    Seasonal variation of macrolide resistance gene abundances in the South Fork Iowa River Watershed

    The Midwestern United States is dominated by agricultural production with high concentrations of swine, leading to application of swine manure onto lands with artificial subsurface drainage. Previous reports have indicated elevated levels of antibiotic resistance genes (ARGs) in surface water and groundwater around confined animal feeding operations which administer antimicrobials. While previous studies have examined the occurrence of ARGs around confined swine feeding operations, little information is known how their transport from tile-drained fields receiving swine manure application impacts downstream environments. To further our knowledge in this area, water samples were collected from five locations in the agriculturally dominated South Fork Iowa River Watershed with approximately 840,000 swine present in the 76,000 ha basin. Samples were collected monthly from three stream sites and two main artificial subsurface drainage outlets. Samples were analyzed for macrolide resistance genes ermB, ermF and 16S rRNAgene abundance using qPCR. Abundance of erm genes ranged from below limits of quantification to \u3e 107 copies 100 mL− 1 water. Eighty-nine percent of stream water samples contained one of these two ARGs. Results indicate significantly more ermB and ermF in main drainage outlets than stream samples when normalized by 16S rRNA abundance (p \u3c 0.0001). Both artificial drainage locations revealed temporal trends for ermB and ermF abundance when normalized to 16S rRNA abundance. The higher resistance gene concentrations identified in artificial drainage samples occurring mid-Spring and late-Fall are likely due to manure application

    Short-term dispersal of Fukushima-derived radionuclides off Japan : modeling efforts and model-data intercomparison

The definitive version was published in Biogeosciences 10 (2013): 4973-4990, doi:10.5194/bg-10-4973-2013.The Great East Japan Earthquake and tsunami that caused a loss of power at the Fukushima nuclear power plants (FNPP) resulted in emission of radioactive isotopes into the atmosphere and the ocean. In June of 2011, an international survey measuring a variety of radionuclide isotopes, including 137Cs, was conducted in surface and subsurface waters off Japan. This paper presents the results of numerical simulations specifically aimed at interpreting these observations and investigating the spread of Fukushima-derived radionuclides off the coast of Japan and into the greater Pacific Ocean. Together, the simulations and observations allow us to study the dominant mechanisms governing this process, and to estimate the total amount of radionuclides in discharged coolant waters and atmospheric airborne radionuclide fallout. The numerical simulations are based on two different ocean circulation models, one inferred from AVISO altimetry and NCEP/NCAR reanalysis wind stress, and the second generated numerically by the NCOM model. Our simulations determine that > 95% of 137Cs remaining in the water within ~600 km of Fukushima, Japan in mid-June 2011 was due to the direct oceanic discharge. The estimated strength of the oceanic source is 16.2 ± 1.6 PBq, based on minimizing the model-data mismatch. We cannot make an accurate estimate for the atmospheric source strength since most of the fallout cesium had left the survey area by mid-June. The model explained several key features of the observed 137Cs distribution. First, the absence of 137Cs at the southernmost stations is attributed to the Kuroshio Current acting as a transport barrier against the southward progression of 137Cs. Second, the largest 137Cs concentrations were associated with a semi-permanent eddy that entrained 137Cs-rich waters, collecting and stirring them around the eddy perimeter. Finally, the intermediate 137Cs concentrations at the westernmost stations are attributed to younger, and therefore less Cs-rich, coolant waters that continued to leak from the reactor in June of that year.Irina Rypina was partially supported by Grant NSF-OCE-0725796.     Chandra Observation of the Cluster Environment of a WAT Radio Source in Abell 1446

    Wide-angle tail (WAT) radio sources are often found in the centers of galaxy clusters where intracluster medium (ICM) ram pressure may bend the lobes into their characteristic C-shape. We examine the low redshift (z=0.1035) cluster Abell 1446, host to the WAT radio source 1159+583. The cluster exhibits possible evidence for a small-scale cluster-subcluster merger as a cause of the WAT radio source morphology. This evidence includes the presence of temperature and pressure substructure along the line that bisects the WAT as well as a possible wake of stripped interstellar material or a disrupted cool core to the southeast of the host galaxy. A filament to the north may represent cool, infalling gas that's contributing to the WAT bending while spectroscopically determined redshifts of member galaxies may indicate some component of a merger occurring along the line-of-sight. The WAT model of high flow velocity and low lobe density is examined as another scenario for the bending of 1159+583. It has been argued that such a model would allow the ram pressure due to the galaxy's slow motion through the ICM to shape the WAT source. A temperature profile shows that the cluster is isothermal (kT= 4.0 keV) in a series of annuli reaching a radius of 400 kpc. There is no evidence of an ongoing cooling flow.     Rapid eye movement (REM) phasic activity refers to brief events that occur in periods of REM sleep, such as individual eye movements (EMs). REM density (RD) is the bestknown measure of such activity, although reports of RD differences among normal, depressed, and schizophrenic subjects have been equivocal. RD is a measure with a large variability, and its physiological substrate is not known. We sought a more consistent measure which might also suggest the underlying physiology. Using the time intervals between individual EMs, we calculated empirical probability distributions which showed that EMs fell into two subgroups or states: "burst" and "isolated". Then, a novel Markov chain model of sequential transition between the states was calculated for nine normal, eight schizophrenic, and seven depressed male veterans. A significantly higher probability of remaining in the burst state was observed in both patient groups. The actual number of EMs in the isolated state was nearly identical in the three groups.     Subtropical mode water variability in a climatologically forced model in the northwestern Pacific Ocean

