Zero gravity tissue-culture laboratory
Hardware was developed for performing experiments to detect the effects that zero gravity may have on living human cells. The hardware is composed of a timelapse camera that photographs the activity of cell specimens and an experiment module in which a variety of living-cell experiments can be performed using interchangeable modules. The experiment is scheduled for the first manned Skylab mission
Muscle protein and glycogen responses to recovery from hypogravity and unloading by tail-cast suspension
Previous studies in this laboratory using the tail-bast hindlimb suspension model have shown that there are specific changes in protein and carbohydrate metabolism in the soleus muscle due to unloading. For example, 6 days of unloading caused a 27% decrease in mass and a 60% increase in glycogen content in the soleus muscle, while the extensor digitorum longus muscle was unaffected. Also, fresh tissue tyrosine and its in vitro release from the muscle are increased in the unloaded soleus, indicating that this condition causes a more negative protein balance. With these results in mind, studies to investigate the effect of hypogravity on protein and carbohydrate metabolism in a number of rat hindlimb muscles were carried out
Interaction of GABA and Excitatory Amino Acids in the Basolateral Amygdala: Role in Cardiovascular Regulation
Activation of the amygdala in rats produces cardiovascular changes that include increases in heart rate and arterial pressure as well as behavioral changes characteristic of emotional arousal. The objective of the present study was to examine the interaction of GABA and excitatory amino acid (EAA) receptors in the basolateral amygdala (BLA) in regulating cardiovascular function. Microinjection of the GABAA receptor antagonist bicuculline methiodide (BMI) or the E A A receptor agonists NMDA or AMPA into the same region of the BLA of conscious rats produced dose-related increases in heart rate and arterial pressure. Injection of the nonselective EAA receptor antagonist kynurenic acid into the BLA prevented or reversed the cardiovascular changes caused by local injection of BMI or the noncompetitive GABA antagonist picrotoxin. Conversely, local pretreatment with the glutamate reuptake inhibitorl-trans-pyrrolidine-2,4-dicarboxylic acid enhanced the effects of intra-amygdalar injection of BMI. The cardiovascular effects of BMI were also attenuated by injection of either the NMDA antagonist 3-(2-carboxypiperazin-4-yl)-propyl-1-phosphonic acid (CPP) or the AMPA receptor antagonist 1,2,3,4-tetrahydro-6-nitro-2,3-dioxo-benzo[f]quinoxaline-7-sulfonamide (NBQX). When these two EAA receptor antagonists were combined, their ability to suppress BMI-induced tachycardic and pressor responses was additive. These findings indicate that the cardiovascular effects caused by blockade of GABAergic inhibition in the BLA of the rat are dependent on activation of local NMDA and AMPA receptors
CD4+ T cell hyporesponsiveness after repeated exposure to Schistosoma mansoni larvae is dependent upon interleukin-10
The effect that multiple percutaneous exposures to Schistosoma larvae has on the development of early CD4+ lymphocyte reactivity is unclear, yet it is important in the context of humans living in areas where schistosomiasis is endemic. In a murine model of multiple infections, we show that exposure of mice to repeated doses (4×) of Schistosoma mansoni cercariae, compared to a single dose (1×), results in CD4+ T cell hyporesponsiveness within the skin-draining lymph nodes (sdLN), manifested as reduced CD4+ cell proliferation and cytokine production. FoxP3+ CD4+ regulatory T cells were present in similar numbers in the sdLN of 4× and 1× mice and thus are unlikely to have a role in effecting hyporesponsiveness. Moreover, anergy of the CD4+ cell population from 4× mice was slight, as proliferation was only partly circumvented through the in vitro addition of exogenous interleukin-2 (IL-2), and the in vivo blockade of the regulatory molecule PD1 had a minimal effect on restoring responsiveness. In contrast, IL-10 was observed to be critical in mediating hyporesponsiveness, as CD4+ cells from the sdLN of 4× mice deficient for IL-10 were readily able to proliferate, unlike those from 4× wild-type cohorts. CD4+ cells from the sdLN of 4× mice exhibited higher levels of apoptosis and cell death, but in the absence of IL-10, there was significantly less cell death. Combined, our data show that IL-10 is a key factor in the development of CD4+ T cell hyporesponsiveness after repeated parasite exposure involving CD4+ cell apoptosis
Testing a Simplified Version of Einstein's Equations for Numerical Relativity
Solving dynamical problems in general relativity requires the full machinery
of numerical relativity. Wilson has proposed a simpler but approximate scheme
for systems near equilibrium, like binary neutron stars. We test the scheme on
isolated, rapidly rotating, relativistic stars. Since these objects are in
equilibrium, it is crucial that the approximation work well if we are to
believe its predictions for more complicated systems like binaries. Our results

Large Magellanic Cloud Microlensing Optical Depth with Imperfect Event Selection
I present a new analysis of the MACHO Project 5.7 year Large Magellanic Cloud
(LMC) microlensing data set that incorporates the effects of contamination of
the microlensing event sample by variable stars. Photometric monitoring of
MACHO LMC microlensing event candidates by the EROS and OGLE groups has
revealed that one of these events is likely to be a variable star, while
additional data has confirmed that many of the other events are very likely to
be microlensing. This additional data on the nature of the MACHO microlensing
candidates is incorporated into a simple likelihood analysis to derive a
probability distribution for the number of MACHO microlens candidates that are
true microlensing events. This analysis shows that 10-12 of the 13 events that
passed the MACHO selection criteria are likely to be microlensing events, with
the other 1-3 being variable stars. This likelihood analysis is also used to
show that the main conclusions of the MACHO LMC analysis are unchanged by the
variable star contamination. The microlensing optical depth toward the LMC is =
1.0 +/- 0.3 * 10^{-7}. If this is due to microlensing by known stellar
populations, plus an additional population of lens objects in the Galactic
halo, then the new halo population would account for 16% of the mass of a
standard Galactic halo. The MACHO detection exceeds the expected background of
2 events expected from ordinary stars in standard models of the Milky Way and
LMC at the 99.98% confidence level. The background prediction is increased to 3
events if maximal disk models are assumed for both the MilkyWay and LMC, but
this model fails to account for the full signal seen by MACHO at the 99.8%

Comparing initial-data sets for binary black holes
We compare the results of constructing binary black hole initial data with
three different decompositions of the constraint equations of general
relativity. For each decomposition we compute the initial data using a
superposition of two Kerr-Schild black holes to fix the freely specifiable
data. We find that these initial-data sets differ significantly, with the ADM
energy varying by as much as 5% of the total mass. We find that all
initial-data sets currently used for evolutions might contain unphysical
gravitational radiation of the order of several percent of the total mass. This
is comparable to the amount of gravitational-wave energy observed during the
evolved collision. More astrophysically realistic initial data will require
more careful choices of the freely specifiable data and boundary conditions for
both the metric and extrinsic curvature. However, we find that the choice of
extrinsic curvature affects the resulting data sets more strongly than the
