    Linalool Acts As A Fast And Reversible Anesthetic In Hydra

    The ability to make transgenic Hydra lines has allowed for quantitative in vivo studies of Hydra regeneration and physiology. These studies commonly include excision, grafting and transplantation experiments along with high-resolution imaging of live animals, which can be challenging due to the animal’s response to touch and light stimuli. While various anesthetics have been used in Hydra studies, they tend to be toxic over the course of a few hours or their long-term effects on animal health are unknown. Here, we show that the monoterpenoid alcohol linalool is a useful anesthetic for Hydra. Linalool is easy to use, non-toxic, fast acting, and reversible. It has no detectable long-term effects on cell viability or cell proliferation. We demonstrate that the same animal can be immobilized in linalool multiple times at intervals of several hours for repeated imaging over 2–3 days. This uniquely allows for in vivo imaging of dynamic processes such as head regeneration. We directly compare linalool to currently used anesthetics and show its superior performance. Linalool will be a useful tool for tissue manipulation and imaging in Hydra research in both research and teaching contexts

    Family in Rehabilitation, Empowering Carers for Improved Malnutrition Outcomes: Protocol for the FREER Pilot Study

    Interventions to improve the nutritional status of older adults and the integration of formal and family care systems are critical research areas to improve the independence and health of aging communities and are particularly relevant in the rehabilitation setting.The primary outcome aimed to determine if the FREER (Family in Rehabilitation: EmpowERing Carers for improved malnutrition outcomes) intervention in malnourished older adults during and postrehabilitation improve nutritional status, physical function, quality of life, service satisfaction, and hospital and aged care admission rates up to 3 months postdischarge, compared with usual care. Secondary outcomes evaluated include family carer burden, carer services satisfaction, and patient and carer experiences. This pilot study will also assess feasibility and intervention fidelity to inform a larger randomized controlled trial.This protocol is for a mixed-methods two-arm historically-controlled prospective pilot study intervention. The historical control group has 30 participants, and the pilot intervention group aims to recruit 30 patient-carer pairs. The FREER intervention delivers nutrition counseling during rehabilitation, 3 months of postdischarge telehealth follow-up, and provides supportive resources using a novel model of patient-centered and carer-centered nutrition care. The primary outcome is nutritional status measured by the Scored Patient-Generated Subjective Global Assessment Score. Qualitative outcomes such as experiences and perceptions of value will be measured using semistructured interviews followed by thematic analysis.     Linking Use of Ship Channels by West Indian Manatees (Trichechus manatus) to Seasonal Migration and Habitat Use

    Research on marine mammal occurrence in ship channels often focuses on large cetaceans in offshore shipping routes, while nearshore research largely addresses small vessel strikes. Marine mammals, such as the West Indian manatee, that reside in or migrate through nearshore areas, have potential to travel through a wide range of channel types, encountering a greater diversity of vessels than previously recognized. We tested the extent and conditions of ship channel use by manatees along the north-central Gulf of Mexico (nGoM) coast by combining data from telemetry-tracked individuals, opportunistic citizen-sourced sightings, and environmental attributes linked to manatee movements. Manatees used both nearshore boat channels (130 and 300 m wide) and open water fairways but used nearshore channels much more frequently, consistent with habitat requirements. Satellite-tracked individuals swam faster and moved more directly in all channel types, indicating use of these channels as migratory and travel corridors. Accordingly, generalized additive models revealed that manatees used channels most often during spring/early summer and fall and at temperatures coincidental with entry to and exit from the nGoM during migration. Manatees also occurred in ship channels when freshwater discharges were low, likely because timing of peak manatee occurrence in the nGoM coincides with seasonally low discharge periods. Expanding shipping activity world-wide is likely to increase interactions between marine mammals and a variety of vessel types, and these effects may be particularly impactful to migratory animals like manatees that use nearshore habitats at the interface of recreational boating and commercial shipping. Linking near- and offshore ship channel use to migration and habitat use will better aid risk-assessment for vessel collision and other shipping related activities for migratory marine species globally

    Decoration of plasmonic Mg nanoparticles by partial galvanic replacement.

