    Procjena serumskoga srčanog troponina I u ovaca s akutnom mliječnokiselinskom acidozom buraga.

    Cardiac troponin I (cTnI) is known to be a sensitive cardio biomarker to determine the myocardial damage in diseases affecting the cardiac muscles. However, there has not been sufficient research about cTnI concentration, which is the most sensitive indicator of myocardial damage in sheep with acute ruminal lactic acidosis (ARLA). For this reason this study aimed to evaluate the serum cTnI concentration in sheep with ARLA. Those diagnosed with ARLA (n = 20) from the total of 40 Akkaraman (White karaman) sheep, aged between 1-2 years used in this study comprised the affected group and the healthy ones (n = 20) comprised the control group. Ruminal fluid was obtained from the animals from both groups with the help of a stomach tube, and examined immediately. Blood samples were taken from the jugular vein of the sheep and the serum was separated. Serum cTnI concentration was measured with a commercial immunoassay system, using the one-step sandwich method. Serum enzyme (ALT, AST, CK-MB and LDH) activities were determined via a clinical biochemistry autoanalyzer. The average serum cTnI concentration was at the level of 0.035 ± 0.015 ng/mL (range; 0.02-0.06 ng/mL) in the control group sheep. It was determined that there was a substantial increase in the group with ARLA and the average concentration reached the level of 0.103 ± 0.080 ng/mL (range; 0.03-1.7 ng/mL) (P<0.0001). It was observed that another cardio marker, CK-MB, was found in the group with ARLA 454.50 ± 191.88 U/L (range; 214-861 U/L) and increased in comparison with the control group 224.35 ± 83.33 U/L (range; 133-421 U/L) (P<0.0001). An increase in LDH (P<0.001) and AST (P<0.001) from liver enzymes in the group with ARLA and ALT activities compared to the control group was identified (P<0.01). In conclusion, this present study determined that the serum cTnI concentration was high in sheep with ARLA and it was concluded that it could be useful to evaluate cTnI concentration as an important marker to determine the prognosis in sheep with ARLA.U bolestima koje zahvaćaju srčani mišić, srčani troponin I (cTnI) poznat je kao osjetljivi biomarker za određivanje oštećenja srčanog mišića. Ipak, nema puno istraživanja o koncentraciji cTnI kao najosjetljivijeg indikatora za oštećenje srčanog mišića kod ovaca s akutnom mliječno-kiselinskom acidozom buraga. Zbog navedenog, u ovom istraživanju namjera je bila procijeniti koncentraciju serumskog cTnI kod ovaca s akutnom mliječno-kiselinskom acidozom. Ovce s dijagosticiranom akutnom mliječno-kiselinskom acidozom (n = 20) u skupini od ukupno 40 akaraman (bijeli karaman) ovaca, u dobi od jedne do dvije godine, činile su pokusnu skupinu bolesnih jedinki. Preostale ovce (n = 20) činile su kontrolnu skupinu zdravih jedinki. Tekućina buraga od svih pretraženih životinja dobivena je sondiranjem i odmah analizirana. Uzorci krvi uzeti su iz jugularne vene te je iz njih izdvojen serum. Koncentracija serumskog cTnI mjerena je komercijalnim imunoenzimnim testom, sendvič postupkom. Aktivnosti serumskih enzima (ALT, AST, CK-MB i LDH) utvrđene su uporabom kliničkog biokemijskog autoanalizatora. Prosječna serumska koncentracija cTnI u kontrolnoj skupini ovaca bila je na razini 0,035 ± 0,015 ng/mL (raspon: 0,02 - 0,06 ng/mL). U skupini ovaca s acidozom utvrđeno je postojano povećanje koncentracije cTnI koja je dosegla razinu od 0,103 ± 0,080 ng/mL (raspon: 0,03 - 1,7 ng/mL) (P<0,0001). U pokusnoj skupini opaženo je povećanje i drugog srčanog markera CK-MB koji je u ovoj skupini iznosio 454,50 ± 191,88 U/I (raspon: 214 - 861 U/I), a u kontrolnoj 224,35 ± 83,33 U/I (raspon: 133 - 421 U/I) (P<0,0001). Također je ustanovljeno da su ovce s acidozom u odnosu na ovce kontrolne skupine imale povišene jetrene enzime LDH (P<0,001) i AST (P<0,001), odnosno ALT (P<0,01). Zaključeno je da ovce s acidozom imaju povećanu koncentraciju serumskog cTnI koji može poslužiti kao važan biljeg za prognozu bolesti

