    A Guy Walks into a Bar...: Exploring Clients\u27 Preferences for Humor and Ratings of Therapy Sessions

    Humor has been identified as an important factor in the establishment of relationships. This study explores the use of humor in mental health therapy and how clients’ preferences for humor impact an evaluation of the therapy session. Forty-eight individuals currently receiving mental health therapy were examined along with the use of three forms of humor: positive, negative, and instrumental. There was a significant relationship between a preference for negative humor and session evaluation scores in which the more negative humor preferred, the lower the session ratings. Although not significant, other trends were noted between self-enhancing humor and session depth, aggressive humor and session depth, and affiliative humor and positivity. Gender differences and preference for humor were also examined with men reporting higher value on negative humor than women and women reporting greater post-session arousal than men. These findings are discussed in terms of the need for further research to consider factors that may have influenced the present study’s results

    Bootstrap approximation for the exchange-correlation kernel of time-dependent density functional theory

    A new parameter-free approximation for the exchange-correlation kernel fxcf_{\rm xc} of time-dependent density functional theory is proposed. This kernel is expressed as an algorithm in which the exact Dyson equation for the response as well as a further approximate condition are solved together self-consistently leading to a simple parameter-free kernel. We apply this to the calculation of optical spectra for various small bandgap (Ge, Si, GaAs, AlN, TiO2_2, SiC), large bandgap (C, LiF, Ar, Ne) and magnetic (NiO) insulators. The calculated spectra are in very good agreement with experiment for this diverse set of materials, highlighting the universal applicability of the new kernel.Comment: 4 figures 5 page

    In vitro activation of complement and contact system by lactic acidosis.

    The activation of complement and contact systems occurs in reperfusion injuries with initial tissue hypoxia, and lactic acidosis such as mycardial infarction and birth asphyxia. The aim of our experiment was the formal proof of activation by sole lactic acidosis. Lactic acid was added to blood and plasma samples from 10 healthy volunteers. C5a and factor XIIa were measured by EIA after incubation at 37 degrees C for 1 h. Both concentrations increased (P < 0.0001 by Friedman analysis) in blood and plasma samples with increasing amount of added lactic acid. Lactic acidosis can activate C5 from the complement system and factor XII from the contact system directly, even in the absence of cellular components

    The Role of the Transcendent in Landscapes

    Studies suggest the need for landscapes of contemplation is as real as ever, despite the seeming lack of spaces designed to provide the possibility of transcendent connection. Reintroducing commonly held physical attributes and characteristics of contemplative spaces, repeated through space and time, is a fundamental and necessary way to reflect renewed interest in and need for integrated ways of knowing in the landscape. This study ultimately shows that the contemplative powers of a site exist on a spectrum and reinforces the belief that every landscape architecture project can successfully incorporate some degree of contemplative design, the individual benefits of which can be great, and as a community continue to be explored. Case studies of three sites reveal that certain combinations of criteria seem to have the potential of being more effective than others. This study contributes to the overarching goal of restorative design by addressing the user need of experiencing cosmologically aligned space, the presence of which contributes to a greater sense of place and, by extension, personal identity, orientation, perspective, and purpose. This study also adds to a postmodern understanding of contemplative space, that is, the notion of an ancient framework and its contemporary application and possible contributions to the current and future betterment of individuals and communities

    In-vitro activation of complement system by lactic acidosis in newborn and adults.

    INTRODUCTION: Complement activation occurs secondary to a variety of external stimuli. Lactic acidosis has been previously shown to activate the complement factors C3a and C5a. In the present investigation we examined the differential effect of lactic acidosis on anaphylatoxin levels in cord and adult blood. Furthermore we aimed to determine if the entire complement cascade could be activated by lactic acidosis. METHODS: Cord and adult blood samples (n = 20 each) were collected and incubated for one hour in either untreated condition or with the addition of lactate in two concentrations (5.5 mmol/l vs. 22 mmol/l). Following incubation, levels of C3a, C5a and sC5b-9, and blood gas parameters were determined. RESULTS: Anaphylatoxin (C3a and C5a) and sC5b-9 levels increased with the addition of lactate in a dose-dependent manner in cord and adult blood (C3a: 1 h, 5.5 mmo/l, 22 mmol/l: 418/498/622 microg/l in cord blood; 1010/1056/1381 microg/l in adult blood, p<0,05; similar results were found for C5a and sC5b-9). CONCLUSION: Lactic acidosis leads to an activation of the entire complement system in neonates and in adults. This activation is dose-dependent and more pronounced in adults as compared to neonates

    Putative ligand binding sites of two functionally characterized bark beetle odorant receptors

