    Ground-Penetrating-Radar Reflection Attenuation Tomography with an Adaptive Mesh

    Ground-penetrating radar (GPR) attenuation-difference analysis can be a useful tool for studying fluid transport in the subsurface. Surface-based reflection attenuation-difference tomography poses a number of challenges that are not faced by crosshole attenuation surveys. We create and analyze a synthetic attenuation-difference GPR data set to determine methods for processing amplitude changes and inverting for conductivity differences from reflection data sets. Instead of using a traditional grid-based inversion, we use a data-driven adaptive-meshing algorithm to alter the model space and to create amore even distribution of resolution. Adaptive meshing provides a method for improving the resolution of the model space while honoring the data limitations and improving the quality of the attenuation difference inversion. Comparing inversions on a conventional rectangular grid with the adaptive mesh, we find that the adaptively meshed model reduces the inversion computation time by an average of 75% with an improvement in the root mean square error of up to 15%. While the sign of the conductivity change is correctly reproduced by the inversion algorithm, the magnitude varies by as much as much as 50% from the true values. Our heterogeneous conductivity model indicates that the attenuation difference inversion algorithm effectively locates conductivity changes, and that surface-based reflection surveys can produce models as accurate as traditional crosshole surveys

    Black Bears Recolonizing Historic Ranges: Indiana Human–Bear Interactions

    Over a century after extirpation from Indiana, USA, 2 American black bears (Ursus americanus) were confirmed in the state during the summers of 2015 and 2016. The first bear encountered a public and management agency unaccustomed to living with large carnivores, which resulted in intentional and unintentional feedings, habituation, and ultimately its euthanasia. The Indiana Department of Natural Resources (DNR) attempted to learn from this encounter and began preparing for the next transient black bear. Education materials were created to help minimize human–bear interactions, promote living safely with bears, and inform about what to do when encountering a bear. Additionally, bear traps were purchased and staff were trained to safely deploy and use these traps. During the summer of 2016, when a second black bear was confirmed in Indiana, the DNR deployed targeted education and outreach materials to try to help maintain a positive living-with-bears environment and minimize human–bear interactions. Expanded public education and a slight increase in preparedness of the DNR provided 2 different outcomes for the recent bears in Indiana. These are likely not the last bears Indiana will host; habitat suitability models suggest that Indiana could potentially support bears in portions of the state. Natural range expansion in neighboring states suggest that Indiana will see more black bears in the future. These 2 bears highlighted the need for Indiana and other state agencies to have some preparations in place—especially related to education—to respond to transient large carnivores that are moving through and eventually recolonizing long extirpated areas

    Differential Expression of Pancreatic Protein and Chemosensing Receptor mRNAs in NKCC1-null Intestine

    AIM: To investigate the intestinal functions of the NKCC1 Na+-K+-2Cl cotransporter (SLC12a2 gene), differential mRNA expression changes in NKCC1-null intestine were analyzed. METHODS: Microarray analysis of mRNA from intestines of adult wild-type mice and gene-targeted NKCC1-null mice (n = 6 of each genotype) was performed to identify patterns of differential gene expression changes. Differential expression patterns were further examined by Gene Ontology analysis using the online Gorilla program, and expression changes of selected genes were verified using northern blot analysis and quantitative real time-polymerase chain reaction. Histological staining and immunofluorescence were performed to identify cell types in which upregulated pancreatic digestive enzymes were expressed. RESULTS: Genes typically associated with pancreatic function were upregulated. These included lipase, amylase, elastase, and serine proteases indicative of pancreatic exocrine function, as well as insulin and regenerating islet genes, representative of endocrine function. Northern blot analysis and immunohistochemistry showed that differential expression of exocrine pancreas mRNAs was specific to the duodenum and localized to a subset of goblet cells. In addition, a major pattern of changes involving differential expression of olfactory receptors that function in chemical sensing, as well as other chemosensing G-protein coupled receptors, was observed. These changes in chemosensory receptor expression may be related to the failure of intestinal function and dependency on parenteral nutrition observed in humans with SLC12a2 mutations. CONCLUSION: The results suggest that loss of NKCC1 affects not only secretion, but also goblet cell function and chemosensing of intestinal contents via G-protein coupled chemosensory receptors

    On the importance of a procedurally fair organizational climate for openness to change in law enforcement

    Objectives Drawing on recent work in policing and organizational psychology, we examined factors related to openness to organizational change and to adopting evidence-based interview techniques among law enforcement investigators. Hypotheses We hypothesized that a procedurally fair organizational climate would predict outcomes tied to organizational change, mediated by organizational identification and perceived legitimacy. We also predicted that procedural justice factors would be stronger predictors than outcome-oriented factors (i.e., rewards and sanctions). Methods Study 1 surveyed law enforcement investigators (N = 711) about their attitudes towards and behaviors within their organization (i.e., perceived procedural fairness of one’s organization, identification, legitimacy, compliance, empowerment, and extra-role behavior). Study 2 conceptually extended this survey to interviewers (N = 71) trained in a new, evidence-based interviewing approach adding likelihood of future use of the novel interviewing approach as an outcome. Results In Study 1, the more investigators thought their organization had a procedurally fair climate, the more they identified with the organization and perceived it as legitimate. Framing compliance, empowerment and extra-role behavior as associated with openness to change, we found that legitimacy predicted compliance and tendency toward extra-role behavior (i.e., going “above and beyond”), while level of identification predicted feelings of empowerment and extra-role behavior. Study 2, partially replicated findings from Study 1, and found that motivation to attend the training also predicted likelihood of future use. Conclusions These studies highlight the value of a procedurally just organizational climate framework in understanding law enforcement interrogators’ propensity towards implementing new evidence-based interrogation techniques

