Specific label-free and real-time detection of oxidized low density lipoprotein (oxLDL) using an immunosensor with three monoclonal antibodies
Increased levels of plasma oxLDL, which is the oxidized fraction of Low Density Lipoprotein (LDL), are
associated with atherosclerosis, an inflammatory disease, and the subsequent development of severe
cardiovascular diseases that are today a major cause of death in modern countries. It is therefore
important to find a reliable and fast assay to determine oxLDL in serum. A new immunosensor
employing three monoclonal antibodies (mAbs) against oxLDL is proposed in this work as a quick and
effective way to monitor oxLDL. The oxLDL was first employed to produce anti-oxLDL monoclonal
antibodies by hybridoma cells that were previously obtained. The immunosensor was set-up by selfassembling
cysteamine (Cyst) on a gold (Au) layer (4 mm diameter) of a disposable screen-printed
electrode. Three mAbs were allowed to react with N-hydroxysuccinimide (NHS) and
ethyl(dimethylaminopropyl)carbodiimide (EDAC), and subsequently incubated in the Au/Cys. Albumin
from bovine serum (BSA) was immobilized further to ensure that other molecules apart from oxLDL
could not bind to the electrode surface. All steps were followed by various characterization techniques
such as electrochemical impedance spectroscopy (EIS) and square wave voltammetry (SWV). The
analytical operation of the immunosensor was obtained by incubating the sensing layer of the device in
oxLDL for 15 minutes, prior to EIS and SWV. This was done by using standard oxLDL solutions prepared
in foetal calf serum, in order to simulate patient's plasma with circulating oxLDL. A sensitive response
was observed from 0.5 to 18.0 mg mL 1
. The device was successfully applied to determine the oxLDL
fraction in real serum, without prior dilution or necessary chemical treatment. The use of multiple
monoclonal antibodies on a biosensing platform seemed to be a successful approach to produce a
specific response towards a complex multi-analyte target, correlating well with the level of oxLDL within
atherosclerosis disease, in a simple, fast and cheap way

Identification and chromosomal localization of one locus of Leishmania (L.) major related with resistance to itraconazole
Ergosterol is an important compound responsible to maintain integrity and fluidity of Leishmania spp. membranes. Starting from an overexpression/selection method, our group has isolated and mapped nine different loci of Leishmania (L.) major related to resistance against two inhibitors of the ergosterol biosynthesis pathway, terbinafine (TBF) and itraconazole (ITZ). Individual functional analysis after overexpression induction of these loci in the presence of TBF and/or ITZ [or the ITZ analog ketoconazole (CTZ)] have shown low but significant levels of resistance after transfection into L. major wild-type parasites. In this work, we have shown the insert mapping and chromosomal identification of one of these loci (cosItz2). Functional analysis experiments associated with chromosomal localization by comparison at genomic database allowed us to identify two prospective gene-protein systems not related to the ergosterol biosynthesis and capable to confer wild-type cells resistance to ITZ-CTZ after transfection. Efficacy of the tubercidin antileishmania action associated with an inhibitor of the nucleoside transport
Tubercidin (TUB) is an adenosine analog with potent antiparasite action, unfortunately associated with severe host toxicity. Prevention of TUB toxicity can be reached associating nitrobenzylthioinosine (NBMPR), an inhibitor of the purine nucleoside transport, specifically target to the mammal cells. It was demonstrated that this nucleoside transport inhibitor has no significant effect in the in vitro uptake of TUB by Schistosoma mansoni and Trypanosoma gambiense. Seeking to evaluate if the association of these compounds is also effective against leishmania, we analyzed the TUB-NBMPR combined treatment in in vitro cultures of promastigote forms of Leishmania (L.) amazonensis, Leishmania (L.) chagasi, Leishmania (L.) major, and Leishmania (V.) braziliensis as well as in cultures of amastigote forms of L. (L.) amazonensis, mice macrophages infected with L. (L.) amazonensis, and in vivo tests in BALB/c mice infected with L. (L.) amazonensis. Lung tissue mechanics in the early stages of induced paracoccidioidomycosis in rats
Pulmonary dysfunction represents the most important cause of death in patients with paracoccidioidomycosis (PBM). In order to investigate the functional changes of the lungs in the early stages of PBM, a model of benign disease was developed by intratracheal challenge of 12-week old isogenic Wistar rats with 1 x 106 yeast forms of Paracoccidioides brasiliensis. Animals were studied 30 and 60 days after infection, when fully developed granulomas were demonstrable in the lungs. Measurements of airway resistance, lung elastance and tissue hysteresis were made during sinusoidal deformations (100 breaths/min, tidal volume = 2 ml) with direct measurement of alveolar pressure using the alveolar capsule technique. Infection caused a significant increase in hysteresis (infected: 1.69, N = 13; control: 1.13, N = 12, P = 0.024, ANOVA), with no alterations in airway resistance or lung elastance. Histopathological analysis revealed the presence of fully developed granulomas located in the axial compartment of the lung interstitial space. Biological activities of lignoids from Amazon Myristicaceae species: Virola michelii, V. mollissima, V. pavonis and Iryanthera juruensis
This work revisits the fruits of Iryanthera juruensis and Virola pavonis and the leaves from V. michelii, as well as describing a study of the fruits of V. mollissima. In I. juruensis aryltetraline neolignan (1) and tetrahydrofuran neolignan (2), were found while from V. pavonis neolignans of benzofuran type (6-9), the tetrahydrofuran type (2, 11-13, 17) and the biphenyl type (10), in addition to diastereoisomers of the 8.O.4'-oxyneolignan type (14 and 15) and others were isolated. The V. mollissima accumulates the aryltetralone neolignan 4 and its seco derivative (5). The lignoids 1 and 2 obtained from I. juruensis arils possess antileishmanial activity against the promastigote form of Leishmania amazonensis.O trabalho consiste na re-investigação dos frutos de Iryanthera juruensis e Virola pavonis e das folhas de V. michelii, bem como no estudo dos frutos de V. mollissima. A partir de I. juruensis foram isoladas uma neolignana ariltetralínica (1) e uma neolignana tetraidrofurânica (2), enquanto que de Virola pavonis foram isoladas neolignanas benzofurânicas (6-9), tetraidrofurânicas (2, 11-13, 17), bifenílica (10), diastereoisômeros de uma hidróxi-neolignana 8.O.4'(14-15) e outras. V. mollissima acumula a neolignana ariltetralônica 4 ou o seu derivado seco (5). As folhas de V. michelii apresentaram a ocorrência de lignanas furofurânicas (18-19). Os lignóides 1 e 2 obtidos dos arilos de I. juruensis apresentaram atividade leishmanicida contra a forma promastigota de Leishmania amazonensis
