    The effects of different combinations of eugenol and acetic acid on quality changes of marinated rainbow trout (Oncorhynchus mykiss, W. 1792) fillets

    In this study, fillets of trout marinated with different concentrations of acetic acid (2% and 4%) and eugenol (0.1% v/w and 0.5% v/w) were preserved at 4 ºC. The samples were analyzed in terms of sensory, microbiological and chemical quality. During the preservation, the total mesophilic anaerobe and Lactobacillus showed significant changes (p0.05), whereas thiobarbituric acid (TBA) showed significant changes (p0.05), tiyobarbitürik asit (TBA) önemli derecede değişimler göstermiştir (p<0.05). Duyusal analizde, lezzet ve genel kabul edilebilirlik muhafaza süresince gruplar arasında önemli farklılıklar göstermiştir. Sonuç olarak, marinatlar 70 gün süresince tüketilebilirlik özelliklerini korumuş ve eugenolün bazı mikrobiyolojik ve kimyasal değerler üzerine olumlu etkileri olmuştur. %0.1 eugenolle hazırlanan marinatlar duyusal bakımdan daha fazla beğeni kazanmıştır

    Dual Suppressive Effect of miR-34a on the FOXM1/eEF2-Kinase Axis Regulates Triple-Negative Breast Cancer Growth and Invasion

    Purpose: Recent studies indicated that dysregulation of noncoding KNAs (ncRNA) such as miRNAs is involved in pathogenesis of various human cancers. However, the molecular mechanisms underlying miR-34a are not fully understood in triple-negative breast cancer (TNBC). Experimental Design: We performed in vitro functional assays on TNBC cell lines to investigate the role of mi R-34a in FOLM1/eEF2K signaling axis. TNBC tumor xenograft models were used for in vivo therapeutic delivery of miR-34a. Results: In this study, we investigated the role of p53-driven ncRNA miR-34a and found that miR-34a is associated with significantly longer patient survival in TNBC and inversely correlated with levels of proto-oncogenic eEF2K, which was associated with significantly shorter overall patient survival, We showed that miR-34a directly binds to the 3'-untranslated region of eEF2K and FOXM1 mRNAs and suppresses their expression, leading to inhibition of TNBC cell proliferation, motility, and invasion. Notably, restoring miR-34a expression recapitulated the effects of inhibition of eEF2K and FOXM1, the transcription factor for eEF2K and the direct target of p53, in TNBC cell lines, whereas overexpression of eEF2K and FOXM1 rescued the effects and signaling pathways mediated by miR-34a. Moreover, in vivo therapeutic delivery of miR-34a nanopartides by systemic intravenous administration delayed tumor growth of two different orthotopic TNBC tumor xenograft models by inhibiting eEF2K and FOXM1, intratumoral proliferation and angiogenesis, and inducing apoptosis. Conclusions: Overall, our findings provide new insights into the tumor suppressor role of miR-34a by dual-targeting of FOXM1/eEF2K signaling axis and suggest that miR-34a-based gene therapy may be a potential therapeutic strategy in TNBC.     Özet: Bu çalışmada, ekonomik yönden önemli bir balık türü olan ve farklı asetik asit solüsyonlarında marine edilen barbunya balığının çeşitli soslarda duyusal olarak değerlendirilmesi amaçlanmıştır. %4 ve %8 oranında asetik asit kullanılarak marine edilen balıklar 4 C'de 1 hafta olgunlaştırıldıktan sonra kimyasal bileşimleri incelenmiş ve 6 farklı sosla hazırlanan örnekler 10 panelist tarafından değerlendirilmiştir. Duyusal değerlendirmeler sonucunda en fazla beğeniyi %4 asetik asit ve %10 tuz ile olgunlaştırıldıktan sonra A sosu (limon suyu, sarımsak, ceviz içi, zeytinyağı) ile sunulan grup kazanmıştır. Bu grup ile diğer gruplar arasındaki farklılığın istatistiksel olarak önemli olduğu tespit edilmiştir (p < 0,05). The marinated red mullet were treated with 4% and 8% acetic acid and then waited 4 C for one week and then their chemical composition were observed and were evaluated by 10 panelist by using 6 different sauces. At the result of sensory evaluations 4% acetic acid and 10% salt marinated and served with A sauce (lemon juice, garlic, walnuts, olive oil) group has got most admination.     Effect of Rosemary Essential Oil Decontamination on Quality Properties ofRainbow Trout (Oncorhynchus mykiss, W. 1972)

    Bu çalışmada, farklı dozlardaki biberiye esansiyel yağının (%0.5, %1, %2) sindirim sistemi çıkartılmamış bütün haldeki gökkuşağı alabalıklarının mikrobiyolojik (mezofil aerob bakteri, psikrofil aerob bakteri, laktik asit bakterileri, Pseudomonas spp., Enterobacteriaceae sayımları) ve kimyasal (pH, toplam uçucu bazik azot ve thiobarbiturik asit) özellikleri üzerine etkisi incelenmiştir. Bu amaçla; her grupta 6'şar adet balık dekontaminantlarla 20 ?C'de 2 dakika muamele edilmiş ve polietilen plastik tabaklarda streç film ile kaplandıktan sonra 41 ?C'de muhafazaya alınarak 0., 3., 6., 9., 12. ve 15. günlerde analiz edilmiştir. Mikrobiyolojik örnekler, USDA/FSIS tarafından kanatlı karkasları için onaylanmış metot da değişiklikler yapılarak kullanılmıştır. Çalışmanın sonuçlarına göre; gökkuşağı alabalıklarında biberiye esansiyel yağı ile dekontaminasyon yönteminin muhafaza süresini arttırdığı saptanmıştır. Ayrıca kullanılan mikrobiyolojik örnek alma yönteminin kulanım kolaylığı ve sonuçların tutarlılığı yönünden bilimsel çalışmalarda kullanılması tavsiye edilmektedir.In this study, the effects of different doses (0.5%, 1%, 2%) of rosemary essential oil on microbiological (mesophilic aerob bacteria, psycrophylic aerob bacteria, lactic acid bacteria, Pseudomonas spp., Enterobactericeae counts) and chemical (pH, total volatile alkaline nitrogen, thiobarbituric acid) properties of whole rainbow trouts were examined. For, this purpose, decontaminats at 20 ?C were applied to each group containing 6 fish for 2 min and placed in polyethylene ware and covered with stretch films and stored at 4&plusmn;1 ?C and samples were analyzed on the analysis day 0th, 3th, 6th, 9th, 12th and 15th days. Microbiological samples were prepared with a modification of USDA/FSIS chicken carcass method. According to the results of the study, decontamination with rosemary essential oils extends the shelf life of Rainbow trout. In addition, microbiological sampling method is recommended to be used in similar scientific studies because of its is easy of use coherent results
