    Anticancer activity of acetogenins from Annona muricata fruit

    Medicinal plants become very important in our days for their therapeutic benefits to humankind. It sustains human health, and it is commonly known as herbal medicines since ancient times. Annona muricata is a heart-shaped fruit that is consumed raw or as the fruit juice in the tropical area. A. muricata is used in traditional and alternative medicine to treat different ailments such as diabetes, hypertension, respiratory and skin illness, inflammation and cancer. A. muricata contains essential anticancer agents named acetogenins that play the significant role in various cancer types. Acetogenins are strong nicotinamide adenine dinucleotide oxidase inhibitors of the cancer cell's mitochondrial membrane but showed neurotoxic effects in rats. Therefore, acetogenins need to be further investigated to determine the exact mechanisms of action, long-term safety, optimal dosage, and potential side effects. Given the extensive studies on A. muricata, this review focuses on the phytochemistry, medicinal uses, biological activities and the mechanisms of action for the fruit extracts and acetogenins, to stimulate further studies on the fruit pulp used for human consumption

    Cytotoxicity effect of ionic liquid-graviola fruit (Annona muricata) extract to human colon cancer (HT29) cell lines

    The present study aimed to investigate the anti-proliferative effect of the ionic liquid-Graviola fruit (IL-GFE) extract on colon adenocarcinoma (HT29) cell lines and their kinetics behaviour to assess the Graviola fruit potential as a therapeutic alternative in cancer treatment. The phytoconstituents content of IL-GFE was identified using GC-TOFMS apparatus and measured its cytotoxicity on HT29 by tetrazolium bromide. Then the cytokinetic behaviour of the treated HT29 cells with IL-GFE was illustrated using the cells' growth curve. Besides, the cell cycle phase perturbation for the treated HT29 was applied using a flow cytometry technique. Qualitative identification of phytoconstituents of IL-GFE showed that Graviola fruit contains acetogenins, alkaloids, flavonoids, tannins and saponins compounds. IL-GF extract displayed a cytotoxicity effect on HT29 cells with the IC50 value of 10.56 µg/mL, while Taxol showed an IC50 value of 1.22 µg/mL. IL-GFE also decreased the cell generation number from 3.93 to 2.96 generations compared to Taxol-treated cells 2.01 generations. The microscope observation of the HT29 cells treated with the crude IL-GFE displayed loss of density and cell detachment. The extract's growth inhibition was related to the cell cycle arrest at the G0/G1 phase. IL-GFE inhibited colon adenocarcinoma HT29 cells' proliferation and affected their kinetic behaviour by lowering cell viability, inducing apoptosis, and arresting the cell cycle at the G0/G1 phase. ******************************************************************************** Kajian ini bertujuan untuk mengkaji kesan anti-proliferatif ekstrak buah-ion Graviola (IL-GFE) pada garis sel adenokarsinoma kolon (HT29) dan tingkah laku kinetik mereka untuk menilai potensi buah Graviola sebagai alternatif terapi untuk barah rawatan. Kandungan fitokonstituen IL-GFE dikenal pasti menggunakan alat GC-TOFMS dan mengukur sitotoksisitasnya pada HT29 oleh tetrazolium bromida. Kemudian tingkah laku sitokinetik sel HT29 yang dirawat dengan IL-GFE digambarkan menggunakan keluk pertumbuhan sel. Selain itu, gangguan fasa kitaran sel untuk HT29 yang dirawat diaplikasikan menggunakan teknik sitometri aliran. Pengenalpastian kualitatif fitokonstituen IL-GFE menunjukkan bahawa buah Graviola mengandungi asetogenin, alkaloid, flavonoid, tanin dan sebatian saponin. Ekstrak IL-GF memperlihatkan kesan sitotoksisiti pada sel HT29 dengan nilai IC50 10.56 µg/mL, sementara Taxol menunjukkan nilai IC50 1.22 µg/mL. IL-GFE juga menurunkan jumlah penjanaan sel dari 3.93 hingga 2.96 generasi berbanding sel yang dirawat Taxol 2.01 generasi. Pemerhatian mikroskop sel HT29 yang dirawat dengan IL-GFE kasar menunjukkan kehilangan ketumpatan dan detasmen sel. Perencatan pertumbuhan ekstrak berkaitan dengan penangkapan kitaran sel pada fasa G0/G1. IL-GFE menghalang percambahan sel HT29 adenokarsinoma kolon dan mempengaruhi tingkah laku kinetik mereka dengan menurunkan daya maju sel, mendorong apoptosis, dan menghentikan kitaran sel pada fasa G0/G1

    Antiproliferative activity of ionic liquid-graviola fruit extract against human breast cancer (MCF-7) cell lines using flow cytometry techniques