    Plasmonic structures have attracted much interest in science and engineering disciplines, exploring a myriad of potential applications owing to their strong light-matter interactions. Recently, the plasmonic concentration of energy in subwavelength volumes has been used to initiate chemical reactions, for instance by combining plasmonic materials with catalytic metals. In this work, we demonstrate that plasmonic nanoparticles of earth-abundant Mg can undergo galvanic replacement in a nonaqueous solvent to produce decorated structures. This method yields bimetallic architectures where partially oxidized 200-300 nm Mg nanoplates and nanorods support many smaller Au, Ag, Pd, or Fe nanoparticles, with potential for a stepwise process introducing multiple decoration compositions on a single Mg particle. We investigated this mechanism by electron-beam imaging and local composition mapping with energy-dispersive X-ray spectroscopy as well as, at the ensemble level, by inductively coupled plasma mass spectrometry. High-resolution scanning transmission electron microscopy further supported the bimetallic nature of the particles and provided details of the interface geometry, which includes a Mg oxide separation layer between Mg and the other metal. Depending on the composition of the metallic decorations, strong plasmonic optical signals characteristic of plasmon resonances were observed in the bulk with ultraviolet-visible spectrometry and at the single particle level with darkfield scattering. These novel bimetallic and multimetallic designs open up an exciting array of applications where one or multiple plasmonic structures could interact in the near-field of earth-abundant Mg and couple with catalytic nanoparticles for applications in sensing and plasmon-assisted catalysis.Support for this project was provided by the EU Framework Programme for Research and Innovation Horizon 2020 (Starting Grant SPECs 804523).     Tektin 2 is required for central spindle microtubule organization and the completion of cytokinesis

    During anaphase, the nonkinetochore microtubules in the spindle midzone become compacted into the central spindle, a structure which is required to both initiate and complete cytokinesis. We show that Tektin 2 (Tek2) associates with the spindle poles throughout mitosis, organizes the spindle midzone microtubules during anaphase, and assembles into the midbody matrix surrounding the compacted midzone microtubules during cytokinesis. Tek2 small interfering RNA (siRNA) disrupts central spindle organization and proper localization of MKLP1, PRC1, and Aurora B to the midzone and prevents the formation of a midbody matrix. Video microscopy revealed that loss of Tek2 results in binucleate cell formation by aberrant fusion of daughter cells after cytokinesis. Although a myosin II inhibitor, blebbistatin, prevents actin-myosin contractility, the microtubules of the central spindle are compacted. Strikingly, Tek2 siRNA abolishes this actin-myosin–independent midzone microtubule compaction. Thus, Tek2-dependent organization of the central spindle during anaphase is essential for proper midbody formation and the segregation of daughter cells after cytokinesis

    Recently, Neubert has suggested that a certain class of nonperturbative corrections dominates the shape of the electron spectrum in the endpoint region of semileptonic BB decay. Perturbative QCD corrections are important in the endpoint region. We study the effects of these corrections on Neubert's proposal.     Mixed-Methods Evaluation of Real-Time Safety Reporting by Hospitalized Patients and Their Care Partners:The MySafeCare Application

    OBJECTIVE: To evaluate the amount and content of data patients and carepartners reported using a real-time electronic safety tool compared to other reporting mechanisms, and understand their perspectives on safety concerns and reporting in the hospital. METHODS: Mixed-methods study including 20 month pre- and post-implementation trial evaluating MySafeCare, a web-based application which allows hospitalized patients/carepartners to report safety concerns in real-time. Comparison of MySafeCare submission rates for three hospital units (oncology acute care; vascular intermediate care; medical intensive care) to submissions rates of Patient Family Relations (PFR) Department, a hospital service to address patient/family concerns. Triangulation of quantitative data with thematic analysis of safety concern submissions and patient/carepartner interviews to understand submission content and perspectives on safety reporting. RESULTS: Thirty-two MySafeCare submissions were received with an average rate of 1.7 submissions per 1,000 patient-days and a range of 0.3 to 4.8 submissions per 1,000 patient-days across all units, indicating notable variation between units. MySafeCare submission rates were significantly higher than PFR submission rates during the post-intervention period on the vascular unit (4.3 [95% CI 2.8 – 6.5] versus 1.5 [95% CI 0.7 – 3.1], Poisson) (P=0.01). Overall trends indicated a decrease in PFR submissions after MySafeCare implementation. Triangulated data indicated patients preferred to report anonymously and did not want concerns submitted directly to their care team. CONCLUSIONS: MySafeCare evaluation confirmed the potential value of providing an electronic, anonymous reporting tool in the hospital to capture safety concerns in real-time. Such applications should be tested further as part of patient safety programs
