    Zinc ions alter morphology and chitin deposition in an ericoid fungus

    A sterile mycelium PS IV, an ascomycete capable of establishing ericoid mycorrhizas, was used to investigate how zinc ions affect the cellular mechanisms of fungal growth. Asignificant reduction of the fungal biomass was observed in the presence of millimolar zinc concentrations; this mirrored conspicuous changes in hyphal morphology which led to apical swellings and increased branching in the subapical parts. Specific probes for fluorescence and electron microscopy localised chitin, the main cell wall polysaccharide, on the inner part of the fungal wall and on septa in control specimens. In Zn-treated mycelium, hyphal walls were thicker and a more intense chitin labelling was detected on the transverse walls.     Volumetric and anatomical MRI for hypoxic-ischemic encephalopathy: relationship to hypothermia therapy and neurosensory impairments.

    OBJECTIVE: To relate volumetric magnetic resonance imaging (MRI) findings to hypothermia therapy and neurosensory impairments. STUDY DESIGN: Newborns \u3e or =36 weeks\u27 gestation with hypoxic-ischemic encephalopathy who participated in the National Institute of Child Health and Human Development hypothermia randomized trial at our center were eligible. We determined the relationship between hypothermia treatment and usual care (control) to absolute and relative cerebral tissue volumes. Furthermore, we correlated brain volumes with death or neurosensory impairments at 18 to 22 months. RESULT: Both treatment groups were comparable before randomization. Total brain tissue volumes did not differ in relation to treatment assignment. However, relative volumes of subcortical white matter were significantly larger in hypothermia-treated than control infants. Furthermore, relative total brain volumes correlated significantly with death or neurosensory impairments. Relative volumes of the cortical gray and subcortical white matter also correlated significantly with Bayley Scales psychomotor development index. CONCLUSION: Selected volumetric MRI findings correlated with hypothermia therapy and neurosensory impairments. Larger studies using MRI brain volumes as a secondary outcome measure are needed

    Exceptional response to immunotherapy in association with radiotherapy in patient with breast metastasis from urothelial carcinoma: A case report

    Most common sites of metastasis of urothelial carcinoma (UC) are lungs, liver, lymph nodes and bone. Pembrolizumab, a humanized monoclonal antibody directed against programmed cell death protein-1 (PD-1), represents an effective second-line therapy for advanced UC. Radiotherapy has been shown to induce a mechanism of immunogenic cell death (ICD) resulting in immune memory and advantageous systemic effects. We present the first case of breast metastasis (BM) from a UC described in literature who had an exceptional response to second-line therapy with pembrolizumab in association with radiotherapy, showing the efficacy of combining immunotherapy and radiotherapy even in patients with atypical metastatic sites

    Prevalence of Mycobacterium avium subsp. paratuberculosis in milk and dairy cattle in Southern Italy; preliminary results

    Paratuberculosis affects all ruminants worldwide. Mycobacterium avium subsp. paratuberculosis could have a role in human diseases like Crohn\u2019s. Some extra EU countries request importation of MAP-free products. Italy has not yet actualized a control program and the diffusion of the infection is still unknown in Southern Italy. The aim of this study was to evaluate the prevalence of the infection in five regions of Southern Italy. Bulk tank milk and in-line milk filters were sampled in 780 dairy cattle herds and respectively analyzed by ELISA and real time PCR. One hundred and fifty-five out of 780 herds (19.9%) were found positive by ELISA and/or real time PCR. Individual milk samples were then collected from all the producing animals of positive herds and from a selection of negative herds. The estimated prevalence varies from region to region between 2.8% and 5.5%. Our results indicate that the disease is widespread in the five regions. The observed prevalence could be underestimated

    ITS-1 versus ITS-2 pyrosequencing: a comparison of fungal populations in truffle-grounds

    International audienceIn a recent study pyrosequencing of the ribosomal internal transcribed spacer-1 (ITS-1) has validated the effectiveness of such technology in the survey of soil fungal diversity. Here we compare the two ITS regions, ITS-1 and ITS-2, of the fungal populations occurring in Tuber melanosporum/Quercus pubescens truffle grounds and sampled in two areas, one devoid of vegetation ("burned", bade in French) where T melanosporum fruiting bodies are usually collected, and outside the brule. TS1F/IT52 and ITS3/ITS4 were used respectively for the amplification of the ITS-1 and ITS-2 regions. Two amplicon libraries were built, one for inside and the other for outside. A set of 15.788 reads was obtained. After the removal of low quality sequences, 3568 and 3156 sequences were obtained from inside the brule with the ITS-1 and ITS-2 primers respectively. The sequences obtained from outside the brule were 4490 with the ITS-1 primers and 2432 with the ITS-2 primers. Most of the sequences obtained for both ITS fragments could be attributed to fungal organisms. The pair of primers, ITS1-F/ITS2, was more selective, producing fewer non-fungal sequences (1% inside, 3% outside), in addition to a higher number of sequences, than the pair ITS3/ITS4 (6% inside, 11% outside). Although differences are present in the taxa percentages between ITS-1 and ITS-2, both reveal that Ascomycota were the dominant fungal phylum and that their number decreased moving from inside the brule to outside, while the number of Basidiomycota increased. Taken together, both the short ITS-1 and ITS-2 reads obtained by the high throughput 454 sequencing provide adequate information for taxon assignment and are suitable to correlate the dynamics of the fungal populations to specific environments

    Integration of rule-based ‘Expert Systems’ on RPAS capable of specific category operations within the U-space: An original mitigation strategy for operational safety risks

    The use of RPAS for civil purposes is spreading across Europe and worldwide; Aviation Authorities are working to layout regulations to assure a safe and secure integration of RPAS with manned aircraft across both controlled and uncontrolled (below 500 Feet of altitude) airspace. Following the identification of a selection of safety risks potentially associated to RPAS Specific Category of operations, an original strategy of risks mitigation focused on rule-based ‘Expert Systems’, has been conceived and it is discussed in this work. The article recalls the main components of rule-based ‘Expert Systems’ that is the knowledge basis and the rules to instruct the ‘Expert system’. Then the work describes the implementation of the rules as statements derived from a safety risk matrix associated to RPAS capable of performing Specific Category operations within the U-space. Finally, the idea of integrating the ‘Expert System’ as a software module within RPAS functional architecture is presented and discussed. Such solution is deemed to be a valuable novelty for future implementations of advanced RPAS autopilots capable of recognizing and solving in flight/on ground operational safety risks in such a way to speed up the integration of RPAS into not segregated airspace and their market development