    Ethnopharmacological relevance Medicinal plants have been used for ages by indigenous communities around the world to help humankind sustain its health. Graviola (Annona muricata), also called soursop, is a member of the Annonaceae family and is an evergreen plant that is generally distributed in tropical and subtropical areas of the world. Graviola tree has a long history of traditional use due to its therapeutic potential including anti-inflammatory, antimicrobial, antioxidant, insecticide and cytotoxic to tumor cells. Aim of the study This study aimed to investigate the in vitro antiproliferative effects and apoptotic events of the ionic liquid extract of Graviola fruit (IL-GFE) on MCF-7 breast cancer cells and their cytokinetics behaviour to observe their potential as a therapeutic alternative in cancer treatment. Materials and methods The cell viability assay of the extract was measured using tetrazolium bromide (MTT assay) to observe the effects of Graviola fruit extract. Then the cytokinetics behaviour of MCF-7 cells treated with IL-GFE is observed by plotting the growth curve of the cells. Additionally, the cell cycle distribution and apoptosis mechanism of IL-GFE action on MCF-7 cancer cells were observed by flow cytometry. Results IL-GFE exhibited anti-proliferative activity on MCF-7 with the IC50 value of 4.75 μg/mL, compared to Taxol with an IC50 value of 0.99 μg/mL. IL- GFE also reduced the number of cell generations from 3.71 to 1.67 generations compared to 2.18 generations when treated with Taxol. Furthermore, the anti-proliferative activities were verified when the growth rate was decreased dynamically from 0.0077 h to 1 to 0.0035 h-1. Observation of the IL-GFE-treated MCF-7 under microscope demonstrated detachment of cells and loss of density. The growth inhibition of the cells by extracts was associated with cell cycle arrest at the G0/G1 phase, and phosphatidylserine externalisation confirms the anti-proliferation through apoptosis. Conclusions ionic liquid Graviola fruit extract affect the cytokinetics behaviour of MCF-7 cells by reducing cell viability, induce apoptosis and cell cycle arrest at the G0/G1 phase

    Active fractions of methanol crude obtained from acacia seyal gum and their anti-proliferative effects against human breast cancer cell lines

    Background: This study is on Acacia seyal gum, which is an exudate from Talha (Acacia seyal) tree. It provides a rich source of prebiotic that is used traditionally in folk medicine. Aims: The anti-proliferative effect (APE) of Acacia seyal gum (ASG) and Prebio-T-commercial (PTC) samples on human breast cancer (MCF-7) cell lines and their antioxidant activities (AA) were investigated. Methods: The methanol crude extracts of both Acacia seyal gum and Prebio-T-commercial were fractioned into acetone and methanol, respectively. The antiproliferative effect on human breast cancer cell lines for each fraction was examined using sulphorhodamine assay (SRB assay). Methanol crude extracts and their active compositions were analysed carefully using gas chromatography-mass spectrometry technique. Results: The most anti-proliferative effect was detected in the sample collected from Prebio-T-commercial (IC50=8.97µg/mL) as compared to Acacia seyal gum (IC50=9.56µg/mL). Regarding total phenolic content (TPC), the methanol crude extracts values are 694±2.58mg, GAE/100g for Prebio-T-commercial as compared to 155.78±2.58, GAE/100g for Acacia seyal gum. However, both acetone and methanol fractions of Acacia seyal gum and Prebio-T-commercial were found to be highly anti-proliferative to human breast cancer. For bioactive compounds determinations, the methanol crude extract from Acacia seyal gum is mainly dominated by Isovitamin C (42.37%), Crypton (5.86%), and Hydroquinone (4.86%) as major components. Conclusion: Finally, the antioxidant and anti-proliferative properties of the active fraction have shown some evidence regarding its use in traditional medicine as well as the prevention of cancer cell growth. This suggests the potential use of their bioactive compounds as natural anticancer agents

    Graviola fruit extraction by ionic liquid microwave-assisted extraction (IL-MAE)

    This chapter discusses the extraction method of Graviola fruit using ionic liquid microwave-assisted extraction (IL-MAE). Three parameters—irradiation power, solid/liquid (s/l) ratio, and extraction time—were investigated to obtain optimum extraction process resulting in a high yield and a low IC50 value for the anti-proliferation assay. The results showed that the extraction time of 1.74 min, the irradiation power of 300Wand the s/l ratio of 1:25 gave the highest extraction yield of 67.6%. Whereas the extraction time of 3 min instead of 2.98 min, the irradiation power of 700Winstead of 690W, and the s/l ratio of 1:39 produced an average IC50 of 4.75 ± 0.36 μg/mL for MCF7 and 10.56 ± 2.04 μg/mL for HT29. The IL-MAE technique was found to be effective in the extraction of plant fruit as described in the case study

