    Aeromonas Isolates from Human Diarrheic Stool and Groundwater Compared by Pulsed-Field Gel Electrophoresis

    Gastrointestinal infections of Aeromonas species are generally considered waterborne; for this reason, Aeromonas hydrophila has been placed on the United States Environmental Protection Agency Contaminant Candidate List of emerging pathogens in drinking water. In this study, we compared pulsed-field gel electrophoresis patterns of Aeromonas isolates from stool specimens of patients with diarrhea with Aeromonas isolates from patients’ drinking water. Among 2,565 diarrheic stool specimens submitted to a Wisconsin clinical reference laboratory, 17 (0.66%) tested positive for Aeromonas. Groundwater isolates of Aeromonas were obtained from private wells throughout Wisconsin and the drinking water of Aeromonas-positive patients. The analysis showed that the stool and drinking water isolates were genetically unrelated, suggesting that in this population Aeromonas gastrointestinal infections were not linked with groundwater exposures

    Emergent spatial patterns of competing benthic and pelagic algae in a river network: A parsimonious basin-scale modeling analysis

    Algae, as primary producers in riverine ecosystems, are found in two distinct habitats: benthic and pelagic algae typically prevalent in shallow/small and deep/large streams, respectively. Over an entire river continuum, spatiotemporal patterns of the two algal communities reflect specificity in habitat preference determined by geomorphic structure, hydroclimatic controls, and spatiotemporal heterogeneity in nutrient loads from point- and diffuse-sources. By representing these complex interactions between geomorphic, hydrologic, geochemical, and ecological processes, we present here a new river-network-scale dynamic model (CnANDY) for pelagic (A) and benthic (B) algae competing for energy and one limiting nutrient (phosphorus, P). We used the urbanized Weser River Basin in Germany (7th-order; ~8.4 million population; ~46 K km2) as a case study and analyzed simulations for equilibrium mass and concentrations under steady median river discharge. We also examined P, A, and B spatial patterns in four sub-basins. We found an emerging pattern characterized by scaling of P and A concentrations over stream-order ω, whereas B concentration was described by three distinct phases. Furthermore, an abrupt algal regime shift occurred in intermediate streams from B dominance in ω≤3 to exclusive A presence in ω≥6. Modeled and long-term basin-scale monitored dissolved P concentrations matched well for ω>4, and with overlapping ranges in ω<3. Power-spectral analyses for the equilibrium P, A, and B mass distributions along hydrological flow paths showed stronger clustering compared to geomorphological attributes, and longer spatial autocorrelation distance for A compared to B. We discuss the implications of our findings for advancing hydro-ecological concepts, guiding monitoring, informing management of water quality, restoring aquatic habitat, and extending CnANDY model to other river basins

    Evaluation of laboratory and on-farm tests to estimate colostrum quality for dairy cows

    The objectives of this study were to evaluate different analytical methods to determine colostrum quality in dairy cattle, including one laboratory-based method (ELISA) and 4 on-farm tests. We hypothesized that the colostral IgG concentration using different analytical methods, such as ELISA (mg/mL), digital Brix refractometer (% Brix), colostrometer (specific gravity and mg/mL), an outflow funnel (seconds), and a lateral flow assay (mg/mL), were highly correlated with the reference method, radial immunodiffusion (RID; mg/mL) and would generate comparable results. Colostrum samples were collected from 209 Holstein Friesian cows on 2 commercial dairy farms in Germany. Colostrum weight and colostrum temperature were measured. Test characteristics, such as optimum thresholds, sensitivity, specificity, and area under the curve (AUC) were determined using a receiver operating characteristic curve analyses for each test. Out of 209 colostrum samples assessed by RID, 186 (89%) samples had high quality (≥50 mg IgG/mL), while 23 colostrum samples (11%) showed poor quality with IgG concentrations less than 50 mg/mL. The mean IgG concentration (±SD) was 101.3 ± 45.9 mg/mL and the range was 6.0 to 244.3 mg/mL. The Pearson correlation coefficient (r) between RID and ELISA was r = 0.78. In comparison to RID, Pearson correlation coefficients for the on-farm tests were: r = 0.79 (digital Brix refractometry), r = 0.58 (colostrometer: specific gravity), r = 0.61 (colostrometer: temperature corrected), r = 0.26 (outflow funnel) and r = 0.43 (lateral flow assay), respectively. The optimal threshold to identify high-quality colostrum using ELISA was 50.8 mg/mL with sensitivity 91.3%, specificity 92.3%, and AUC of 0.94. For the on-farm tests sensitivity ranged from 95.7% (Brix refractometry) to 60.9% (lateral flow assay). Specificity ranged from 88.6% (lateral flow assay) to 75.9% (colostrometer: temperature corrected). The AUC ranged from 0.93 (Brix refractometry) to 0.73 (outflow funnel). Based on the AUC, ELISA (0.94) and Brix refractometry (0.93) can be considered highly accurate. In conclusion, the ELISA is accurate to assess colostrum quality. Regarding the on-farm tests only the digital Brix refractometer and the colostrometer were adequate to determine colostrum quality

    Instabilities and turbulence in stellarators from the perspective of global codes

