    Clinicopathological findings in cats tested for feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV)

    This retrospective study aimed to evaluate the clinicopathological changes in a population of cats tested for feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV), in an Italian Veterinary University Hospital, in the period between January 2002 and May 2016. During the period of 14 years, 1834 cats were tested, and of these 241/1834 (13.1%) were positive for FIV antibodies and 92/1834 (5%) cats were positive for FeLV antigen. These data confirm the presence of a high prevalence of these viruses on Italian territory. To the authors’ knowledge, this study describes findings that have never been evaluated before, such as iron status in retrovirus-infected cats and urinalysis in FeLV-positive cats. In this study, FIV-positive cats were more likely to have higher serum protein concentration and lower albumin-globulin ratio than other groups of cats. Lower urine specific gravity and higher urine protein to creatinine ratio were also detected for FIV-positive cats when compared with negative and healthy cats. FeLV-positive cats were more likely to have cytopenia, decreased haemoglobin, haematocrit and RBC compared with other groups of cats. The data obtained underline the importance of considering retroviral infections in the presence of a broad spectrum of risk factors and laboratory anomalies

    Urinary reference values and first insight into the urinary proteome of captive giraffes

    Urinalysis is widely recognized to be a useful tool in routine health investigations, since it can diagnose numerous pathologies. Considering the paucity of knowledge concerning giraffes, urine from 44 giraffes (Giraffa camelopardalis) (18 males and 26 females, from 3 months of age to 21 years of age) underwent routine urinalysis, 1D-electrophoresis, and protein identification using mass spectrometry, with the aim of identifying the urinary reference values and the urine proteome. The urine specific gravity (USG), urine total proteins (uTP), urine creatinine (uCr), and urine protein:creatinine ratio (UPC) reference values, reported as the median, and lower limit (LL) and upper limit (UL), were 1.030 (1006\u20131.049), 17.58 (4.54\u201335.31) mg/dL, 154.62 (39.59\u2013357.95) mg/dL, and 0.11 (0.07\u20130.16), respectively. Mass spectrometry, together with electrophoresis, revealed a pattern of common urinary proteins; albumin, lysozyme C, and ubiquitin were the most represented proteins in the giraffe urine. It has been hypothesized that these proteins could act as a defense against microbes. Moreover, in giraffes, urinalysis could be a valid tool for gauging renal function and physiological status changes

    AGN Black Hole Masses and Bolometric Luminosities

    Black hole mass, along with mass accretion rate, is a fundamental property of active galactic nuclei. Black hole mass sets an approximate upper limit to AGN energetics via the Eddington limit. We collect and compare all AGN black hole mass estimates from the literature; these 177 masses are mostly based on the virial assumption for the broad emission lines, with the broad-line region size determined from either reverberation mapping or optical luminosity. We introduce 200 additional black hole mass estimates based on properties of the host galaxy bulges, using either the observed stellar velocity dispersion or using the fundamental plane relation to infer σ\sigma; these methods assume that AGN hosts are normal galaxies. We compare 36 cases for which black hole mass has been generated by different methods and find, for individual objects, a scatter as high as a couple of orders of magnitude. The less direct the method, the larger the discrepancy with other estimates, probably due to the large scatter in the underlying correlations assumed. Using published fluxes, we calculate bolometric luminosities for 234 AGNs and investigate the relation between black hole mass and luminosity. In contrast to other studies, we find no significant correlation of black hole mass with luminosity, other than those induced by circular reasoning in the estimation of black hole mass. The Eddington limit defines an approximate upper envelope to the distribution of luminosities, but the lower envelope depends entirely on the sample of AGN included.     Endolysosomal targeting of a clinical chlorin photosensitiser for light-triggered delivery of nano-sized medicines

    A major problem with many promising nano-sized biotherapeutics including macromolecules is that owing to their size they are subject to cellular uptake via endocytosis, and become entrapped and then degraded within endolysosomes, which can significantly impair their therapeutic efficacy. Photochemical internalisation (PCI) is a technique for inducing cytosolic release of the entrapped agents that harnesses sub-lethal photodynamic therapy (PDT) using a photosensitiser that localises in endolysosomal membranes. Using light to trigger reactive oxygen species-mediated rupture of the photosensitised endolysosomal membranes, the spatio-temporal selectivity of PCI then enables cytosolic release of the agents at the selected time after administration so that they can reach their intracellular targets. However, conventional photosensitisers used clinically for PDT are ineffective for photochemical internalisation owing to their sub-optimal intracellular localisation. In this work we demonstrate that such a photosensitiser, chlorin e6, can be repurposed for PCI by conjugating the chlorin to a cell penetrating peptide, using bioorthogonal ligation chemistry. The peptide conjugation enables targeting of endosomal membranes so that light-triggered cytosolic release of an entrapped nano-sized cytotoxin can be achieved with consequent improvement in cytotoxicity. The photoproperties of the chlorin moiety are also conserved, with comparable singlet oxygen quantum yields found to the free chlorin