    Introduction: in recent years, platelets are known to have a large variety of functions in many pathophysiological processes and their interaction with endothelial cells and leukocytes is known to play an important role in the pathophysiology of vascular inflammation. The aim of this study was to investigate the relationship between white blood cell count in conditions resulting in leukocytosis and platelet count and platelet parameters including mean platelet volume, platelet distribution width, and plateletcrit. Methods: white blood cell counts count and all platelet parameters were evaluated in 341 results of normal complete blood count (of which the white blood cell counts were within reference range, group 1) and 327 results of elevated white blood cell counts count (group 2). Results: there was a significant difference between these two groups in PLT counts and PCT values, being higher in Group 2. However, there was no statistically significant difference between two groups in MPV and PDW values. On the other hand, there were statistically significant, but weak, correlations between the WBC and platelet counts in both groups (p&lt;0.01, r=0.235 for group 1, p&lt;0.05, r=0.116 for group 2). Conclusion: as a conclusion PLT count and PCT values increase in infectious conditions. This study and previous studies show that PLTs are employed in infectious conditions but the exact mechanism and the exact clinical importance of this response remains to be cleared by further studies.The Pan African Medical Journal 2016;2

    The effect of exercise in some sport branches on urinary second messenger cyclic nucleotide levels

    In this study, the possible effects of exercise on hormonal secretions in some sport branches were investigated on the basis of urinary cyclic nucleotide (cAMP and cGMP) excretion, since it was thought that the total, ultimate effects of secreted hormones, under the effect of exercise stress, may be estimated by means of the changes in the second messenger, cAMP, and cGMP, concentrations in urine. A total of 60 subjects from different sport branches were included in the study, and pre- and post-training urinary cyclic nucleotide levels of them were determined by EIA method. The differences in cAMP and cGMP between pre-training and post-training periods were statistically significant (p < 0.05 for both), with the post-training levels being higher. The increased post-training cyclic nucleotide concentrations in urine showed that a complex hormonal response occurs as a result of both metabolic state and the exercise stress in sporters. It was concluded that the complicated hormonal status of the sporters may be explained partly by determining the second messenger concentrations. Since, the present study looking at this perspective is a preliminary one in this area, the determination of all second messenger changes for the evaluation of the total hormonal status of sporters is needed in future

    Comparison of Cobas 6500 and Iris IQ200 fully-automated urine analyzers to manual urine microscopy.

    Introduction: Urine screening is achieved by either automated or manual microscopic analysis. The aim of the study was to compare Cobas 6500 and Iris IQ200 urine analyzers, and manual urine microscopic analysis. Materials and methods: A total of 540 urine samples sent to the laboratory for chemical and sediment analysis were analyzed on Cobas 6500 and Iris IQ200 within 1 hour from sampling. One hundred and fifty three samples were found to have pathological sediment results and were subjected to manual microscopic analysis performed by laboratory staff blinded to the study. Spearman’s and Gamma statistics were used for correlation analyses, and the McNemar test for the comparison of the two automated analyzers. Results: The comparison of Cobas u701 to the manual method yielded the following regression equations: y = - 0.12 (95% CI: - 1.09 to 0.67) + 0.78 (95% CI: 0.65 to 0.95) x for WBC and y = 0.06 (95% CI: - 0.09 to 0.25) + 0.66 (95% CI: 0.57 to 0.73) x for RBC. The comparison of IQ200 Elite to manual method the following equations: y = 0.03 (95% CI: - 1.00 to 1.00) + 0.88 (95% CI: 0.66 to 1.00) x for WBC and y = - 0.22 (95% CI: - 0.80 to 0.20) + 0.40 (95% CI: 0.32 to 0.50) x for RBC. IQ200 Elite compared to Cobas u701 yielded the following equations: y = - 0.95 (95% CI: - 2.13 to 0.11) + 1.25 (95% CI: 1.08 to 1.44) x for WBC and y = - 1.20 (95% CI: - 1.80 to -0.30) + 0. 80 (95% CI: 0.55 to 1.00) x for RBC. Conclusions: The two analyzers showed similar performances and good compatibility to manual microscopy. However, they are still inadequate in the determination of WBC, RBC, and EC in highly-pathological samples. Thus, con?rmation by manual microscopic analysis may be useful
