    TERT promoter mutations are highly recurrent in SHH subgroup medulloblastoma

    Telomerase reverse transcriptase (TERT) promoter mutations were recently shown to drive telomerase activity in various cancer types, including medulloblastoma. However, the clinical and biological implications of TERT mutations in medulloblastoma have not been described. Hence, we sought to describe these mutations and their impact in a subgroup-specific manner. We analyzed the TERT promoter by direct sequencing and genotyping in 466 medulloblastomas. The mutational distributions were determined according to subgroup affiliation, demographics, and clinical, prognostic, and molecular features. Integrated genomics approaches were used to identify specific somatic copy number alterations in TERT promoter-mutated and wild-type tumors. Overall, TERT promoter mutations were identified in 21 % of medulloblastomas. Strikingly, the highest frequencies of TERT mutations were observed in SHH (83 %; 55/66) and WNT (31 %; 4/13) medulloblastomas derived from adult patients.     Abstract Controlling the crystallization of Metal–Organic Frameworks (MOFs) at the nanoscale is currently challenging, and this hinders their utilization for multiple applications including photo(electro)chemistry and sensors. In this work, we show a synthetic protocol that enables the preparation of highly homogeneous Cu2O@MOF nanowires standing on a conductive support with extensive control over the crystallization of the MOF nanoparticles at the surface of the Cu2O nanowires. Cu2O nanowires were first prepared via templated electrodeposition, and then partially converted into the well-known Cu-MOF HKUST-1 by pulsed electrochemical oxidation. We show that the use of PVP as a capping agent during the electrochemical oxidation of Cu2O into HKUST-1 provides control over the growth of the MOF nanocrystals on the surface of the Cu2O nanowires, and that the size of the MOF crystals obtained can be tuned by changing the concentration of PVP dissolved in the electrolyte. In addition, we propose the use of benzoic acid as an alternative to achieve control over the size of the obtained MOF nanocrystals when the use of a capping agent should be avoided

    Genetic and Epigenetic Inactivation of Kruppel-like Factor 4 in Medulloblastoma1

    Although medulloblastoma is the most common pediatric malignant brain tumor, its molecular underpinnings are largely unknown. We have identified rare, recurrent homozygous deletions of Kruppel-like Factor 4 (KLF4) in medulloblastoma using high-resolution single nucleotide polymorphism arrays, digital karyotyping, and genomic real-time polymerase chain reaction (PCR). Furthermore, we show that there is loss of physiological KLF4 expression in more than 40% of primary medulloblastomas both at the RNA and protein levels. Medulloblastoma cell lines drastically increase the expression of KLF4 in response to the demethylating agent 5-azacytidine and demonstrate dense methylation of the promoter CpG island by bisulfite sequencing. Methylation-specific PCR targeting the KLF4 promoter demonstrates CpG methylation in approximately 16% of primary medulloblastomas. Reexpression of KLF4 in the D283 medulloblastoma cell line results in significant growth suppression both in vitro and in vivo. We conclude that KLF4 is inactivated by either genetic or epigenetic mechanisms in a large subset of medulloblastomas and that it likely functions as a tumor suppressor gene in the pathogenesis of medulloblastoma

    Paulus als interkultureller Vermittler: Wie der Jude Paulus als Christusgläubiger sein Evangelium unter den Menschen aus den Völkern verbreitete. Der als Jude geborene Apostel Paulus sah sich berufen, das Evangelium der Auferstehung Christi unter den Völkern zu vermitteln. Die vorliegende kulturwissenschaftlich geprägte Studie zeigt auf, dass und in welcher Weise Paulus seine bikulturelle Persönlichkeit einsetzte, um die Menschen aus den Völkern für seine Version des Evangeliums von Jesus Christus zu gewinnen. Im Fokus der Untersuchung zu Paulus als Vermittler in einem Kulturtransfergeschehen stehen die paulinischen Selbstbeschreibungen, insbesondere deren „Spitzensätze“ (1 Kor 9,19–23) sowie als beispielhafte Manifestation seiner Adaptabilität die Selbstdarstellung als Wettkämpfer (1 Kor 9,24–27)
