoes and clathrin clusters or 2) retrograde cargoes and coatomer clusters

    Genetic Determinants of Circulating Sphingolipid Concentrations in European Populations

    Sphingolipids have essential roles as structural components of cell membranes and in cell signalling, and disruption of their metabolism causes several diseases, with diverse neurological, psychiatric, and metabolic consequences. Increasingly, variants within a few of the genes that encode enzymes involved in sphingolipid metabolism are being associated with complex disease phenotypes. Direct experimental evidence supports a role of specific sphingolipid species in several common complex chronic disease processes including atherosclerotic plaque formation, myocardial infarction (MI), cardiomyopathy, pancreatic beta-cell failure, insulin resistance, and type 2 diabetes mellitus. Therefore, sphingolipids represent novel and important intermediate phenotypes for genetic analysis, yet little is known about the major genetic variants that influence their circulating levels in the general population. We performed a genome-wide association study (GWAS) between 318,237 single-nucleotide polymorphisms (SNPs) and levels of circulating sphingomyelin (SM), dihydrosphingomyelin (Dih-SM), ceramide (Cer), and glucosylceramide (GluCer) single lipid species (33 traits); and 43 matched metabolite ratios measured in 4,400 subjects from five diverse European populations. Associated variants (32) in five genomic regions were identified with genome-wide significant corrected p-values ranging down to 9.08 x 10(-66). The strongest associations were observed in or near 7 genes functionally involved in ceramide biosynthesis and trafficking: SPTLC3, LASS4, SGPP1, ATP10D, and FADS1-3. Variants in 3 loci (ATP10D, FADS3, and SPTLC3) associate with MI in a series of three German MI studies. An additional 70 variants across 23 candidate genes involved in sphingolipid-metabolizing pathways also demonstrate association (p = 10(-4) or less). Circulating concentrations of several key components in sphingolipid metabolism are thus under strong genetic control, and variants in these loci can be tested for a role in the development of common cardiovascular, metabolic, neurological, and psychiatric diseases

    Site fidelity and movement patterns of reef manta rays (Mobula alfredi: Mobulidae) using passive acoustic telemetry in northern Raja Ampat, Indonesia

    Though extremely valuable to the local marine tourism industry, there is a dearth of published information on the ecology and population dynamics of reef manta rays (Mobula alfredi) in Raja Ampat, West Papua, Indonesia. Knowledge of the movement ecology in particular of this large and scattered population is urgently needed to better manage the rapidly expanding manta-focused tourism. Here we report the results of an initial passive acoustic telemetry study designed to provide local managers with the first detailed knowledge of the site use and movement patterns of reef mantas in northern Raja Ampat. A total of 39 reef mantas were tagged with Vemco V16 acoustic transmitters over a 15-month period between 27 November 2013 and 22 February 2015. To monitor their movements, VR2W acoustic receivers were deployed at eight sites corresponding to known manta cleaning and feeding aggregation sites, with receivers downloaded every six months over a two-year initial monitoring period. The duration between tag deployments and last date of detections at sites ranged from 1 to 682 days (mean ± SE = 237 ± 27). The cumulative number of days of manta detections at receiver sites by individual mantas ranged from 1 to 188 days (mean ± SE = 42 ± 7). Manta Ridge was the most visited site with 565 days of detections. The tagged mantas demonstrated strong site fidelity to the observed aggregation sites. At the same time, they also exhibited seasonal movements within an approximately 150 km long corridor between sites in the Dampier Strait and the northwest of Waigeo Island. Data analysed from a nearby array of six VR2W receivers in southern Raja Ampat (approximately 180 km to the south of the study area) confirmed that none of the tagged mantas were detected in this array, providing further evidence of strong site fidelity and limited movements within northern Raja Ampat. More than 96% of detections occurred during the daytime. The number of detections reached a peak around noon at Yefnabi Kecil and Eagle Rock and slightly earlier at Manta Ridge. These findings have been shared with the Raja Ampat Marine Protected Area Management Authority and are now being used in the formulation of a management plan for this vulnerable and economically important species to ensure the long-term health of Raja Ampat's reef mantas and the sustainability of manta tourism in the region

    Can Historical Institutionalism be Applied to Political Regime Development in Africa?

