Pathogenicity of multiple Providencia species (Enterobacteriales: Morganellaceae) to the mass-reared Mexican fruit fly (Diptera: Tephritidae)
Mexican fruit fly (Anastrepha ludens (Loew)) (Diptera: Tephritidae) represents a major threat to fruit production in the Western Hemisphere. Sterile insect technique is used to suppress and eradicate wild populations. Success of this control method necessitates weekly production of hundreds of millions of flies, their sterilization by irradiation, and their aerial release. Diet needed to produce large fly numbers are conducive to the spread of bacteria. Pathogenic bacteria were isolated from 3 rearing facilities and from multiple sources: eggs, larvae, pupae and spent diet, and were found to include some isolates identified to the genus Providencia (Enterobacteriales: Morganellaceae). We identified 41 Providencia isolates and tested their pathogenicity to A. ludens. Based on 16s rRNA sequences, 3 groups were clustered into several species of Providencia with varying capacities to affect the Mexican fruit fly production. Isolates putatively identified as P. alcalifaciens/P. rustigianii were all pathogenic causing larval and pupal yield reduction of 46–64% and 37–57%, respectively. Among them, Providencia isolate 3006 was the most pathogenic reducing larval and pupae yield by 73 and 81%, respectively. Isolates identified as P. sneebia were not pathogenic. The final cluster, P. rettgeri/P. vermicola, were variable in pathogenicity with 3 isolates yielding like the control and the rest causing larval and pupal yield reduction of 26–53% and 23–51%, respectively. Isolates putatively identified as P. alcalifaciens/P. rustigianii were more virulent than P. rettgeri/P. vermicola. Accurate identification of species is needed to diagnose and monitor pathogenic versus nonpathogenic Providencia strains
Morganella morganii (Enterobacteriales: Enterobacteriaceae) is a lethal pathogen of Mexican fruit fly (Diptera: Tephritidae) larvae
Tephritid pests, such as the Mexican fruit fly, Anastrepha ludens (Loew), represent a major threat to fruit production worldwide. In order to control these pests, sterile insect technique is used to suppress and eradicate wild populations. For this control method to be successful, hundreds of millions of flies must be produced weekly in mass rearing facilities. The large quantity of artificial diet and close proximity of flies at various life stages allows bacteria from family Enterobacteriaceae, Bacillaceae, Pseudomonadaceae, and others to multiply and spread more easily. In this study, bacteria with a possible pathogenic effect were isolated from Mexican fruit fly eggs and dead Mexican fruit fly larvae. Two strains of bacteria associated with dead and dying larvae were identified using the 16S rRNA sequence as a species of Morganella. Further sequencing of multiple genes and the entire genomes identified both strains as Morganella morganii. Pathogenicity tests were completed to assess this bacterium as a Mexican fruit fly pathogen. Several measures of pathogenicity including effects on larval and pupal weight, adult percent emergence, and flight ability were measured for the 2 strains of Morganella compared against a control. In all cases, the presence of the Morganella strains significantly reduced all quality control measurements compared to the control. Also, at 105 colony forming units per ml or higher levels of inoculum, the presence of Morganella resulted in 100% mortality of larvae. This study illustrates that Morganella morganii is an extremely lethal pathogen of mass reared Mexican fruit flies.

Las moscas tefritidas como la mosca Mexicana de la fruta (Anastrepha ludens Loew), representan un peligro para la producciĂłn de la fruta a nivel mundial. Para controlar estas plagas, la tĂ©cnica del insecto estĂ©ril es usada para suprimir y erradicar poblaciones naturales. Para que este mĂ©todo de control tenga Ă©xito, cientos de millones de moscas deben ser producidos en plantas de crianza masiva. La gran cantidad de dieta artificial y la cercanĂa de moscas en diversos estadios de desarrollo permiten que las bacterias perteneciendo a la familia Entobacteriacea, Baciliiacea, Pseudomonadacea, y otros se multipliquen y diseminen rápidamente. En este estudio, bacterias con posibilidad de ser patogĂ©nicas fueron aislados de huevos y larvas muertas de la mosca Mexicana de la fruta. Dos cepas de bacteria aisladas de larvas muertas o moribundas fueron identificados usando secuencias de ARN ribosoma 16S como una especie de Morganella. Adicionalmente, la realizaciĂłn de secuencias de mĂşltiples genes y el genoma entero revelĂł que las dos cepas de bacteria fueron identificados como Morganella morganii. Pruebas de patogenicidad fueron completadas para determinar la patogenicidad de Morganella hacia la mosca Mexicana de la fruta. Diferentes variables de patogenicidad como el efecto sobre el peso de las larvas y pupas, porcentaje de emergencia de moscas adultas y habilidad de vuelo fueron evaluadas para las cepas de dos Morganella y comparados contra el control. En todos los casos, la presencia de cepas de Morganella redujeron significativamente las medidas de control de calidad comparado con el control. TambiĂ©n, al 105 de unidades formadoras de colonias, la presencia de Morganella resultĂł en una mortalidad del 100% de larvas. Does Foreign Exchange Reserve Decumulation Lead to Currency Appreciation?
