    Plausible responses to the threat of rapid sea-level rise for the Thames Estuary

    This paper considers the perceptions and responses of selected stakeholders to a scenarion of rapid rise in sea-level due to the collapse of the West Antarctic ice sheet, which could produce a global rise in sea-level of 5 to 6 metres. Through a process of dialogue involving one-to one interviews and a one-day policy exercise, we addressed influences on decision-making when information is uncertain and our ability to plan, prepare for and implement effective ways of coping with this extreme scenario. Through these interactions we hoped to uncover plausible responses to the scenario and identify potential weaknesses in our current flood management approaches to dealing with such an occurrence. By undertaking this exploratory exercise we hoped to find out whether this was a feasible way to deal with such a low probability but high consequence scenario. It was the process of finding a solution that interested us rather than the technical merits of one solution over another. We were not intending to produce definitive set of recommendations on how to respond but to gain insights into the process of making a decision, specifically what influences it and what assumptions are made.Sea level rise, London

    BIOMECHANICAL VALIDATION OF A QUALITATIVE ANALYSIS OF BASEBALL PITCHING

    Correct biomechanics minimise the risk of injury and improve performance during baseball pitching. The mechanics of 20 youth pitchers were analysed during outdoor practice using digital camcorders and a checklist of kinematic and temporal elements. The pitchers were also analysed indoors with a six-camera 240Hz Motion Analysis System. In both conditions, mechanics were graded using accepted norms for youth pitchers. Kappa coefficients were calculated between the qualitative measurements and motion analysis data for 17 kinematic parameters. 11 variables showed acceptable relationships between qualitative and quantitative data, indicating the practical value of this qualitative analysis as a field tool

    Diffusion-controlled generation of a proton-motive force across a biomembrane

    Respiration in bacteria involves a sequence of energetically-coupled electron and proton transfers creating an electrochemical gradient of protons (a proton-motive force) across the inner bacterial membrane. With a simple kinetic model we analyze a redox loop mechanism of proton-motive force generation mediated by a molecular shuttle diffusing inside the membrane. This model, which includes six electron-binding and two proton-binding sites, reflects the main features of nitrate respiration in E. coli bacteria. We describe the time evolution of the proton translocation process. We find that the electron-proton electrostatic coupling on the shuttle plays a significant role in the process of energy conversion between electron and proton components. We determine the conditions where the redox loop mechanism is able to translocate protons against the transmembrane voltage gradient above 200 mV with a thermodynamic efficiency of about 37%, in the physiologically important range of temperatures from 250 to 350 K.Comment: 26 pages, 4 figures.     Breakage in the SNRPN locus in a balanced 46,XY,t(15;19) Prader-Willi syndrome patient

    A patient with Prader-Willi syndrome (PWS) was found to carry a de novo balanced reciprocal translocation, t(15;19)(q12;q13.41), which disrupted the small nuclear ribonucleoprotein N (SNRPN) locus. The translocation chromosome 15 was found to be paternal in origin. Uniparental disomy and abnormal DNA methylation were ruled out. The translocation breakpoint was found to have occurred between exon 0 (second exon) and 1 (third exon) of the SNRPN locus outside of the SmN open reading frame (ORF), which is intact. The transcriptional activities of ZNF127, IPW, PAR-1, and PAR-5 were detected with RT-PCR from fibroblasts of the patient, suggesting that these genes may not play a significant role in the PWS phenotype in this patient. Transcription from the first two exons and last seven exons of the SNRPN gene was also detected with RT-PCR; however, the complete mRNA (10 exons) was not detected. Thus, the PWS phenotype in the patient is likely to be the result of disruption of the SNRPN locus

    The effects of community interventions on unplanned healthcare use in patients with multimorbidity: a systematic review