    Historical institutionalism has been used to explain the emergence of democracy and dictatorship in various regions of the world, but not applied to political development in Africa. Based on the recently refined concepts of historical institutionalism, the aim of this study is to provide a framework for the analysis of the various regime types that have been established in Africa during the last two decades: democratic, hybrid and authoritarian. Surprisingly little effort has been dedicated to a historically grounded explanation of these regime types. Against a common claim that African politics is mainly driven by informal institutions or behaviours, we argue that an institution-based examination of African politics is justified. We then provide a proposition of how to link up concepts of historical institutionalism with empirical cases in Africa, within a comparative approach. Our proposition for tracing specific development paths will not be based on the regimes as a 'whole', but on the deconstruction of a political regime into partial regimes and subsequently into selected formal and informal institutions. This will allow for an empirical analysis of the different components of a regime over long periods of time, and thus for path-dependent analyses of regime development

    Measurement of the lepton charge asymmetry in W-boson decays produced in p-pbar collisions

    We describe a measurement of the charge asymmetry of leptons from W boson decays in the rapidity range 0 enu, munu events from 110+/-7 pb^{-1}of data collected by the CDF detector during 1992-95. The asymmetry data constrain the ratio of d and u quark momentum distributions in the proton over the x range of 0.006 to 0.34 at Q2 \approx M_W^2. The asymmetry predictions that use parton distribution functions obtained from previously published CDF data in the central rapidity region (0.0<|y_l|<1.1) do not agree with the new data in the large rapidity region (|y_l|>1.1).Comment: 13 pages, 3 tables, 1 figur

    A full angular analysis has been performed for the pseudo-scalar to vector-vector decays, B0 --> J/psi K* and B_s --> J/psi phi, to determine the amplitudes for decays with parity-even longitudinal and transverse polarization and parity-odd transverse polarization. The measurements are based on 190 B0 candidates and 40 B_s candidates collected from a data set corresponding to 89 inverse pb of pbarp collisions at root(s) = 1.8 TeV at the Fermilab Tevatron. In both decays the decay amplitude for longitudinal polarization dominates and the parity-odd amplitude is found to be small.Comment: 7 pages, 3 figures, 1 tabl

    Observation of Hadronic W Decays in t-tbar Events with the Collider Detector at Fermilab

    We observe hadronic W decays in t-tbar -> W (-> l nu) + >= 4 jet events using a 109 pb-1 data sample of p-pbar collisions at sqrt{s} = 1.8 TeV collected with the Collider Detector at Fermilab (CDF). A peak in the dijet invariant mass distribution is obtained that is consistent with W decay and inconsistent with the background prediction by 3.3 standard deviations. From this peak we measure the W mass to be 77.2 +- 4.6 (stat+syst) GeV/c^2. This result demonstrates the presence of two W bosons in t-tbar candidates in the W (-> l nu) + >= 4 jet channel.Comment: 20 pages, 4 figures, submitted to PR

    Measurement of J/Psi and Psi(2S) Polarization in ppbar Collisions at sqrt(s) = 1.8 TeV

    We have measured the polarization of J/Psi and Psi(2S) mesons produced in p\bar{p} collisions at \sqrt{s} = 1.8 TeV, using data collected at CDF during 1992-95. The polarization of promptly produced J/Psi [Psi(2S)] mesons is isolated from those produced in B-hadron decay, and measured over the kinematic range 4[5.5] < P_T < 20 GeV/c and |y| < 0.6. For P_T \gessim 12 GeV/c we do not observe significant polarization in the prompt component.Comment: Revised version, accepted for publication in Physical Review Letter