Despite the use of annually repeating atmospheric forcing, significant interannual to decadal variability is evident in the volume, temperature, and age of STMW formed in the region. This long time-scale variability is intrinsic to the ocean. The formation and characteristics of STMW are comparable to those observed in nature. STMW is found to be cooler, denser, and shallower in the east than in the west, but time variations in these properties are generally correlated across the full water mass. Formation is found to occur south of the Kuroshio Extension, and after formation STMW is advected westward, as shown by the transport streamfunction. The ideal age and chlorofluorocarbon tracers are used to analyze the life cycle of STMW. Over the full model run, the average age of STMW is found to be 4.1 yr, but there is strong geographical variation in this, from an average age of 3.0 yr in the east to 4.9 yr in the west. This is further evidence that STMW is formed in the east and travels to the west. This is qualitatively confirmed through simulated dye experiments known as transit-time distributions. Changes in STMW formation are correlated with a large meander in the path of the Kuroshio south of Japan. In the model, the large meander inhibits STMW formation just south of Japan, but the export of water with low potential vorticity leads to formation of STMW in the east and an overall increase in volume. This is correlated with an increase in the outcrop area of STMW.     Mitochondrial abnormalities in ageing macular photoreceptors

    PURPOSE. To evaluate somatic mitochondrial (mt)DNA mutations in the macula during ageing. METHODS. Ten 30-m cryostat sections from the macula (foveal and perifoveal regions) and peripheral retina of 14 donors (aged 14 -94 years) were cut for cytochrome c oxidase cytochemistry. The photoreceptor layer was microdissected and DNA extracted for 4977-bp mtDNA (mtDNA 4977 ) quantification using PCR. Dual cytochemistry for cytochrome c oxidase and succinate dehydrogenase allowed the detection of cytochrome c oxidase-deficient cones. RESULTS. Findings showed a progressive accumulation of mtDNA 4977 from ages 14 to 94 years. From ages 14 to 60 years there was an increase from 0.006% to 0.25%, and from ages 60 to 94 years there was a steeper increase from 0.25% to 5.39%. Counts of cones in the dual-reacted preparations showed more cytochrome c oxidase-deficient cones in the foveal region than elsewhere. CONCLUSIONS. The results show that mitochondrial DNA deletions and cytochrome c oxidase-deficient cones accumulate in the ageing retina, particularly in the foveal region. These defects may contribute to the changes in macular function observed in ageing and age-related maculopathy. Morphologic changes accompanying this ageing process primarily involve the photoreceptors and the cells of the retinal pigment epithelium (RPE). This is manifested by atrophy and loss of both cell types, depigmentation and hyperpigmentation of the RPE, a progressive accumulation of lipofuscin, drusen formation, thickening of Bruch's membrane, and the appearance of basal laminar deposits

    HIV Testing of At Risk Patients in a Large Integrated Health Care System

    OBJECTIVE: Early identification of HIV infection is critical for patients to receive life-prolonging treatment and risk-reduction counseling. Understanding HIV screening practices and barriers to HIV testing is an important prelude to designing successful HIV screening programs. Our objective was to evaluate current practice patterns for identification of HIV. METHODS: We used a retrospective cohort analysis of 13,991 at-risk patients seen at 4 large Department of Veterans Affairs (VA) health-care systems. We also reviewed 1,100 medical records of tested patients. We assessed HIV testing rates among at-risk patients, the rationale for HIV testing, and predictors of HIV testing and of HIV infection. RESULTS: Of the 13,991 patients at risk for HIV, only 36% had been HIV-tested. The prevalence of HIV ranged from 1% to 20% among tested patients at the 4 sites. Approximately 90% of patients who were tested had a documented reason for testing. CONCLUSION: One-half to two-thirds of patients at risk for HIV had not been tested within our selected VA sites. Among tested patients, the rationale for HIV testing was well documented. Further testing of at-risk patients could clearly benefit patients who have unidentified HIV infection by providing earlier access to life-prolonging therapy
