    Pengaruh Pemberian Ekstrak Phyllanthus Niruri Linn terhadap Infiltrasi Limfosit dan Ekspresi Perforin pada Kanker Kolon Tikus Sprague-Dawley

    The effect of Phyllanthus niruri Linn extract on lymphocytes infiltration and perforin expression in colon cancer of Sprague-Dawley RatBackground: Colon cancer treatment currently involves immunotherapy that aims to improve the quality of life and survival of patients. Phyllanthus niruri Linn (P. niruri L) may act as an immunomodulator, but its potency in antitumor immune responses has not been revealed. The study was conducted to evaluate the differences between the immunological status of rats suffering colon cancer which were not given to those given the extract of P. niruri L.Methods: The study was randomized posttest-only control group design. Samples were 30 Sprague-Dawley male rats, bodyweight 170-220 gr which induced by 1.2 DMH 30 mg/kgBW subcutaneously. On the weeks 9, 11 and 13, four induced rats each week were sacrified to detect the development of colon cancer. On the weeks of 13th all of 4 rats were developed colon cancer, so the induction were stopped. The rest of 18 induced rats were randomly into two groups: without P. niruri L or positive control (K+)=9 rats and given P. niruri L extract 13.5 mg/kg orally or X group=9 rats. After 19th week all of rats were then terminated and tumor lesion of colon were processed hystophatologically. The tissues of colon cancer were stained by H&E for evaluate the lymphocytes infiltration and immunohistochemistry monoclonal antibody anti-perforin for perforin expression. Non pairs t-test was used with considered significant if p<0.05.Results: The mean of lymphocytes infiltration of the group X was 401.89±70.19, it was higher compared to K+ 191.89±50.68 (p=0.000). The mean percentage of perforin expression of group X was 39.00±1.80%, it was higher compared to K+ 23.00± 3.00% (p=0.000). Conclusion: The extract of P. niruri L increases immunologic status through mechanism of lymphocytes infiltration and perforin expression elevation of colon cancer in animal mode

    Effect of Phyllanthus niruri Linn extract on tumor growth and cell proliferation of colorectal cancer: experimental study in 1.2 DMH-induced Sprague-Dawley ratsBackground: The development of chemotherapy for colorectal cancer (CRC) is so very advanced, however, the survival of patients has not been satisfactory, therefore, the current therapy also involve immunotherapy. Phyllanthus niruri Linn (P. niruri L) may act as an immunomodulator and anticancer, but its potency has not been revealed. Study was conducted to confirm the effects of P. niruri L extract in the treatment of CRC.Method: The study was randomized posttest-only control group design. Samples were Sprague-Dawley male rats, bodyweight 170- 220 gr, were divided into two groups: non induced or negative control (K-) consisted of 9 normal rats, and induced by 1.2 DMH 30 mg/kgBW subcutaneously group consisted 30 rats. On the weeks 9, 11 and 13, four induced rats each week were sacrified to detect the development of CRC. On the weeks of 13th all of 4 rats were developed CRC, so the induction were stopped. The rest of 18 induced rats were randomly into two groups: without P. niruri L or positive control (K+)=9 rats and given P. niruri L extract 13.5 mg/kg orally or X group=9 rats. After 19th week all of rats were then terminated and tumor lesion of colon were examined macroscopically and hystophatologic tissues were stained with AgNORs for evaluate the cells proliferation. Oneway Anova and Post Hoc LSD test for the growth of colon tumor and non pairs t-test for cell proliferation were used. Considered significant if p was <0.05. Result: There was no tumor growth on K- group, while for K+ was 83.33±14.34% and on X was 40.44±13.23% (p=0.000). The mean of AgNORs on K+ was 4.60±0.55 while on X was 2.25±0.39 (p=0.000).Conclusion: The extract of P. niruri L suppress the tumor growth and cell proliferation of CRC