    The Association of Maternal Obesity and Race with Pregnancy Weight Gain and Small for Gestational Age Infant Birth: The Effect of Prenatal Care

    Objective: To examine the association of maternal obesity, race/ethnicity, and prenatal care on high gestational weight gain (GWG) and small for gestational age (SGA) infant birth. Methods: This was a retrospective cohort study of births included in the PRAMS Phase 8 dataset (2016-2017). The study population was 53,893 non-diabetic women with a singleton in-hospital birth between 37 and 42 weeks gestational age. Results: Only obese non-Hispanic white and Hispanic women showed a consistent decrease in adjusted odds of high GWG as prenatal care visit category increased. Only non-Hispanic white women showed a lower increase in adjusted odds of an SGA infant birth with more compared to intermediate prenatal care. Conclusions: The effectiveness of prenatal care in reducing high GWG varies by race for women with a BMI outside a healthy range. More prenatal care did not reduce SGA infant births amongst overweight or obese women. Policy implications: Interventions to improve prenatal care delivery for overweight or obese women should consider race

    The Incidence of Malignancy and the Preoperative Assessment of Women Undergoing Hysterectomy with Morcellation for Benign Indications

    Background: The use of power morcellation in gynecologic surgery has come under scrutiny secondary to concerns for occult malignancy dissemination. The incidence of undiagnosed gynecologic malignancy when hysterectomy performed for benign indications is not definitive but has been quoted as high as 2.7% (1:37). There is not a standard recommended preoperative evaluation, and variation is anticipated by preoperative complaint or diagnosis. Objectives: To quantify the malignancy incidence in women undergoing hysterectomy for benign indications and to compare the preoperative evaluation of patients undergoing hysterectomy with and without morcellation. Methods: Retrospective cohort of women undergoing hysterectomies between October 2007 and June 2014 was identified by procedural codes through the hospital billing system. Exclusions included hysterectomies performed by gynecologic oncologists or non-gynecologic surgeons and surgeries performed outside the UMass healthcare system. Chart abstraction included demographics; pre-hysterectomy evaluation, including current cervical cytology, pathologic endometrial assessment (biopsy, dilation and curettage), and imaging (ultrasound, MRI, CT scan, sonohysterogram, or hysteroscopy); intraoperative factors; and final diagnosis. Results: Analytic cohort included 2,332 women undergoing hysterectomy with 396 (17.0%) including use of morcellation. The malignancy incidence on final pathology was 2.1% and was different between non-morcellated versus morcellated specimens (2.5% vs. 0.3%, p Conclusion: The incidence of malignancy at time ofhysterectomy performed by non­-oncology trained gynecologists was 2.1% overall, and 0.3% in morcellated cases. The pre-operative evaluation of patients undergoing hysterectomy with morcellation is similar to those without morcellation, except for lower rates of pathologic endometrial assessment. An argument could be made that a pathology assessment is indicated in this group due to risk of dissemination in the case of occult malignancy. The risk of occult malignancy is rare, but this should be discussed with patients and taken into account during the pre-operative evaluation

    A Cytosolic Multiprotein Complex Containing p85\u3cem\u3eα\u3c/em\u3e is Required for \u3cem\u3eβ\u3c/em\u3e-Catenin Activation in Colitis and Colitis-Associated Cancer

    Wnt/β-catenin signaling is required for crypt structure maintenance. We previously observed nuclear accumulation of Ser-552 phosphorylated β-catenin (pβ-CatSer-552) in intestinal epithelial cells (IEC) during colitis and colitis-associated cancer. Data here delineate a novel multiprotein cytosolic complex (MCC) involved in β-catenin signaling in the intestine. The MCC contains p85α, the class IA subunit of PI3K, along with β-catenin, 14-3-3ζ, Akt, and p110α. MCC levels in IEC increase in colitis and colitis-associated cancer patients. IEC-specific p85α-deficient (p85ΔIEC) mice develop more severe dextran sodium sulfate colitis due to delayed ulcer healing and reduced epithelial β-catenin activation. In colonic IEC, p85α deficiency did not alter PI3K signaling. In vitro shRNA depletion of individual complex members disrupts the MCC and reduces β-catenin signaling. Despite worse colitis, p85ΔIEC mice have reduced tumor burden after azoxymethane/dextran sodium sulfate treatment. Together the data indicate that the β-catenin MCC is needed for mucosal repair and carcinogenesis. This novel MCC may be an attractive therapeutic target in preventing cancer in colitis patients