    Background: Bark beetles are major pests of conifer forests, and their behavior is primarily mediated via olfaction. Targeting the odorant receptors (ORs) may thus provide avenues towards improved pest control. Such an approach requires information on the function of ORs and their interactions with ligands, which is also essential for understanding the functional evolution of these receptors. Hence, we aimed to identify a high-quality complement of ORs from the destructive spruce bark beetle Ips typographus (Coleoptera, Curculionidae, Scolytinae) and analyze their antennal expression and phylogenetic relationships with ORs from other beetles. Using 68 biologically relevant test compounds, we next aimed to functionally characterize ecologically important ORs, using two systems for heterologous expression. Our final aim was to gain insight into the ligand-OR interaction of the functionally characterized ORs, using a combination of computational and experimental methods. Results: We annotated 73 ORs from an antennal transcriptome of I. typographus and report the functional characterization of two ORs (ItypOR46 and ItypOR49), which are responsive to single enantiomers of the common bark beetle pheromone compounds ipsenol and ipsdienol, respectively. Their responses and antennal expression correlate with the specificities, localizations, and/or abundances of olfactory sensory neurons detecting these enantiomers. We use homology modeling and molecular docking to predict their binding sites. Our models reveal a likely binding cleft lined with residues that previously have been shown to affect the responses of insect ORs. Within this cleft, the active ligands are predicted to specifically interact with residues Tyr84 and Thr205 in ItypOR46. The suggested importance of these residues in the activation by ipsenol is experimentally supported through site-directed mutagenesis and functional testing, and hydrogen bonding appears key in pheromone binding. Conclusions: The emerging insight into ligand binding in the two characterized ItypORs has a general importance for our understanding of the molecular and functional evolution of the insect OR gene family. Due to the ecological importance of the characterized receptors and widespread use of ipsenol and ipsdienol in bark beetle chemical communication, these ORs should be evaluated for their potential use in pest control and biosensors to detect bark beetle infestations

    Biosensing platform combining label-free and labelled analysis using Bloch surface waves

    Bloch surface waves (BSW) propagating at the boundary of truncated photonic crystals (1D-PC) have emerged as an attractive approach for label-free sensing in plasmon-like sensor configurations. Due to the very low losses in such dielectric thin film stacks, BSW feature very low angular resonance widths compared to the surface plasmon resonance (SPR) case. Besides label-free operation, the large field enhancement and the absence of quenching allow utilizing BSW coupled fluorescence detection to additionally sense the presence of fluorescent labels. This approach can be adapted to the case of angularly resolved resonance detection, thus giving rise to a combined label-free / labelled biosensor platform. It features a parallel analysis of multiple spots arranged as a one-dimensional array inside a microfluidic channel of a disposable chip. Application of such a combined biosensing approach to the detection of the Angiopoietin-2 cancer biomarker in buffer solutions is reported

    Acidosis activates complement system in vitro.

    We investigated the in vitro effect of different forms of acidosis (pH 7.0) on the formation of anaphylatoxins C3a and C5a. Metabolic acidosis due to addition of hydrochloric acid (10 micromol/ml blood) or lactic acid (5.5 micromol/ml) to heparin blood (N=12) caused significant activation of C3a and C5a compared to control (both p=0.002). Respiratory acidosis activated C3a (p=0.007) and C5a (p=0.003) compared to normocapnic controls. Making blood samples with lactic acidosis hypocapnic resulted in a median pH of 7.37. In this respiratory compensated metabolic acidosis, C3a and C5a were not increased. These experiments show that acidosis itself and not lactate trigger for activation of complement components C3 and C5

    Insulin-like growth factor-1 in CNS and cerebrovascular aging

    Insulin-like growth factor-1 (IGF-1) is an important anabolic hormone that decreases with age. In the past two decades, extensive research has determined that the reduction in IGF-1 is an important component of the age-related decline in cognitive function in multiple species including humans. Deficiency in circulating IGF-1 results in impairment in processing speed and deficiencies in both spatial and working memory. Replacement of IGF-1 or factors that increase IGF-1 to old animals and humans reverses many of these cognitive deficits. Despite the overwhelming evidence for IGF-1 as an important neurotrophic agent, the specific mechanisms through which IGF-1 acts have remained elusive. Recent evidence indicates that IGF-1 is both produced by and has important actions on the cerebrovasculature as well as neurons and glia. Nevertheless, the specific regulation and actions of brain- and vascular-derived IGF-1 is poorly understood. The diverse effects of IGF-1 discovered thus far reveal a complex endocrine and paracrine system essential for integrating many of the functions necessary for brain health. Identification of the mechanisms of IGF-1 actions will undoubtedly provide critical insight into regulation of brain function in general and the causes of cognitive decline with age