    OBJECTIVES: To summarise the impact of community-based interventions for multimorbid patients on unplanned healthcare use. The prevalence of multimorbidity (co-existence of multiple chronic conditions) is rapidly increasing and affects one-third of the global population. Patients with multimorbidity have complex healthcare needs and greater unplanned healthcare usage. Community-based interventions allow for continued care of patients outside hospitals, but few studies have explored the effects of these interventions on unplanned healthcare usage. DESIGN: A systematic review was conducted. MEDLINE, EMBASE, PsychINFO and Cochrane Library online databases were searched. Studies were screened and underwent risk of bias assessment. Data were synthesised using narrative synthesis. SETTING: Community-based interventions. PARTICIPANTS: Patients with multimorbidity. MAIN OUTCOME MEASURES: Unplanned healthcare usage. RESULTS: Thirteen studies, including a total of 6148 participants, were included. All included studies came from high-income settings and had elderly populations. All studies measured emergency department attendances as their primary outcome. Risk of bias was generally low. Most community interventions were multifaceted with emphasis on education, self-monitoring of symptoms and regular follow-ups. Four studies looked at improved care coordination, advance care planning and palliative care. All 13 studies found a decrease in emergency department visits post-intervention with risk reduction ranging from 0 (95% confidencec interval [CI]: -0.37 to 0.37) to 0.735 (95% CI: 0.688-0.785). CONCLUSIONS: Community-based interventions have potential to reduce emergency department visits in patients with multimorbidity. Identification of specific successful components of interventions was challenging given the overlaps between interventions. Policymakers should recognise the importance of community interventions and aim to integrate aspects of these into existing healthcare structures. Future research should investigate the impact of such interventions with broader participant characteristics

    Imaging Ca2+ concentration changes at the secretory vesicle surface with a recombinant targeted cameleon

    AbstractRegulated exocytosis involves the Ca2+-triggered fusion of secretory vesicles with the plasma membrane, by activation of vesicle membrane Ca2+-binding proteins [1]. The Ca2+-binding sites of these proteins are likely to lie within 30 nm of the vesicle surface, a domain in which changes in Ca2+ concentration cannot be resolved by conventional fluorescence microscopy. A fluorescent indicator for Ca2+ called a yellow ‘cameleon’ (Ycam2) – comprising a fusion between a cyan-emitting mutant of the green fluorescent protein (GFP), calmodulin, the calmodulin-binding peptide M13 and an enhanced yellow-emitting GFP – which is targetable to specific intracellular locations, has been described [2]. Here, we generated a fusion between phogrin, a protein that is localised to secretory granule membranes [3], and Ycam2 (phogrin–Ycam2) to monitor changes in Ca2+ concentration ([Ca2+]) at the secretory vesicle surface ([Ca2+]gd) through alterations in fluorescence resonance energy transfer (FRET) between the linked cyan and yellow fluorescent proteins (CFP and YFP, respectively) in Ycam2. In both neuroendocrine PC12 and MIN6 pancreatic β cells, apparent resting values of cytosolic [Ca2+] and [Ca2+]gd were similar throughout the cell. In MIN6 cells following the activation of Ca2+ influx, the minority of vesicles that were within ∼1 μm of the plasma membrane underwent increases in [Ca2+]gd that were significantly greater than those experienced by deeper vesicles, and greater than the apparent cytosolic [Ca2+] change. The ability to image both global and compartmentalised [Ca2+] changes with recombinant targeted cameleons should extend the usefulness of these new Ca2+ probes

    Achievement goals, self-handicapping, and performance: A 2 × 2 achievement goal perspective

    Elliot and colleagues (2006) examined the effects of experimentally induced achievement goals, proposed by the trichotomous model, on self-handicapping and performance in physical education. Our study replicated and extended the work of Elliot et al. by experimentally promoting all four goals proposed by the 262 model (Elliot & McGregor, 2001), measuring the participants’ own situational achievement goals, using a relatively novel task, and testing the participants in a group setting. We used a randomized experimental design with four conditions that aimed to induce one of the four goals advanced by the 262 model. The participants (n¼138) were undergraduates who engaged in a dart-throwing task. The results pertaining to self-handicapping partly replicated Elliot and colleagues’ findings by showing that experimentally promoted performance-avoidance goals resulted in less practice. In contrast, the promotion of mastery-avoidance goals did not result in less practice compared with either of the approach goals. Dart-throwing performance did not differ among the four goal conditions. Personal achievement goals did not moderate the effects of experimentally induced goals on selfhandicapping and performance. The extent to which mastery-avoidance goals are maladaptive is discussed, as well as the interplay between personal and experimentally induced goals
