    Antigen receptor variable region repertoires expressed by T cells infiltrating thyroid, retroorbital, and pretibial tissue in Graves' disease

    To date, it has remained unclear whether T cells infiltrating thyroid, retroorbital, and pretibial tissue of patients with Graves' ophthalmopathy and pretibial dermopathy represent a primary immune response that is directed against certain antigenic determinants shared among these involved tissues. To characterize these T cells at the molecular level, we compared the T cell antigen receptor (TcR) variable (V) region gene usage in thyroid, retroorbital, pretibial tissue, and peripheral blood mononuclear cells of two patients with Graves' disease, ophthalmopathy, and pretibial dermopathy. Ribonucleic acid was extracted, reverse transcribed, and amplified using the PCR and 22 V alpha and 23 V beta gene-specific oligonucleotide primers. The resulting TcR V alpha and V beta transcripts were verified by Southern hybridization analysis using TcR C region-specific, digoxigenin-labeled oligonucleotide probes. In addition, complementarity determining regions 3 and junctional regions of TcR V beta genes were sequenced. Marked similarities of intrathyroidal, retroorbital, and pretibial TcR V alpha and V beta gene repertoires were noted with respect to the degree of TcR V gene restriction and the patterns of individual V genes expressed. Sequence analysis of junctional domains of V beta families revealed oligoclonality of intrahyroidal, retroorbital, and pretibial T cells. In addition, certain conserved junctional motifs were shared by T cells derived the thyroid gland and the extrathyroidal sites. Our results suggest that in the two patients with Graves' disease and extrathyroidal manifestations studied, similar antigenic determinants may have contributed to the recruitment and oligoclonal expansion of T cells both within the thyroid gland and at the involved extrathyroidal sites

    Performance of a J85-13 compressor with clean and distorted inlet flow

    The results presented are of a series of experimental tests in which a J85-13 turbojet engine was subjected to both distorted and undistorted inlet total pressure conditions. A distinctive feature of the data base obtained is that it includes compressor interstage information not previously recorded for a J85-13 engine. Each of the eight compressor stages was instrumented to obtain the characteristics of the individual stages for undistorted inlet conditions, and these data are documented in the report along with the undistorted compressor overall performance. Also included in the report is the overall performance of the compressor exposed to 14 different distorted-inlet conditions - 10 circumferential patterns and 4 radial patterns. The distortion patterns were introduced using screens that spoiled from 8 to 50 percent of the compressor face area; the distortion screen density, or the area blocked by the screen wire per unit area of screen, varied from 26 to 69 percent

    Zero-temperature Monte Carlo study of the non-coplanar phase of the classical bilinear-biquadratic Heisenberg model on the triangular lattice

    We investigate the ground-state properties of the highly degenerate non-coplanar phase of the classical bilinear-biquadratic Heisenberg model on the triangular lattice with Monte Carlo simulations. For that purpose, we introduce an Ising pseudospin representation of the ground states, and we use a simple Metropolis algorithm with local updates, as well as a powerful cluster algorithm. At sizes that can be sampled with local updates, the presence of long-range order is surprisingly combined with an algebraic decay of correlations and the complete disordering of the chirality. It is only thanks to the investigation of unusually large systems (containing ∼108\sim 10^8 spins) with cluster updates that the true asymptotic regime can be reached and that the system can be proven to consist of equivalent (i.e., equally ordered) sublattices. These large-scale simulations also demonstrate that the scalar chirality exhibits long-range order at zero temperature, implying that the system has to undergo a finite-temperature phase transition. Finally, we show that the average distance in the order parameter space, which has the structure of an infinite Cayley tree, remains remarkably small between any pair of points, even in the limit when the real space distance between them tends to infinity.Comment: 15 pages, 10 figure