Some of its phytochemicals were reported to have a neurotoxicity effect causing neurodegenerative diseases. However, different parts of this tree have been used for ages in traditional medicine due to their biological activities, such as anti-inflammatory, antimicrobial, antioxidant and anticancer effects. This study aimed to qualitatively screen the crude ionic liquid-Graviola fruit extract (IL-GFE) phytochemical composition, assess its acute toxicity and determine the lethal concentrations using zebrafish (Danio rerio) embryos. IL-GFE contains acetogenins, alkaloids, phenols, terpenoids, tannins and flavonoids. Acute toxicity effects of IL-GFE on zebrafish embryos were observed from 24 to 120 hours of post fertilisation (hpf). The survival rate, LC50, sublethal endpoints and effect of IL-GFE on the heart rate of zebrafish larvae were assessed. Results showed that the lethal concentration (LC50) of the crude IL-GFE was 173.45 μg/mL. Interestingly, no significant changes on the morphology of the treated zebrafish were observed at a concentration of 125 μg/mL. However, the heart rate of zebrafish larvae at 96 hpf was significantly decreased by 33.76% after treated with crude IL-GFE at 125 μg/mL (119.00 ± 4.72 beats/min) as compared to the untreated group (179.67 ± 4.66 beats/min). This preliminary finding showed that crude ionic liquid-Graviola fruit extract and its phytoconstituents might have the potential to be developed as a food supplement or herbal product. However, further tests need to be conducted to evaluate its medicinal properties and adverse effects on organisms of higher orders

    Consumer awareness and actual buying behaviour of organic food in Algeria

    Abstract Introduction. The adverse effects awareness of the chemicals misuse applied in food production is continuously increasing among consumers. In parallel, there is a rising trend in purchasing organically grown foods. Objective. This study aimed to evaluate Algerian consumers awareness and attitudes toward organic food products, and identify factors influencing their decision to purchase these organically grown foods. Material and methods. Survey data were obtained from 429 consumers via a 32-scale structured questionnaire, and evaluated with SPSS 25.0 and AMOS 25.0 using confirmatory factor analysis (CFA), and structural equation modeling (SEM). Hypotheses were verified by evaluating data via SEM, where mediating variables were attitude and purchasing intention. Results. The main reasons for Algerian consumers to consume organic food were health concerns, followed by environmentally friendly, pesticide-free, and better taste. Moreover, three factors namely health consciousness, subjective norms, and perception of availability were revealed to influence consumer buying behaviour towards organic food products. This occurred either by a direct effect such as availability perception or by a partial mediation of purchase intention, like subjective norms or by full mediating of attitude toward organic food, and purchase intention as the health consciousness factor did. Conclusion. The present study can guide and help agricultural policymakers to promote shifting to organic farming, which would benefit the ecosystem and human wellbeing. Furthermore, the report provides clear guidance for industry professionals who need to develop successful marketing strategies by addressing the main motivators of organic food consumption

    Evaluation of metabolomics behavior of human colon cancer HT29 celllines treated with ionic liquid graviola fruit pulp extract

    Ethnopharmacological relevance: Medicinal plants have been used by indigenous people across the world for centuries to help individuals preserve their wellbeing and cure diseases. Annona muricata L. (Graviola) which is belonging to the Annonaceae family has been traditionally used due to its medicinal abilities including antimicrobial, anti-inflammatory, antioxidant and cancer cell growth inhibition. Graviola is claimed to be a potential antitumor due to its selective cytotoxicity against several cancer cell lines. However, the metabolic mechanism information underlying the anticancer activity remains limited. Aim of the study: This study aimed to investigate the effect of ionic liquid-Graviola fruit pulp extract (IL-GPE) on the metabolomics behavior of colon cancer (HT29) by using an untargeted GC-TOFMS-based metabolic profiling. Materials and methods: Multivariate data analysis was used to determine the metabolic profiling, and the ingenuity pathway analysis (IPA) was used to predict the altered canonical pathways after treating the HT29 cells with crude IL-GPE and Taxol (positive control). Results: The principal components analysis (PCA) identified 44 metabolites with the most reliable factor loading, and the cluster analysis (CA) separated three groups of metabolites: metabolites specific to the non-treated HT29 cells, metabolites specific to the treated HT29 cells with the crude IL-GPE and metabolites specific to Taxol treatment. Pathway analysis of metabolomic profiles revealed an alteration of many metabolic pathways, including amino acid metabolism, aerobic glycolysis, urea cycle and ketone bodies metabolism that contribute to energy metabolism and cancer cell proliferation. Conclusion: The crude IL-GPE can be one of the promising anticancer agents due to its selective inhibition of energy metabolism and cancer cell proliferation

    Extraction method plays an essential role in extracting bioactive compounds. Ionic liquid microwave-assisted extraction (ILMAE) method has been successfully applied to get the optimum extraction yield from Annona muricata fruit. A series of the 1-butyl-3-methylimidazolium cation with different anions Cl-, BF4-, PF6- were assessed for extraction efficiency, and 1-butyl-3-methylimidazolium chloride was selected as the optimal solvent. Also, the microwave-assisted extraction parameters, including extraction time, power and solid-liquid ratio had been investigated. The response surface methodology was applied by using the extraction yield as response value to identify the optimum extraction condition. The optimum extraction condition is extraction time of 2 min, power of 300 W and a solid-liquid ratio of 1:25 g/mL. The extraction yield was 67.64% which is in close agreement with the value predicted by the model. Finally, ionic liquids as a green and designer solvents combined with microwave-assisted extraction could provide high extraction yields in less extraction time
