Three-Dimensional Characterization of Mechanical Interactions between Endothelial Cells and Extracellular Matrix during Angiogenic Sprouting
We studied the three-dimensional cell-extracellular matrix interactions of endothelial cells that form multicellular structures called sprouts. We analyzed the data collected in-situ from angiogenic sprouting experiments and identified the differentiated interaction behavior exhibited by the tip and stalk cells. Moreover, our analysis of the tip cell lamellipodia revealed the diversity in their interaction behavior under certain conditions (e.g., when the heading of a sprout is switched approximately between the long-axis direction of two different lamellipodia). This study marks the first time that new characteristics of such interactions have been identified with shape changes in the sprouts and the associated rearrangements of collagen fibers. Clear illustrations of such changes are depicted in three-dimensional views.Singapore-MIT Alliance in Research and Technology (SMART
Derivation of Rabbit Embryonic Stem Cells from Vitrified–Thawed Embryos
The rabbit is a useful animal model for regenerative medicine. We previously developed pluripotent rabbit embryonic stem cell (rbESC) lines using fresh embryos. We also successfully cryopreserved rabbit embryos by vitrification. In the present work, we combined these two technologies to derive rbESCs using vitrified?thawed (V/T) embryos. We demonstrate that V/T blastocysts (BLs) can be used to derive pluripotent rbESCs with efficiencies comparable to those using fresh BLs. These ESCs are undistinguishable from the ones derived from fresh embryos. We tested the developmental capacity of rbESCs derived from V/T embryos by BL injection experiments and produced chimeric kits. Our work adds cryopreservation to the toolbox of rabbit stem cell research and applications and will greatly expand the available research materials for regenerative medicine in a clinically relevant animal model.Peer Reviewedhttps://deepblue.lib.umich.edu/bitstream/2027.42/140328/1/cell.2015.0044.pd
The Lambek calculus with iteration: two variants
Formulae of the Lambek calculus are constructed using three binary
connectives, multiplication and two divisions. We extend it using a unary
connective, positive Kleene iteration. For this new operation, following its
natural interpretation, we present two lines of calculi. The first one is a
fragment of infinitary action logic and includes an omega-rule for introducing
iteration to the antecedent. We also consider a version with infinite (but
finitely branching) derivations and prove equivalence of these two versions. In
Kleene algebras, this line of calculi corresponds to the *-continuous case. For
the second line, we restrict our infinite derivations to cyclic (regular) ones.
We show that this system is equivalent to a variant of action logic that
corresponds to general residuated Kleene algebras, not necessarily
*-continuous. Finally, we show that, in contrast with the case without division
operations (considered by Kozen), the first system is strictly stronger than
the second one. To prove this, we use a complexity argument. Namely, we show,
using methods of Buszkowski and Palka, that the first system is -hard,
and therefore is not recursively enumerable and cannot be described by a
calculus with finite derivations
Ultrafast Hole Trapping and Relaxation Dynamics in p-Type CuS Nanodisks
CuS nanocrystals are potential materials for developing low-cost solar energy conversion devices. Understanding the underlying dynamics of photoinduced carriers in CuS nanocrystals is essential to improve their performance in these devices. In this work, we investigated the photoinduced hole dynamics in CuS nanodisks (NDs) using the combination of transient optical (OTA) and X-ray (XTA) absorption spectroscopy. OTA results show that the broad transient absorption in the visible region is attributed to the photoinduced hot and trapped holes. The hole trapping process occurs on a subpicosecond time scale, followed by carrier recombination (~100 ps). The nature of the hole trapping sites, revealed by XTA, is characteristic of S or organic ligands on the surface of CuS NDs. These results not only suggest the possibility to control the hole dynamics by tuning the surface chemistry of CuS but also represent the first time observation of hole dynamics in semiconductor nanocrystals using XTA
