    The NIRCam instrument on the James Webb Space Telescope will have a Lyot coronagraph for high contrast imaging of extrasolar planets and circumstellar disks at λ=2 - 5 μm. Half-tone patterns are used to create graded-transmission image plane masks. These are generated using electron beam lithography and reactive ion etching of a metal layer on an antireflection coated sapphire substrate. We report here on the manufacture and evaluation of the flight occulters

    The NIRCam instrument on the James Webb Space Telescope will provide coronagraphic imaging from λ =1-5 μm of high contrast sources such as extrasolar planets and circumstellar disks. A Lyot coronagraph with a variety of circular and wedge-shaped occulting masks and matching Lyot pupil stops will be implemented. The occulters approximate grayscale transmission profiles using halftone binary patterns comprising wavelength-sized metal dots on anti-reflection coated sapphire substrates. The mask patterns are being created in the Micro Devices Laboratory at the Jet Propulsion Laboratory using electron beam lithography. Samples of these occulters have been successfully evaluated in a coronagraphic testbed. In a separate process, the complex apertures that form the Lyot stops will be deposited onto optical wedges. The NIRCam coronagraph flight components are expected to be completed this year

    BACKGROUND: HSV-2 is the major cause of genital herpes. We previously demonstrated that the host viral restriction factor tetherin restricts HSV-2 release and is antagonized by several HSV-2 glycoproteins. However, the mechanisms underlying HSV-2 glycoproteins mediated counteraction of tetherin remain unclear. In this study, we investigated whether tetherin restricts the cell-to-cell spread of HSV-2 and the mechanisms underlying HSV-2 gD mediated antagonism of tetherin. METHODS: Infectious center assays were used to test whether tetherin could affect cell-to-cell spread of HSV-2. Coimmunoprecipitation assays were performed to map the tetherin domains required for HSV-2 gD-mediated downregulation. Immunoflurence assays were performed to detect the accumulation of tetherin in lysosomes or proteasomes. All experiments were repeated for at least three times and the data were performed statistical analysis. RESULTS: 1) Tetherin restricts cell-to-cell spread of HSV-2; 2) HSV-2 gD specifically interacts with the CC domain of tetherin; 3) HSV-2 gD promotes tetherin to the lysosomal degradation pathway. CONCLUSIONS: Tetherin not only restricts HSV-2 release but also its cell-to-cell spread. In turn, HSV-2 gD targets the CC domain of tetherin and promotes its degradation in the lysosome. Findings in this study have increased our understanding of tetherin restriction and viral countermeasures

    Investigation of the Performance of Neat and Modified Asphalt Binders

    The Superpave binder specifications are performance-based criteria for binder selection for use in hot-mix asphalt mixtures. However, these specifications may not be applicable to modified binders. This research investigates the a new method for determining hot-mix asphalt mixing and compaction temperatures as well as the performance of neat (unmodified) and modified binders of the same performance grade. The first phase of the research focuses on determining mixing and compaction temperatures for hot-mix asphalt mixtures. Modified binders are shear rate and temperature dependent and the conventional methods of determining mixing and compaction temperatures can yield extreme results. The Zero Shear Viscosity theory is used as an alternative method to determine hot-mix asphalt mixing and compaction temperatures. The results reveal that the method is applicable to determining mixing and compaction temperatures for hot-mix asphalt mixtures containing modified binders, but additional work is needed in order to make the method applicable to neat binders. The second phase of the study investigates the performance of hot-mix asphalt mixtures containing neat and modified binders. Laboratory tests were performed on similar mixtures that contained neat and modified binders. Simple performance tests, such as accelerated rutting, flexural beam fatigue, and indirect tensile indicate that modified binders may contribute to improved hot-mix asphalt resistance to various distresses. Overall, performance-graded binders of the same grade appear to offer similar performance regardless of the high temperature at which the binder meets the high temperature specification