Many developing countries have increased their foreign reserve stocks dramatically in recent years, in large part motivated by the desire for precautionary self-insurance. One of the negative consequences of large accumulations for these countries is the risk of valuation losses. In this paper we examine the implications of systematic reserve decumulation by the Czech authorities aimed at mitigating valuation losses on euro-denominated assets. The policy was explicitly not intended to influence the value of the koruna relative to the euro. Initially the timing and size of reserve sales was not predictable, eventually sales occurred on a daily basis (in three equal installments within the day). This project examines whether these reserve sales, both during the regime of discretionary timing as well as when sales occurred every day, had unintended consequences for the domestic currency. Our findings using intraday exchange rate data and time-stamped reserve sales indicate that when decumulation occurred every day these sales led to significant appreciation of the koruna. STRUCTURE OF METHYLPHEOPHORBIDE-RCI
he methanolic extract of the cyanobacterium (blue-green alga) Spirulina geitleri has been treated with methanolic acid to convert all chlorophyllous pigments to their methylpheophorbides. Fractionation of the latter from methylpheophorbide a by thin layer chromatography and high pressure liquid chromatography yielded methylpheophorbide-RCI. Its structure has been determined as 132S-hydroxy-20-chloro-methylpheophorbide a by 1H-nuclear magnetic resonance, absorption and circular dichroism spectroscopy, mass spectrometry and by partial synthesis from chlorophyll a. The pigment is isolated from Spirulina geitleri irrespective of the use or omission of chlorinated substances during the isolation procedure
Development of a custom on-line ultrasonic vapour analyzer/flowmeter for the ATLAS inner detector, with application to gaseous tracking and Cherenkov detectors
Precision sound velocity measurements can simultaneously determine binary gas
composition and flow. We have developed an analyzer with custom electronics,
currently in use in the ATLAS inner detector, with numerous potential
applications. The instrument has demonstrated ~0.3% mixture precision for
C3F8/C2F6 mixtures and < 10-4 resolution for N2/C3F8 mixtures. Moderate and
high flow versions of the instrument have demonstrated flow resolutions of +/-
2% F.S. for flows up to 250 l.min-1, and +/- 1.9% F.S. for linear flow
velocities up to 15 ms-1; the latter flow approaching that expected in the
vapour return of the thermosiphon fluorocarbon coolant recirculator being built

A Combine On-Line Acoustic Flowmeter and Fluorocarbon Coolant Mixture Analyzer for The ATLAS Silicon Tracker
An upgrade to the ATLAS silicon tracker cooling control system may require a
change from C3F8 (octafluoro-propane) to a blend containing 10-30% of C2F6
(hexafluoro-ethane) to reduce the evaporation temperature and better protect
the silicon from cumulative radiation damage with increasing LHC luminosity.
Central to this upgrade is a new acoustic instrument for the real-time
measurement of the C3F8/C2F6 mixture ratio and flow. The instrument and its
Supervisory, Control and Data Acquisition (SCADA) software are described in
this paper. The instrument has demonstrated a resolution of 3.10-3 for
C3F8/C2F6 mixtures with ~20%C2F6, and flow resolution of 2% of full scale for
mass flows up to 30gs-1. In mixtures of widely-differing molecular weight (mw),
higher mixture precision is possible: a sensitivity of < 5.10-4 to leaks of
C3F8 into the ATLAS pixel detector nitrogen envelope (mw difference 160) has
been seen. The instrument has many potential applications, including the
analysis of mixtures of hydrocarbons, vapours for semi-conductor manufacture
and anaesthesia
Label-free electrochemical monitoring of DNA ligase activity
This study presents a simple, label-free electrochemical technique for the monitoring of DNA ligase activity. DNA ligases are enzymes that catalyze joining of breaks in the backbone of DNA and are of significant scientific interest due to their essential nature in DNA metabolism and their importance to a range of molecular biological methodologies. The electrochemical behavior of DNA at mercury and some amalgam electrodes is strongly influenced by its backbone structure, allowing a perfect discrimination between DNA molecules containing or lacking free ends. This variation in electrochemical behavior has been utilized previously for a sensitive detection of DNA damage involving the sugar-phosphate backbone breakage. Here we show that the same principle can be utilized for monitoring of a reverse process, i.e., the repair of strand breaks by action of the DNA ligases. We demonstrate applications of the electrochemical technique for a distinction between ligatable and unligatable breaks in plasmid DNA using T4 DNA ligase, as well as for studies of the DNA backbone-joining activity in recombinant fragments of E. coli DNA ligase
Elastic Scattering and Total Cross-Section in p+p reactions measured by the LHC Experiment TOTEM at sqrt(s) = 7 TeV
Proton-proton elastic scattering has been measured by the TOTEM experiment at
the CERN Large Hadron Collider at TeV in special runs with the
Roman Pot detectors placed as close to the outgoing beam as seven times the
transverse beam size. The differential cross-section measurements are reported
in the |t|-range of 0.36 to 2.5 GeV^2. Extending the range of data to low t
values from 0.02 to 0.33 GeV^2,and utilizing the luminosity measurements of
CMS, the total proton-proton cross section at sqrt(s) = 7 TeV is measured to be
