    Effect of ageing time on consumer-perceived quality of Italian Simmental beef

    The aim of the experiment was to provide an objective indication on the optimal ageing time of meat from ItalianSimmental (IS) young bulls, the most important commercial category of the breed. The research was carried out on 10young bulls slaughtered at the average weight of 688 kg (SE: 7.3 kg). The right side of each carcass was stored for 7days and the left one for further 7 days, at normal refrigeration temperatures. At the end of the ageing period, the sideswere sampled at the loin for meat pH, color, cooking loss and shear force measurement. A central location, affective testinvolving 74 consumers was carried out to measure the degree of liking for grilled beef aged 7 or 14 days.The pH and color parameters were not significantly affected by the duration of post-mortem storage, while cooking lossincreased (31.4 vs 32.6 %; P<0.05) and shear force values diminished (56.7 vs 46.8 N; P< 0.01) when ageing timeincreased from 7 to 14 days. The degree of liking by the untrained respondents for the two types of cooked meat wasdifferent, considering that consumers gave the highest hedonistic scores to beef aged 14 days. Tenderness was the sensoryattribute that allowed the best discrimination between beef stored for periods of different length. In fact, while thedifference between 14-day-aged and 7-day-aged beef for flavour ratings (7.0 vs 6.9) did not reach the threshold of significance,the former meat was perceived as significantly finer than the latter regarding tenderness (6.5 vs 5.6; P= 0.01)and marginally preferred in overall terms (6.9 vs 6.5; P= 0.07). The classification test, carried out at the end of the quantitativetest by asking consumers to select from a list the most appropriate attributes describing the stimuli associatedwith meat consumption, confirmed the effectiveness of texture attributes in ranking different-aged beef acceptability. Infact, the number of ticks reported for chewiness and juiciness descriptors were different for the two types of meat: the14-day-aged beef was perceived as easier to chew (57 vs 34 ticks) and juicier (42 vs 24) than the shortest-aged beef.Ageing for 14 days should be recommended as a process control point for the beef industry to improve consumer acceptanceof IS young bull beef

    Serum thyroid hormone evaluation during transition periods in dairy cows

    Abstract. Thirty-five multiparous Holstein dairy cows were selected from a high-producing dairy farm in northeastern Italy: 16 in second lactation (L2), 10 in third lactation (L3) and 9 in fourth lactation (L4). Blood sampling was carried out 7 ± 5 days before calving (Pre/C) and 7 ± 5 days after calving (Post/C). Serum triiodothyronine (T3), thyroxine (T4) and a thyroid-stimulating hormone (TSH) were assessed. Two-way analysis of variance (ANOVA) showed statistically significant effects of class (L2, L3, L4) and of period (Pre/C, Post/C) on the parameters studied (T3, T4, TSH). In particular, Bonferroni's multiple comparison test showed lower values in post-calving than in the pre-calving in L2 and L3 for TSH; lower values in post-calving than in the pre-calving in L2 for T3; and lower values in post-calving than in the pre-calving in L2, L3 and L4 for T4. Our results improve the knowledge of endocrine and metabolic changes occurring in dairy cows during transition periods and may be useful to supply a new strategy for the improvement of dairy cow farm management and reproductive performance

    Abnormal Upregulation of GPR17 Receptor Contributes to Oligodendrocyte Dysfunction in SOD1 G93A Mice

    Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by progressive loss of motor neurons (MN). Importantly, MN degeneration is intimately linked to oligodendrocyte dysfunction and impaired capacity of oligodendrocyte precursor cells (OPCs) to regenerate the myelin sheath enwrapping and protecting neuronal axons. Thus, improving OPC reparative abilities represents an innovative approach to counteract MN loss. A pivotal regulator of OPC maturation is the P2Y-like G protein-coupled receptor 17 (GPR17), whose role in ALS has never been investigated. In other models of neurodegeneration, an abnormal increase of GPR17 has been invariably associated to myelin defects and its pharmacological manipulation succeeded in restoring endogenous remyelination. Here, we analyzed GPR17 alterations in the SOD1G93A ALS mouse model and assessed in vitro whether this receptor could be targeted to correct oligodendrocyte alterations. Western-blot and immunohistochemical analyses showed that GPR17 protein levels are significantly increased in spinal cord of ALS mice at pre-symptomatic stage; this alteration is exacerbated at late symptomatic phases. Concomitantly, mature oligodendrocytes degenerate and are not successfully replaced. Moreover, OPCs isolated from spinal cord of SOD1G93A mice display defective differentiation compared to control cells, which is rescued by treatment with the GPR17 antagonist montelukast. These data open novel therapeutic perspectives for ALS management

    Vitamin B12 and homocysteine levels in blood of dairy cows during subacute ruminal acidosis