Constraints on Spin-Independent Nucleus Scattering with sub-GeV Weakly Interacting Massive Particle Dark Matter from the CDEX-1B Experiment at the China Jin-Ping Laboratory
We report results on the searches of weakly interacting massive particles
(WIMPs) with sub-GeV masses () via WIMP-nucleus spin-independent
scattering with Migdal effect incorporated. Analysis on time-integrated (TI)
and annual modulation (AM) effects on CDEX-1B data are performed, with 737.1
kgday exposure and 160 eVee threshold for TI analysis, and 1107.5
kgday exposure and 250 eVee threshold for AM analysis. The sensitive
windows in are expanded by an order of magnitude to lower DM masses
with Migdal effect incorporated. New limits on at
90\% confidence level are derived as 1010
for TI analysis at 50180 MeV/, and
1010 for AM analysis at
75 MeV/3.0 GeV/.Comment: 5 pages, 4 figure
Search for Light Weakly-Interacting-Massive-Particle Dark Matter by Annual Modulation Analysis with a Point-Contact Germanium Detector at the China Jinping Underground Laboratory
We present results on light weakly interacting massive particle (WIMP)
searches with annual modulation (AM) analysis on data from a 1-kg mass -type
point-contact germanium detector of the CDEX-1B experiment at the China Jinping
Underground Laboratory. Datasets with a total live time of 3.2 yr within a 4.2
yr span are analyzed with analysis threshold of 250 eVee. Limits on
WIMP-nucleus (-) spin-independent cross sections as function of WIMP
mass () at 90\% confidence level (C.L.) are derived using the dark
matter halo model. Within the context of the standard halo model, the 90\% C.L.
allowed regions implied by the DAMA/LIBRA and CoGeNT AM-based analysis are
excluded at 99.99\% and 98\% C.L., respectively. These results correspond to
the best sensitivity at 6 among WIMP AM
measurements to date.Comment: 5 pages, 4 figure
Recombinant Rabbit Leukemia Inhibitory Factor and Rabbit Embryonic Fibroblasts Support the Derivation and Maintenance of Rabbit Embryonic Stem Cells
Abstract The rabbit is a classical experimental animal species. A major limitation in using rabbits for biomedical research is the lack of germ-line-competent rabbit embryonic stem cells (rbESCs). We hypothesized that the use of homologous feeder cells and recombinant rabbit leukemia inhibitory factor (rbLIF) might improve the chance in deriving germ-line-competent rbES cells. In the present study, we established rabbit embryonic fibroblast (REF) feeder layers and synthesized recombinant rbLIF. We derived a total of seven putative rbESC lines, of which two lines (M5 and M23) were from culture Condition I using mouse embryonic fibroblasts (MEFs) as feeders supplemented with human LIF (hLIF) (MEF+hLIF). Another five lines (R4, R9, R15, R21, and R31) were derived from Condition II using REFs as feeder cells supplemented with rbLIF (REF+rbLIF). Similar derivation efficiency was observed between these two conditions (8.7% vs. 10.2%). In a separate experiment with 2?3 factorial design, we examined the effects of feeder cells (MEF vs. REF) and LIFs (mLIF, hLIF vs. rbLIF) on rbESC culture. Both Conditions I and II supported satisfactory rbESC culture, with similar or better population doubling time and colony-forming efficiency than other combinations of feeder cells with LIFs. Rabbit ESCs derived and maintained on both conditions displayed typical ESC characteristics, including ESC pluripotency marker expression (AP, Oct4, Sox2, Nanog, and SSEA4) and gene expression (Oct4, Sox2, Nanog, c-Myc, Klf4, and Dppa5), and the capacity to differentiate into three primary germ layers in vitro. The present work is the first attempt to establish rbESC lines using homologous feeder cells and recombinant rbLIF, by which the rbESCs were derived and maintained normally. These cell lines are unique resources and may facilitate the derivation of germ-line-competent rbESCs.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/98439/1/cell%2E2012%2E0001.pd
