    Functional Stoichiometry at the Nicotinic Receptor. The Photon Cross Section for Phase 1 Corresponds to Two Bis-Q Molecules per Channel

    These experiments examine changes in the agonist-induced conductance that occur when the agonist-receptor complex is perturbed. Voltage-clamped Electrophorus electroplaques are exposed to the photoisomerizable agonist trans-Bis-Q A 1-µs laser flash photoisomerizes some trans-Bis-Q molecules bound to receptors; because the cis configuration is not an agonist, receptor channels close within a few hundred microseconds. This effect is called phase 1. We compare (a) the fraction of channels that close during phase 1 with (b) the fraction of trans-Bis-Q molecules that undergo trans → cis photoisomerization. Parameter a is measured as the fractional diminution in voltage-clamp currents during phase 1. Parameter b is measured by changes in the optical spectra of Bis-Q solutions caused by flashes . At low flash intensities, a is twice b, which shows that the channel can be closed by photoisomerizing either of two bound agonist molecules. Conventional dose-response studies with trans-Bis-Q also give a Hill coefficient of two. As a partial control for changes in the photochemistry caused by binding of Bis-Q to receptors, spectral measurements are performed on the photoisomerizable agonist QBr, covalently bound to solubilized acetylcholine receptors from Torpedo. The bound and free agonist molecules have the same photoisomerization properties. These results verify the concept that the open state of the acetylcholine receptor channel is much more likely to be associated with the presence of two bound agonist molecules than with a single such molecule

    State Power Plant Siting: a Sketch of the Main Features of a Possible Approach

    Work on various phases of power plant technology and siting has been underway within the Environmental Quality Laboratory (EQL) at the California Institute of Technology for some time. Of particular relevance to this memorandum, a good deal of effort has been devoted to institutional aspects of the siting process. Our purpose in what follows is to draw from our past work -- and from the discussions and work of others -- a sketch of the major outlines of one possible approach to power plant siting for the state. We hope in doing so to give our present views about the issues and how they might rationally be resolved, not so much to convince as to inform, stimulate fruitful ideas, and help provide the basis for constructive debate. We ourselves are not necessarily wedded to any of the discussion that follows; we find our own minds changing from time to time as we study the problem further or confront sound suggestions from others. Part I of this memorandum briefly outlines the major features of what we see as a fruitful approach to the siting problem. Sections A through E of Part I describe some elements of the approach; Section F sketches the actual siting decision process we suggest, and in doing so shows how the elements play into the process. Section G comments briefly on a suggested role for judicial review. In Part II we attempt to reduce our ideas to a fairly precise outline for a state siting statute, and to deal with certain matters of detail not covered in Part I. Section A of Part II introduces the statutory outline by summarizing each of its provisions; Section B sets forth the outline itself. The Appendix to this memorandum depicts our suggested approach in time-line fashion; it should be helpful in reading and understanding the proposal

    Current analytical methods for determining steroids in sewage sludge and sediment are reviewed and experimental data comparing LC/MS ESI and GC/MS quantification of extracts from these matrices is presented. The effect of matrix influence on method analysis and steroid recoveries was investigated and a comparison of GC and LC determination with mass spectrometry performed. Consequently upon this, an environmental analytically robust methodology is presented and using this technique analysis of sediments determined E1 to be the main steroid (<18 ng/g), EE2 and E3 were determined at far lower concentrations with minimal detection of E

    A cross-cultural study of attitudes toward suicide among young people in India, Italy and Australia

    Background: An understanding of the cultural aspects of suicidal behavior is essential for the development of culturally appropriate suicide prevention and intervention strategies. Aims: This study explored the attitudes toward youth suicide in 686 Italian, Indian and Australian undergraduate students (18-24 years old). Method: A 21-item suicide attitude inventory titled Attitude towards Youth Suicide (AtYS) Scale was used in the three samples. Results: Four factors were extracted, labelled negative attitudes toward suicide, belief that suicide was not preventable, suicide as acceptable and normal, and the existence of risk signs for suicide. Country differences were found for all four subscales, with Indian students having the most negative attitudes toward suicide. Sex differences were found in all three countries with women, on the whole, having less negative attitudes toward suicide, more belief in the preventability of suicide in India and more belief in risk signs for suicide in Italy. Conclusions: Attitudes are linked to suicide in a complex manner. More quantitative and qualitative studies, including in lower-income and non-English speaking Western societies, are needed

    Conductance increases produced by bath application of cholinergic agonists to Electrophorus electroplaques

    When solutions containing agonists are applied to the innervated face of an Electrophorus electroplaque, the membrane's conductance increases. The agonist-induced conductance is increased at more negative membrane potentials. The "instantaneous" current-voltage curve for agonist-induced currents is linear and shows a reversal potential near zero mV; chord conductances, calculated on the basis of this reversal potential, change epsilon-fold for every 62-mV change in potential when the conductance is small. Conductance depends non- linearly on small agonist concentrations; at all potentials, the dose-response curve has a Hill coefficient of 1.45 for decamethonium (Deca) and 1.90 for carbamylcholine (Carb). With agonist concentrations greater than 10^(-4) M Carb or 10^(-5) M Deca, the conductance rises to a peak 0.5-1.5 min after introduction of agonist, then declines with time; this effect resembles the "desensitization" reported for myoneural junctions. Elapid alpha-toxin, tubocurarine, and desensitization reduce the conductance without changing the effects of potential; the apparent dissociation constant for tubocurarine is 2 X 10^(-7) M. By contrast, procaine effects a greater fractional inhibition of the conductance at high negative potentials

    Assessment of Basic Concept Knowledge of Children from Differing Socio-Economic Status Homes

    In the field of education, research suggests that socioeconomic status (SES) influences vocabulary development and academic achievement. Children from high SES backgrounds tend to score better on standardized vocabulary tests than children from low SES backgrounds; however, to date only general vocabulary measures have been utilized. Basic concepts, a specific subset of vocabulary, serve as the foundation upon which children build their conceptual knowledge. For this study, two groups of preschool children (one from high SES homes and one from low SES homes) were administered the Bracken Basic Concept Scale - Third Edition (BBCS:3). The purpose of this study was to determine if children from higher SES homes do, in fact, have a higher knowledge of basic concepts than their lower SES peers. An independent samples t-test was used to statistically analyze the differences between test scores of the two groups (high and low SES). Results revealed that children from higher SES homes scored significantly higher on the BBCS:3 than their peers from lower SES homes. Implications of this study are discussed
