Calcium Signaling and Contractility in Cardiac Myocyte of Wolframin Deficient Rats

Wolframin (Wfs1) is a membrane protein of the sarco/endoplasmic reticulum. Wfs1 mutations are responsible for the Wolfram syndrome, characterized by diabetic and neurological symptoms. Although Wfs1 is expressed in cardiac muscle, its role in this tissue is not clear. We have characterized the effect of invalidation of Wfs1 on calcium signaling-related processes in isolated ventricular myocytes of exon5-Wfs1 deficient rats (Wfs1-e5/-e5) before the onset of overt disease. Calcium transients and contraction were measured in field-stimulated isolated myocytes using confocal microscopy with calcium indicator fluo-3 AM and sarcomere length detection. Calcium currents and their calcium release-dependent inactivation were characterized in whole-cell patch-clamp experiments. At 4 months, Wfs1-e5/-e5 animals were euglycemic, and echocardiographic examination revealed fully compensated cardiac function. In field-stimulated isolated ventricular myocytes, both the amplitude and the duration of contraction of Wfs1-e5/-e5 animals were elevated relative to control Wfs1+/+ littermates. Increased contractility of myocytes resulted largely from prolonged cytosolic calcium transients. Neither the amplitude of calcium currents nor their voltage dependence of activation differed between the two groups. Calcium currents in Wfs1-e5/-e5 myocytes showed a larger extent of inactivation by short voltage prepulses applied to selectively induce calcium release-dependent inactivation of calcium current. Neither the calcium content of the sarcoplasmic reticulum, measured by application of 20 mmol/l caffeine, nor the expression of SERCA2, determined from Western blots, differed significantly in myocytes of Wfs1-e5/-e5 animals compared to control ones. These experiments point to increased duration of calcium release in ventricular myocytes of Wfs1-e5/-e5 animals. We speculate that the lack of functional wolframin might cause changes leading to upregulation of RyR2 channels resulting in prolongation of channel openings and/or a delay in termination of calcium release

Hashtagging on Instagram: Functions of Hashtags as Employed by #Instagirls

Hastagging, similarly to tagging, has become an inseparable part of online communication and users’ motivation for employing hashtags—especially on the platform of their origin, i.e. Twitter—has attracted the attention of scholars from various fields, including linguistics. In reference to previous studies, this paper aims to expand existing knowledge regarding the role and functions of hashtags by examining Instagram posts, which are, in contrast to tweets, predominantly visual and remain an insufficiently researched area. The analytical data consisting of posts uploaded by three Instafamous personalities, also known as Instagirls, were scanned for the presence of hashtags to determine their frequency and their characteristic traits. The qualitative analysis of posts shows that on Instagram, hashtags are employed to fulfil one of two opposing goals: 1. to serve as a tool for aggregating posts of similar content; 2. to serve as a resource for creative self-expression. Furthermore, since hashtags are often designated as a means of promoting one’s visibility, their role as a self-presentation strategy in the construction of micro-celebrity on Instagram is also discussed

Tato diplomová práce na téma HR ve vybraném hotelovém zařízení se zaměřuje hlavně na pojmy lidské zdroje, motivace a spokojenost zaměstnanců. Hlavním cílem práce je zjistit současný stav spokojenosti zaměstnanců ve vybraném oddělení hotelového zařízení a navrhnout doporučení, které povedou ke zlepšení motivace a spokojenosti zaměstnanců. Diplomová práce je rozdělena na dvě základní části. V první části jsou rozebrány teoretické stránky uvedené problematiky. Uvádí se v ní pojem hotelnictví, lidské zdroje, motivace zaměstnanců a jejich spokojenost. Ve druhé části se práce zaměřuje na praktické uskutečnění výzkumu, jeho kroky a postupy. Jsou zde rozpracované vedlejší cíle, kterými jsou získání interních informací o pracovní motivaci a spokojenosti zaměstnanců v dané společnosti. Nachází se zde také analýza dotazníkového šetření ve vybraném oddělení. V závěru práce jsou definovány a popsány konkrétní návrhy a doporučení manažerovi oddělení.This thesis on HR in selected hotel focuses mainly on the concepts of human resources, motivation and employee satisfaction. The main aim of the thesis is to find out the current state of employee‘s satisfaction in the selected department of the hotel facility and to propose recommendations that will lead to the improvement of motivation and satisfaction of employees. The thesis is divided into two basic parts. The first part deals with the theoretical aspects of the mentioned issue. It mentions the concept of hotel industry, human resources, employee motivation and satisfaction. In the second part, the thesis focuses on the practical realization of research, it‘s steps and procedures. There are sub-goals that are being developed to obtain internal information about work motivation and employee satisfaction in a given company. There is also an analysis of the questionnaire survey in the selected department. The first part of this Bachelor's thesis discussed the issue of vegetation along the river margins and banks according to the recommended literature. The literature re-search, based on the information obtained, dealt with the primary functions of wetlands and riparian vegetation. It also described individual parts (zones) of riverbed. In conclusion, the research covered the issue of riparian vegetation establishment and management. The second part contained the methodology, used in this thesis, basic characteris-tics of the river Jizera and a description of chosen areas. The area of interest was divided into seven parts, which were explored and described in detail in terms of plant species and their locations.