    Penelitian ini dilakukan untuk mengetahui perbedaan antara pengenaan pajak final dan pajak non final. Penelitian ini menggunakan data laporan keuangan yang telah dipublikasi selama kurun waktu 2010-2014 (lima tahun). Metode statistik dalam penelitian ini menggunakan uji normalitas, uji homogenitas, uji ANOVA, dan Uji Independent t-Test. Berdasarkan hasil analisis statistik bahwa terdapat perbedaan yang signifikan antara pengenaan pajak final dan pajak non final dalam menentukan besarnya pajak yang terutang. Pengenaan pajak final lebih besar secara signifikan dibandingkan dengan pengenaan pajak non final. Bagi pemerintah, pengenaan pajak secara final dinilai sangat menguntungkan karena dapat menambah sumber penerimaan dalam keuangan negara. Namun, disisi lain bagi wajib dinilai sangat merugikan karena pengenaan pajak secara final tidak memperhatikan biaya-biaya yang dikeluarkan dalam memperoleh penghasilan tersebut

    Influence of Connection Placement to the Behavior of Precast Concrete Exterior Beam-Column Joint. This paperpresents an experimental study on the influence of connection placement to the behaviour of exterior beamcolumnjoint of precast concrete structure under semi cyclic loading. Four half-scale beam-column specimens wereinvestigated. Three beam-columns were jointed through connection that are placed in beam-column joint region and theforth is connected at the plastic hinge potensial region of the beam.     Evaluation of the changes in working limits in an automobile assembly line using simulation

    The aim of the work presented in this paper consists of the development of a decision-making support system, based on discrete-event simulation models, of an automobile assembly line which was implemented within an Arena simulation environment and focused at a very specific class of production lines with a four closed-loop network configuration. This layout system reflects one of the most common configurations of automobile assembly and preassembly lines formed by conveyors. The sum of the number of pallets on the intermediate buffers, remains constant, except for the fourth closed-loop, which depends on the four-door car ratio (x) implemented between the door disassembly and assembly stations of the car body. Some governing equations of the four closed-loops are not compatible with the capacities of several intermediate buffers for certain values of variable x. This incompatibility shows how the assembly line cannot operate in practice for x0,97 in a stationary regime, due to the starvation phenomenon or the failure of supply to the machines on the production line. We have evaluated the impact of the pallet numbers circulating on the first closed-loop on the performance of the production line, translated into the number of cars produced/hour, in order to improve the availability of the entire manufacturing system for any value of x. Until the present date, these facts have not been presented in specialized literature. © 2012 American Institute of Physics

    Simulation study for investment decisions on the EcoBoost camshaft machining line

    Design/redesign of manufacturing systems is a complex, risky, and expensive task. Ford Motor Company’s Valencia Engine Plant faces this challenge as it plans to upgrade its machining and assembly lines to introduce the new EcoBoost engines. The research project described in this paper aimed to support the transition process particularly at the camshaft machining line by using simulation modelling techniques. A series of experiments was carried out using the simulation model developed, and recommendations were proposed based on the results of these experiments to support the decision as to where to invest on the line. The outcomes from the research project indicated that investment is required in terms of increasing the capacity of two bottleneck operations through retooling and improving the conveyor routing logic in one key area.     Carotenoid Distribution in Living Cells of Haematococcus pluvialis (Chlorophyceae)

    Haematococcus pluvialis is a freshwater unicellular green microalga belonging to the class Chlorophyceae and is of commercial interest for its ability to accumulate massive amounts of the red ketocarotenoid astaxanthin (3,3′-dihydroxy-β,β-carotene-4,4′-dione). Using confocal Raman microscopy and multivariate analysis, we demonstrate the ability to spectrally resolve resonance–enhanced Raman signatures associated with astaxanthin and β-carotene along with chlorophyll fluorescence. By mathematically isolating these spectral signatures, in turn, it is possible to locate these species independent of each other in living cells of H. pluvialis in various stages of the life cycle. Chlorophyll emission was found only in the chloroplast whereas astaxanthin was identified within globular and punctate regions of the cytoplasmic space. Moreover, we found evidence for β-carotene to be co-located with both the chloroplast and astaxanthin in the cytosol. These observations imply that β-carotene is a precursor for astaxanthin and the synthesis of astaxanthin occurs outside the chloroplast. Our work demonstrates the broad utility of confocal Raman microscopy to resolve spectral signatures of highly similar chromophores in living cells