    Abstract. The aim of this study was to investigate the variations of vitamin B12 and homocysteine in blood of dairy cows during subacute ruminal acidosis (SARA). On 228 subjects ruminal liquid was collected through rumenocentesis technique and rumen pH was immediately measured by a portable pH-meter. On the basis of pH values all cows were classified (bovine class) in Group A (animals with rumen pH>5.7), Group B (animals with rumen pH between 5.6 and 5.7) and Group C (animals with rumen pH<5.6). In relation to the acidosis risk depending on the rumen pH (herd class), the herds were classified in Group 1 (normal herds: less than 33 % cows with rumen pH<5.8), Group 2 (critical herds: more than 33 % cows with rumen pH between 5.5 and 5.8) and Group 3 (acidosis herds: more than 33 % cows with rumen pH<5.5). On blood samples, collected by jugular venipuncture, vitamin B12 and homocysteine were measured by chemiluminescent immunological tests. One-way analysis of variance (ANOVA), followed by Bonferroni test, showed significant differences (P<0.05) for vitamin B12 in bovine class and significant differences (P<0.05) for homocysteine in herd class. The influence of rumen pH values resulted in adequate vitamin B12 and homocysteine levels to meet microbial and cow requirements and fatty acids modifications in dairy cows affected by SARA. Moreover, the increase of vitamin B12 could be due to the presence of analogues which interfere with the transport of the vitamin. These findings provide more information on blood modifications during SARA

    Metabolic and hormonal adaptation in Bubalus bubalis around calving and early lactation

    Pregnancy and lactation are physiological periods that result in an increased metabolic demand that, if not satisfied, could provoke a threat to homeostasis. In this study changes in the values of Non-Esterified Fatty Acids (NEFA) and β-hydroxybutyrate (BHB), glucose, insulin, thyroid hormones, milk composition and yield were investigated in buffaloes during the late pregnancy and early lactation. From a total of 50 buffaloes, blood samples were collected -7±5 days before expected calving; +7±5; +30±5 and +50±5 days post-partum; milk samples were collected at the same post-partum time points. On serum samples, the values of Non-Esterified Fatty Acids (NEFA), β-hydroxybutyrate (BHB), glucose, insulin, Triiodothyronine (T3), Thyroxine (T4) and Thyroid-stimulating hormone (TSH) were evaluated. On milk, fat %, protein %, lactose %, somatic cells score (SCS), milk yield and daily milk production (DMP) were assessed. Peripartum period significantly influenced all studied parameters (P<0.05). Milk constituents and productivity statistically changed throughout monitoring period (P<0.005). Milk yield resulted positively correlated with insulin and TSH values, negatively correlated with NEFA and BHB. Insulin was negatively correlated with lactose % and positively correlated with SCS. The obtained results showed that the peripartum period and lactation are accompanied by marked changes in some biochemical variables and in the thyroid hormones values in Italian Mediterranean Buffaloes. Moreover, the relationship found between TSH values and milk yield seems to suggest a possible role of thyroid gland on the maintenance of lactogenesis. This study underlines the importance of monitoring the hormonal status of buffalo during the transition period in order to understand when adjustments of regulatory mechanisms break through physiological limits predisposing the buffalo to metabolic problems

    The dictionary matching with gaps problem is to preprocess a dictionary DD of dd gapped patterns P1,,PdP_1,\ldots,P_d over alphabet Σ\Sigma, where each gapped pattern PiP_i is a sequence of subpatterns separated by bounded sequences of don't cares. Then, given a query text TT of length nn over alphabet Σ\Sigma, the goal is to output all locations in TT in which a pattern PiDP_i\in D, 1id1\leq i\leq d, ends. There is a renewed current interest in the gapped matching problem stemming from cyber security. In this paper we solve the problem where all patterns in the dictionary have one gap with at least α\alpha and at most β\beta don't cares, where α\alpha and β\beta are given parameters. Specifically, we show that the dictionary matching with a single gap problem can be solved in either O(dlogd+D)O(d\log d + |D|) time and O(dlogεd+D)O(d\log^{\varepsilon} d + |D|) space, and query time O(n(βα)loglogdlog2min{d,logD}+occ)O(n(\beta -\alpha )\log\log d \log ^2 \min \{ d, \log |D| \} + occ), where occocc is the number of patterns found, or preprocessing time and space: O(d2+D)O(d^2 + |D|), and query time O(n(βα)+occ)O(n(\beta -\alpha ) + occ), where occocc is the number of patterns found.     Hypoxia promotes the inflammatory response and stemness features in visceral fat stem cells from obese subjects

    Low-grade chronic inflammation is a salient feature of obesity and many associated disorders. This condition frequently occurs in central obesity and is connected to alterations of the visceral adipose tissue (AT) microenvironment. Understanding how obesity is related to inflammation may allow the development of therapeutics aimed at improving metabolic parameters in obese patients. To achieve this aim, we compared the features of 2 subpopulations of adipose-derived stem cells (ASC) isolated from both subcutaneous and visceral AT of obese patients with the features of 2 subpopulations of ASC from the same isolation sites of non-obese individuals. In particular, the behavior of ASC of obese vs non-obese subjects during hypoxia, which occurs in obese AT and is an inducer of the inflammatory response, was evaluated. Obesity deeply influenced ASC from visceral AT (obV-ASC); these cells appeared to exhibit clearly distinguishable morphology and ultrastructure as well as reduced proliferation, clonogenicity and expression of stemness, differentiation and inflammation-related genes. These cells also exhibited a deregulated response to hypoxia, which induced strong tissue-specific NF-kB activation and an NF-kB-mediated increase in inflammatory and fibrogenic responses. Moreover, obV-ASC, which showed a less stem-like phenotype, recovered stemness features after hypoxia. Our findings demonstrated the peculiar behavior of obV-ASC, their influence on the obese visceral AT microenvironment and the therapeutic potential of NF-kB inhibitors. These novel findings suggest that the deregulated hyper-responsiveness to hypoxic stimulus of ASC from visceral AT of obese subjects may contribute via paracrine mechanisms to low-grade chronic inflammation, which has been implicated in obesity-related morbidity