    Clinical, electrodiagnostic findings and quality of life of dogs and cats with brachial plexus injury

    Brachial plexus injury (BPI) represents a common consequence of road traffic accidents in humans and small animals. In humans, neuropathic pain is a common symptom after BPI. The aim of the study was to describe the clinical signs, the electrodiagnostic findings, the outcome and the quality of life (QoL) of a cohort of dogs and cats with BPI. Clinical records of 40 dogs and 26 cats with BPI were retrospectively reviewed. Specific attention was put on the evaluation of electrodiagnostic findings (35/40 dogs; 14/26 cats) and telephonic interview results (26/40 dogs; 18/26 cats). The most common neurological condition was the inability to bear weight and sensory deficits on the affected limb. Radial and ulnar motor nerve conduction studies (MNCSs) were absent respectively in 47% (radial) and 62% (ulnar) of dogs and 57% (radial) and 57% (ulnar) of cats. The absence of radial (p = 0.003) and ulnar (p = 0.007) MNCSs in dogs and ulnar MNCSs in cats (p = 0.02) was significantly associated to the amputation of the affected limb. The owners described signs of pain/discomfort in 73% of dogs and 56% of cats. This is the first report suggesting that neuropathic pain/discomfort should be adequately considered in order to improve the QoL

    Finding and counting vertex-colored subtrees

    The problems studied in this article originate from the Graph Motif problem introduced by Lacroix et al. in the context of biological networks. The problem is to decide if a vertex-colored graph has a connected subgraph whose colors equal a given multiset of colors MM. It is a graph pattern-matching problem variant, where the structure of the occurrence of the pattern is not of interest but the only requirement is the connectedness. Using an algebraic framework recently introduced by Koutis et al., we obtain new FPT algorithms for Graph Motif and variants, with improved running times. We also obtain results on the counting versions of this problem, proving that the counting problem is FPT if M is a set, but becomes W[1]-hard if M is a multiset with two colors.     Vitamin D status in cord blood and newborns: ethnic differences.

    BACKGROUND: A deficiency in vitamin D (25OHD) is common throughout the world in both adults and children, being related to skin pigmentation, sun exposure, dietary intake and obesity. Limited data are available for the neonatal age. The aim of the study is to understand the differences in 25OHD levels with respect to skin colour and ethnicity in newborns. METHODS: We randomly enrolled 62 neonates, born at term and appropriate for gestational age. Thirty two were born from Italian mothers with fair skin (FS) and 30 from non-Caucasian mothers (North African, African, Asian and Latin American): 10 with light olive/light brown (LOB) and 20 with medium brown/black skin (MBB). Vitamin D was measured in the cord blood at birth and in neonatal serum during metabolic screening. RESULTS: 25OHD levels were (mean\u2009\ub1\u2009SD) 21.4\u2009\ub1\u200911 ng/ml in cord blood and 14.9\u2009\ub1\u20097 ng/ml in serum after birth. 25OHD values were higher in cord blood (p\u2009<\u20090.01) and neonatal serum (p\u2009<\u20090.001) in subjects supplemented with Vitamin D. Newborn FS showed higher vitamin D levels in cord blood when compared to LOB and MBB (p\u2009<\u20090.01), and higher levels in neonatal serum when compared to LOB (p\u2009<\u20090.01). In cord blood, 25OHD levels were higher in Italian newborns than in North African (p\u2009<\u20090.004) and African (p\u2009<\u20090.01). In neonatal serum, 25OHD levels were higher in Italian infants only when compared with North African infants (p\u2009<\u20090.03). CONCLUSIONS: The present study shows a high prevalence of vitamin D insufficiency and deficiency in newborns with significant differences observed to be due to ethnicity, skin colour and maternal supplementation during the pregnancy

    The role of surface microstructure on the resistance to stains of porcelain stoneware tiles

    Porcelain stoneware tiles frequently undergo a polishing process, aimed at improving their aesthetical appearence, that brings about a consistent material removal, with formation of superficial defects and opening of closed pores. The consequent degradation of surface characteristics and especially the increased sensitivity to stains-represent the main limit to the use of polished tiles in many indoor and outdoor applications. In order to better understand the role of microstructure on the resistance to stains, a phenomenological study of staining/cleaning operations (ISO 10545 parts 14 and 16) and a thoroughful physico-microstructural characterisation of tile working surfaces (SEM, open and closed porosity, rugosimetry, MIP) were carried out on twelve industrially manufactured and polished products. Diverse staining behaviours proved to be connected with different tile micro structures, being the surface roughness as well as the amount and shape of coarser pores the most influent variables. Through a statistical approach, an empirical predictional model of the amount of stain retained by the tile surface after mild washing with warm water was set up.