    In this work, a comparison of the global gyrokinetic codes EUTERPE and GENE-3D in stellarator configurations of LHD and W7-X is carried out. In linear simulations with adiabatic electrons, excellent agreement is found in the mode numbers, growth rate and frequency, mode structure, and spatial localization of the most unstable mode in LHD. In W7-X, the dependence of the growth rate and frequency with the mode number is well reproduced by both codes. The codes are also compared in linear simulations with kinetic ions and electrons in W7-X using model profiles, and reasonable agreement is found in the wavenumber of the most unstable modes. A stabilization of small-scale modes in kinetic-electron simulations with respect to the adiabatic-electron case is consistently found in both codes. Nonlinear simulations using adiabatic electrons and model profiles are also studied and the heat fluxes are compared. Very good agreement is found in the turbulent ion heat fluxes in both LHD and W7-X. Two problems that cannot be properly accounted for in local flux tube codes are studied: the localization of instabilities and turbulence over the flux surface and the influence of a background long-wavelength electric field. Good agreement between codes is found with respect to the spatial localization of instabilities and turbulence over the flux surface. The localization of saturated turbulence is found in both codes to be much smaller than that of the linear instabilities and smaller than previously reported in full-surface radially-local simulations. The influence of the electric field on the localization is also found to be smaller in the developed turbulent state than in the linear phase, and smaller than in previous works. A stabilizing effect of a constant electric field on the linearly unstable modes is found in both codes. A moderate reduction of turbulent transport by the radial electric field..

    On Measuring Non-Recursive Trade-Offs

    We investigate the phenomenon of non-recursive trade-offs between descriptional systems in an abstract fashion. We aim at categorizing non-recursive trade-offs by bounds on their growth rate, and show how to deduce such bounds in general. We also identify criteria which, in the spirit of abstract language theory, allow us to deduce non-recursive tradeoffs from effective closure properties of language families on the one hand, and differences in the decidability status of basic decision problems on the other. We develop a qualitative classification of non-recursive trade-offs in order to obtain a better understanding of this very fundamental behaviour of descriptional systems

    Zeros of the i.i.d. Gaussian power series: a conformally invariant determinantal process

    Consider the zero set of the random power series f(z)=sum a_n z^n with i.i.d. complex Gaussian coefficients a_n. We show that these zeros form a determinantal process: more precisely, their joint intensity can be written as a minor of the Bergman kernel. We show that the number of zeros of f in a disk of radius r about the origin has the same distribution as the sum of independent {0,1}-valued random variables X_k, where P(X_k=1)=r^{2k}. Moreover, the set of absolute values of the zeros of f has the same distribution as the set {U_k^{1/2k}} where the U_k are i.i.d. random variables uniform in [0,1]. The repulsion between zeros can be studied via a dynamic version where the coefficients perform Brownian motion; we show that this dynamics is conformally invariant.Comment: 37 pages, 2 figures, updated proof

    Broad-Spectrum Antiviral Activities of Neplanocin A, 3-Deazaneplanocin A, and Their 5'-Nor Derivatives

    The neplanocin A analogs, 3-deazaneplanocin A, 9-(trans-2',trans-3'-dihydroxycyclopent-4'-enyl)adenine (DHCA), and 9-(trans-2',trans-3'-dihydroxycyclopent-4'-enyl)-3-deazaadenine (DHCDA), all potent inhibitors of S-adenosylhomocysteine (AdoHcy) hydrolase, were studied for their broad-spectrum antiviral potential. 3-Deazaneplanocin A, DHCA, and DHCDA proved specifically effective against vesicular stomatitis virus, vaccinia virus, parainfluenza virus, reovirus, and rotavirus. Their selectivity was greater than that of neplanocin A, particularly against vesicular stomatitis virus and rotavirus. As could be expected from adenosine analogs that are directly targeted at AdoHcy hydrolase, 3-deazaneplanocin A, DHCA, and DHCDA were fully active in adenosine kinase-deficient cells, implying that their activity did not depend on phosphorylation by adenosine kinase. None of the AdoHcy hydrolase inhibitors showed selective activity against human immunodeficiency virus (type 1). 3-Deazaneplanocin A at a dose of 0.5 mg/kg per day conferred marked protection against a lethal infection of newborn mice with vesicular stomatitis virus.This work was supported by the Belgian Fonds voor Geneeskundig Wetenschappelijk Onderzoek (project no. 3.0040.83) and the Belgian Geconcerteerde Onderzoeksacties (project no. 85/90-79).     The Second Messenger Cyclic Di-GMP Regulates Clostridium difficile Toxin Production by Controlling Expression of sigD

    The Gram-positive obligate anaerobe Clostridium difficile causes potentially fatal intestinal diseases. How this organism regulates virulence gene expression is poorly understood. In many bacterial species, the second messenger cyclic di-GMP (c-di-GMP) negatively regulates flagellar motility and, in some cases, virulence. c-di-GMP was previously shown to repress motility of C. difficile. Recent evidence indicates that flagellar gene expression is tightly linked with expression of the genes encoding the two C. difficile toxins TcdA and TcdB, which are key virulence factors for this pathogen. Here, the effect of c-di-GMP on expression of the toxin genes tcdA and tcdB was determined, and the mechanism connecting flagellar and toxin gene expressions was examined. In C. difficile, increasing c-di-GMP levels reduced the expression levels of tcdA and tcdB, as well as that of tcdR, which encodes an alternative sigma factor that activates tcdA and tcdB expression. We hypothesized that the C. difficile orthologue of the flagellar alternative sigma factor SigD (FliA; σ28) mediates regulation of toxin gene expression in response to c-di-GMP. Indeed, ectopic expression of sigD in C. difficile resulted in increased expression levels of tcdR, tcdA, and tcdB. Furthermore, sigD expression enhanced toxin production and increased the cytopathic effect of C. difficile on cultured fibroblasts. Finally, evidence is provided that SigD directly activates tcdR expression and that SigD cannot activate tcdA or tcdB expression independent of TcdR. Taken together, these data suggest that SigD positively regulates toxin genes in C. difficile and that c-di-GMP can inhibit both motility and toxin production via SigD, making this signaling molecule a key virulence gene regulator in C. difficile
